Project description:Tumor-derived exosomes are key factors that mediate the communication between tumor cells and stromal cells. In this study, we demonstrated that there was an uptake of cancer-derived exosomes by lung fibroblasts both in vivo and in vitro, leading to upregulation of inflammatory genes in fibroblasts, such as Il-6, Ccl1, Ccl2, Ccl5 and Cxcl2. Further analysis indicated that exosomal miR-3473b-mediated suppression of Nfkbid caused the inflammatory cytokine secertion in fibroblasts and further resulted in B cells accumulation in lungs. Inhibition of miR-3473b blocked inflammatory cytokine genes such as IL-6 expression in lung fibroblasts and B cell accumulation in lung. We further demonstrated that the lung metastasis areas and numbers were significantly decreased by using B cell neutralizing antibodies, suggesting B cell may play an important role in establishment of tumor promoting microenvironment for metastatic cancers cells colonization in the lung. In the present study, we showed that intercellular crosstalk occurred between cancer cells and fibroblasts via cancer-derived exosomes. Moreover, inhibition of the NF-kB activation in fibroblasts and B cells accumulation may become a potential target for the prevention and treatment of lung cancer.

Project description:Tumour-derived exosomes have been shown to induce pre-metastatic niche formation, favoring metastatic colonization of tumour cells, but the underlying molecular mechanism is still not fully understood. In this study, we showed that exosomes derived from the LLC cells could indeed significantly enhance their intrapulmonary colonization. Circulating LLC-derived exosomes were mainly engulfed by lung fibroblasts and led to the NF-κB signalling activation. Further studies indicated that the exosomal miR-3473b was responsible for that by hindering the NFKB inhibitor delta's (NFKBID) function. Blocking miR-3473b could reverse the exosome-mediated NF-κB activation of fibroblasts and decrease intrapulmonary colonization of lung tumour cells. Together, this study demonstrated that the miR-3473b in exosomes could mediate the interaction of lung tumour cells and local fibroblasts in metastatic sites and, therefore, enhance the metastasis of lung tumour cells.

Project description:Our findings have clinical implications. Identification of sputum exosomal miRNA helps explore the important biological pathways underlying the pathogenesis of bronchiectasis, thus unraveling candidate targets for future interventions of PA colonization. Apart from canonical inflammatory pathways, we have unraveled the modulation of longevity regulation pathway which opens a new avenue for exploring how PA colonization interacts with the airway epithelium. The significant correlation between sputum inflammatory biomarkers and miR-92b-5p and miR-223-3p provided further evidence on the unresolved inflammation in the PA-colonized microenvironment. However, causality cannot be inferred based on the current study design.

Project description:Development of effective therapies against brain metastasis is currently hindered by limitations in our understanding of the molecular mechanisms driving it. Here we define the contributions of tumour-secreted exosomes to brain metastatic colonization and demonstrate that pre-conditioning the brain microenvironment with exosomes from brain metastatic cells enhances cancer cell outgrowth. Proteomic analysis identified cell migration-inducing and hyaluronan-binding protein (CEMIP) as elevated in exosomes from brain metastatic, but not lung or bone metastatic cells. CEMIP depletion in tumour cells impaired brain metastasis, disrupting invasion and tumour cell association with the brain vasculature, phenotypes rescued by pre-conditioning the brain microenvironment with CEMIP+ exosomes. Moreover, uptake of CEMIP+ exosomes by brain endothelial and microglial cells induced endothelial cell branching and inflammation in the perivascular niche by upregulating Ptgs2, Tnf, and Ccl/Cxcl 86 cytokines, known to promote brain vascular remodeling and metastasis. CEMIP was elevated in tumour tissues and exosomes from patients with brain metastasis and predicted brain metastasis progression and patient survival. Collectively, our findings suggest that targeting of exosomal CEMIP could constitute a future avenue for the prevention and treatment of brain metastasis.

Project description:Helicobacter pylori (H. pylori) infection is characterized as progressive processes of bacterial persistence and chronic gastritis with features of infiltration of mononuclear cells more than granulocytes in gastric mucosa. Angiopoietin-like 4 (ANGPTL4) is considered a double-edged sword in inflammation-associated diseases, but its function and clinical relevance in H. pylori-associated pathology is unknown. Here we demonstrate both pro-colonization and pro-inflammation roles of ANGPTL4 in H. pylori infection. Increased ANGPTL4 in the infected gastric mucosa was produced from gastric epithelial cells (GECs) synergistically induced by H. pylori and IL-17A in a cagA-dependent manner. Human gastric ANGPTL4 correlated with H. pylori colonization and the severity of gastritis, and mouse ANGPTL4 from non-bone marrow-derived cells promoted bacteria colonization and inflammation. Importantly, H. pylori colonization and inflammation were attenuated in Il17a-/-, Angptl4-/-, Il17a-/-Angptl4-/- mice. Mechanistically, ANGPTL4 bound to integrin αV (ITGAV) on GECs to suppress CXCL1 production by inhibiting ERK, leading to decreased gastric influx of neutrophils, thereby promoting H. pylori colonization; ANGPTL4 also bound to ITGAV on monocytes to promote CCL5 production by activating PI3K-AKT-NF-κB, resulting in increased gastric influx of regulatory CD4+ T cells (Tregs) via CCL5-CCR4-dependent migration. In turn, ANGPTL4 induced Treg proliferation through binding to ITGAV to activate PI3K-AKT-NF-κB, promoting H. pylori-associated gastritis. Overall, we propose a model in which ANGPTL4 collectively ensures H. pylori persistence and promotes gastritis. Efforts to inhibit ANGPTL4-associated pathway may prove valuable strategies in treating H. pylori infection.

Project description:Early detection and surgical excision of tumors have helped improve the survival rate of 24 patients with breast cancer. However, patients with metastatic cancer have a poor prognosis. In 25 this study, we propose that ANKRD1 promotes metastasis of breast cancer. ANKRD1 was found 26 to be highly expressed in the MDA-MB-231 and MDA-LM-2 malignant metastatic breast cancer 27 cell lines compared to the non-metastatic breast cancer cells (MCF-7, ZR-75-30, T47D) and normal 28 breast cancer cells (MCF-10A). Furthermore, high-grade tumors showed increased levels of 29 ANKRD1 compared to low-grade tumors. Both in vitro and in vivo functional studies demon- 30 strated the essential role of ANKRD1 in cancer cell migration and invasion. The previous studies 31 have suggested an important role of NF-κB and MAGE-A6 in breast cancer metastasis, but the 32 upstream regulators of this axis are not well characterized. Our study suggests that ANKRD1 33 promotes metastasis of breast cancer by activating NF-κB as well as MAGE-A6 signaling. Our 34 findings show that ANKRD1 is a potential therapeutic target and a diagnostic marker for breast 35 cancer metastasis.
