Project description:Animals in transport are exposed to both psychological and physical stressors, which affect performance and health. The objectives of this study were to investigate whether and how the transportation stress effect on female reproduction in mice, and detect differentially expressed genes association with stress response and reproductive capacity utilizing gene expression profiles. After transported mice by car to and from laboratory for a total period of 10 h, nearly 1000 km distance, measurements were carried out to explore effects of transportation stress on reproductive traits including puberty, estrous cycle, follicular number within ovaries, fertility and superovulation quality. Early life stress suffered mice before puberty promoted puberty onset, altered estrous cycle length and reduced large antral follicle number. Pubertal and adult mice exposure to transportation stress induced their pregnancy rate decline. Furthermore, adult stressed mice showed an obvious reduction in fertility with less litter size and litter weight in birth, which may cause reduced ovulation oocytes identified by superovulation. Assuming ovulated oocytes number as reproductive parameter, genes differently expressed between mice with extremely high and extremely low oocytes were compared within control and stress group separately.

Project description:Seminal fluid factors modulate the female immune response at conception to facilitate embryo implantation and reproductive success. Whether sperm affect this response has not been clear. We evaluated global gene expression by microarray in the mouse uterus after mating with intact or vasectomized males. Intact males induced greater changes in gene transcription, prominently affecting pro-inflammatory cytokine and immune regulatory genes, with TLR4 signaling identified as a top-ranked upstream driver. Recruitment of neutrophils and expansion of peripheral regulatory T cells were elevated by seminal fluid of intact males. In vitro, epididymal sperm induced IL6, CXCL2, and CSF3 in uterine epithelial cells of wild-type, but not Tlr4 null females. Collectively these experiments show that sperm assist in promoting female immune tolerance by eliciting uterine cytokine expression through TLR4-dependent signaling. The findings indicate a biological role for sperm beyond oocyte fertilization, in modulating immune mechanisms involved in female control of reproductive investment. miRNA profile of duck duodenum mucosa damage caused by transportation stress

Project description:We deep sequenced and analyzed miRNAs using deep RNA sequencing (RNA-seq) in transported and control duck's duodenum sample of Jingjiang duck. We analyzed the miRNA data with 9467248 reads and 9808143 million reads, obtained 9338224 and 9677777 clean reads in transported and control duck's by high-throughput sequencing, respectively. we respectively gained 4636135 and 4759049 miRNAs sequences in two groups by filtering the known non-miRNA reads, such as rRNA, tRNA, snRNA, and snoRNA by screening against ncRNA deposited in the GenBank and Rfam databases. These findings provided insights into the expression profiles of miRNAs in duck duodenum, and deepened our understanding of miRNAs in transportation injury of duck.
