Project description:Total RNASeq of 5 HREC donors exposed to TNF-alpha and IL1-beta. Here we seek to investigate the transcriptional regulation of ICAM-1 in human retinal endothelial cells (HREC). In order to do this we have collected primary isolates from 5 HREC donors and exposed them to Tumour Necrosis Factor alpha and Interleukin-1. Total RNA was sequenced on a NovaSeq to 1.2 illion 150bp read pairs per sample. Samples here cluster primarily by donor and then by time (60 minutes vs 24 hours).

Project description:Super-selective intra-ophthalmic artery chemotherapy (SSIOAC) is an organ-specific drug-delivery strategy to treat retinoblastoma, the most common primary ocular malignancy in children. Unfortunately, recent clinical reports associate adverse vascular toxicities with SSIOAC using melphalan, the most commonly used chemotherapeutic. To explore the reason for the unexpected vascular toxicities, we have developed in vitro studies with human retinal endothelial cells to test the effects of the chemotherapeutics and a non-human primate model to monitor the SSIOAC treatment in real-time and post-treatment. Melphalan and carboplatin (another chemotherapeutic used to treat retinoblastoma via SSIOAC) triggered migration, proliferation, and apoptosis when used to treat human retinal endothelial cells. Melphalan was associated with increased adhesion of leukocytes to human retinal endothelial cells, and tended to increase with increased cell expression of adhesion proteins (ICAM-1) and soluble chemotactic factors (IL-8). Histopathology post-SSIOAC indicated vessel wall sloughing, leukostasis, and vessel occlusion. We have established an in vitro human cell culture model and a non-human primate model to evaluate strategies designed to obviate vascular side effects, and optimize the efficacy of SSIAOC and the drug preparations used in SSIOAC. 4 non-treated (CNT) vs. 4 carboplatin-treated primary human retinal endothelial cells (RECs).

Project description:Transcriptional profiles of Fz4-/- retinal endothelial cells were compared to that of wild type endothelial cells under various culture conditions. The goal was to identify the transcriptional response to Frizzled 4 signaling in cultured retinal endothelial cells. To analyze the Norrin response of WT and Fz4-/- retinal endothelial cells in culture, we co-cultured these cells either with HEK293 cell line that stably expresses Norrin or with control 293 cells.

Project description:Retinal histogenesis occurs over an extended period, which necessitates retinal progenitors to self-renew to sustain the temporal generation of diverse cell types. Embryonic day 18 retinal cells showed self-renewal properties such as clonal propagation without compromising multipotentiality when grown in the presence of endothelial cell conditioned medium and not EGF alone. Timed pregnant Sprague Dawley (Embryonic day 18) rats were obtained from Charles River Laboratories. Embryos were harvested and enucleated. Retinae were dissected out and dissociated by trypsin-EDTA followed by manual trituration. The retinal cells obtained from 10 embryos were cultured in the presence of EGF (condition1) or EGF and Endothelial cell conditioned medium (condition 2) to obtain neurospheres. To get biological replicate for both conditions, another batch of embryos were pooled and subjected to neurosphere formation. Total RNA was extracted from both conditions using Qiagen RNeasy mini kit following manufacturer's protocol. The RNA was subjected to microarray analysis using Affymetrix Rat 230 2.0 platform by UNMC microarray Core facility. RMA analysis was performed using Genepattern software. Log2 transformed values of each condition was used to obtain fold change of specific genes.

The goal was to identify the transcriptional response to Frizzled 4 signaling in cultured retinal endothelial cells. To analyze the Norrin response of WT and Fz4-/- retinal endothelial cells in culture, we co-cultured these cells either with HEK293 cell line that stably expresses Norrin or with control 293 cells. Experiment Overall Design: To isolate the requisite retinal endothelial cell lines, we crossed Fz4-/- and control animals into the Immorto-mouse line in which a gamma interferon responsive H2Kb promoter directs the expression of a temperature sensitive SV40 large T-antigen (Jat et al., 1991). When cultured at 33°C in the presence of gamma interferon, this conditional oncogene system extends the proliferative capacity of a variety of cell types, which can be subsequently analyzed under phenotypically reverting conditions (37°C without gamma interferon). The cell immuno-purification procedure follows that described by Matsubara et al. (2000) and Su et al. (2003). The purified cells were then cultured and passaged at 33°C on dishes coated with 1% gelatin. For RNA preparation, cells were cultured at 37°C in the absence of interferon gamma. For the Matrigel experiments, endothelial cells previously starved for 24 hrs in DMEM with 1% FBS were trypsinized and plated on the Matrigel layer. The cells were incubated in DMEM with 1% FBS for 6 hours at 37°C before harvesting for RNA extraction. For the 293 cell co-culture experiments, endothelial cells were co-cultured with a human embryonic kidney (HEK) 293 cell line that stably expresses Norrin, or with control 293 cells.
