Project description:<p><strong>BACKGROUND:</strong> Protein undernutrition is prevalent mainly among elderly people, and it could develop diseases such as sarcopenia. Urinary metabolites may become biomarkers reflecting protein undernutrition, but this has been insufficiently investigated.</p><p><strong>OBJECTIVE:</strong> The purpose of this study was to identify novel urinary metabolites as biomarker candidates responsive to protein undernutrition.</p><p><strong>METHODS:</strong> Adult male Wistar rats were fed an AIN-93 M diet [14% casein, control (CT) diet] or an AIN-93 M-based isocaloric diet [5% casein, low protein (LP) diet] for 4 weeks. 1H nuclear magnetic resonance metabolomic analysis was performed on urine samples weekly and on plasma and liver samples at week 4 to identify metabolites that respond to protein undernutrition. Liver samples were also subjected to mRNA microarray and quantitative PCR analyses at week 4 to investigate alterations of the gene expressions responsive to protein undernutrition.</p><p><strong>RESULTS:</strong> Urinary taurine levels were significantly lower in the LP group than in the CT group (79.7% lower) at week 1, and remained constant until week 4. Hepatic taurine level and gene expression level of cysteine dioxygenase type 1, the rate-limiting enzyme for taurine biosynthesis, were also significantly lower in the LP group than in the CT group (81.1% and 32.5% lower, respectively). Urinary trimethylamine N-oxide (TMAO) levels were significantly higher in the LP group than in the CT group (49.7% higher) at week 2, and remained constant until week 4. Hepatic TMAO level was also higher in the LP group than in the CT group (81.2% higher). Hepatic gene expression levels of flavin-containing monooxygenase 1 and 5, the enzymes for TMAO biosynthesis, were also significantly higher in the LP group than in the CT group (61.6% and 311.8% higher, respectively).</p><p><strong>CONCLUSIONS:</strong> Urinary taurine and TMAO levels substantially respond to LP diet ingestion, and may act as non-invasive biomarker candidates to detect protein undernutrition.</p>

Project description:The hypothesis of the present study is that metabolic phenotyping of blood plasma allows to (i) discriminate between colorectal cancer patients and control subjects and (ii) identify new biomarkers for colorectal cancer. In order to test this hypothesis, the investigators will apply proton nuclear magnetic resonance (1H-NMR) spectroscopy to perform metabolic phenotyping of blood plasma in 50 colorectal cancer patients and 50 control subjects. Multivariate statistics will be performed to assess the discriminative power of the applied methodology in distinguishing between both groups and to identify metabolites with potential as biomarkers for colorectal cancer.

Project description:BackgroundPerinatally HIV-infected children on anti-retroviral treatment (ART) are reported to have metabolic abnormalities such as dyslipidemia, lipodystrophy, and insulin resistance which potentially increase the risk of diabetes, kidney, liver and cardiovascular disease.ObjectiveTo elucidate HIV-mediated metabolic complications that sustain even during ART in perinatally HIV-infected children.MethodWe have carried out metabolic profiling of the plasma of treatment-naïve and ART-suppressed perinatally HIV-infected children and uninfected controls using 1H nuclear magnetic resonance (NMR) spectroscopy followed by statistical analysis and annotation.ResultValidated multivariate analysis showed clear distinction among our study groups. Our results showed elevated levels of lactate, glucose, phosphoenolpyruvic acid, propionic acid, 2-ketobutyric acid and tricarboxylic acid (TCA) cycle metabolites in untreated HIV-infected children compared to uninfected controls. ART normalized the levels of several metabolites, however the level of lactate, phosphoenolpyruvic acid, oxoglutaric acid, oxaloacetic acid, myoinositol and glutamine remained upregulated despite ART in HIV-infected children. Pathway analysis revealed perturbed propanoate metabolism, amino acid metabolism, glycolysis and TCA cycle in untreated and ART-suppressed HIV-infected children.ConclusionDeveloping therapeutic strategies targeting metabolic abnormalities may be beneficial for preventing diabetes, cardiovascular disease or other associated complications in perinatally HIV-infected children.

Project description:Human epidemiological evidence has led scientists to theorize that undernutrition during gestation is an important early origin of adult diseases. Animal models have successfully demonstrated that maternal diet could contribute to some of adult diseases. Undernutrition is perceived harmful in pregnant women whereas calorie restriction is a strategy proven to extend healthy and maximum life span in adult. This diagrammatically opposite effect of nutritional condition might provide us hints to search for genes underlying health conditions. Here, we have initiated a study examining the effect of undernutrition on maternal and fetal livers, utilizing high-throughput DNA microarray analysis for screening genome-wide changes in their transcriptomes. Briefly, pregnant mice were exposed to food deprivation (FD) on gestation day (GD) 17, and caesarean section was performed on GD18. Control mice were supplied with chow ad libitum till sacrifice. Total RNA extracted from mother and fetal livers for each control and treatment (FD) was analyzed with an Agilent mouse whole genome DNA chip. Total of 3058 and 3126 up (>1.5 fold)- and down (<0.75 fold)-regulated genes, and 1475 and 1225 up- (>1.5 fold)- and down (<0.75 fold)-regulated genes showed differential expression at the mRNA level, in the maternal and fetal livers, respectively. Interestingly, 103 genes up-regulated in mother were down-regulated in fetus, whereas 108 down-regulated maternal genes were up-regulated in fetus; these 211 genes are potential candidates related to longevity or health. The role of some of these genes, in context of the proposed mechanisms for developmental origins of health and disease is discussed. Comparison between mouse control and FD livers in fetus and mother was performed. Five to ten biological replicates were used, and pooled total RNA from each condition (control and FD) was dye-swaped.

Project description:In utero undernutrition is associated with obesity and insulin resistance, although its effect on skeletal muscle remains poorly defined. We report that, in mice, adult offspring from undernourished dams have decreased energy expenditure, decreased skeletal muscle mitochondrial content, and altered energetics in isolated mitochondria and permeabilized muscle fibers. Strikingly, when these mice are put on a 40% calorie restricted diet they lose half as much weight as calorie restricted controls. Our results reveal for the first time that in utero undernutrition alters metabolic physiology having a profound effect on skeletal muscle energetics and response to calorie restriction in adulthood.
