Project description:Excess nutrient uptake and altered hormone secretion in the gut contribute to a systemic energy imbalance causing obesity and increased risk for type 2 diabetes (T2D) and colorectal cancer. This functional maladaptation is thought to emerge at the level of the intestinal stem cells (ISCs). But, how an obesogenic diet affects ISC identity and fate decisions is not well understood. Here, we study the intestinal responses to an obesogenic diet in mice by combining single-cell profiling of more than 27,000 small intestinal crypt cells with genetic lineage labelling and tracing of ISC fate decisions and in situ metabolomics. We find that an obesogenic diet induces ISC and progenitor hyperproliferation, but also enhances ISC differentiation and cell turnover. Furthermore, the regional identity of ISCs and enterocytes is reprogrammed from a distal to a proximal phenotype and enterocyte zonation is altered to adapt cell function to nutrient availability. Single-cell resolution of the enteroendocrine lineage shows an increase in endocrine progenitors and peptidergic enteroendocrine cell (EEC) types and a decrease in serotonergic EEC types. Mechanistically, we can link increased fatty acid synthesis, Ppar signalling and the Insr/Igf1r/Akt pathway to ISC dysfunction, hyperproliferation and impaired endocrine differentiation. In summary, our work describes key molecular mechanisms of diet-induced intestinal maladaptation in mice that promote obesity, and, thus, underlie the pathogenesis of the metabolic syndrome and associated complications.

Project description:This dataset consists of single-cell RNA-seq (10X) data from small intestinal crypts of healthy and HFHSD induced obese mice Foxa2-FVF/FVF mice and from small intestinal villi of healthy and HFHSD induced obese C57BL/6N mice. Mice were fed a HFHSD for 12 weeks. Intestinal crypt cell samples were FVF-enriched with FVF+ cells in a 1:1 ratio (50% FVF+, 50% FVF-) before sequencing.

Project description:Epidemiological surveys first indicated that obesity and type II diabetes generated by lipid rich diets may be efficiently inherited, both paternally and maternally. This apparent exception to the fundamental law of non heritability of acquired characters was more recently documented in laboratory organisms but the transgenerational signal remained a matter of speculation. Here we show that experimental transfer to healthy one-cell embryos of testis RNA prepared from males raised on high-fat-diet (HFD) results in the development of obesity and diabetes in the adult. Body weight, fasting glucose levels, response to insulin, glucose tolerance, RNA levels of leptin and stearoyl-CoA desaturase were all affected in mice born after RNA microinjection to the same extent as in the progenitors and in their sexual offspring. Surprisingly, neither microarray hybridization nor deep sequencing of the inducer and control RNAs showed a differential expression of known regulators of adipogenesis, but only modest variations in a series of transcripts. Transgenerational maintenance appears as one instance of the RNA-mediated heredity of epigenetic traits previously demonstrated in experimental systems ranging from the worm to the mouse. The relationship between lipid-rich diet and changes in testicular RNAs on one side and the nature of the mark imprinted on the one-cell embryo by RNA transfer and responsible for pathological developments at later stages are questions of interest for future studies. Here, we illustrate an important role of the TGF-beta/Smad3 signaling pathway in regulating glucose and energy homeostasis. Smad3 deficient mice are protected from diet-induced obesity and diabetes. Interestingly, the metabolic protection is accompanied by Smad3-/- white adipose tissue acquiring the bioenergetic and gene expression profile of brown fat/skeletal muscle. Smad3-/- adipocytes demonstrate a marked increase in mitochondrial biogenesis, with a corresponding increase in basal respiration, and Smad3 acts as a repressor of PGC-alpha1 expression. We observe significant correlation between TGF-beta1 levels and adiposity in rodents and humans. Further, systemic blockade of TGF-beta1 signaling protects mice from obesity, diabetes and hepatic steatosis. Together, these results demonstrate that TGF-beta signaling regulates glucose tolerance and energy homeostasis and suggest that modulation of TGF-beta1 activity might be an effective treatment strategy for obesity and diabetes. Although adipocytes are critical in obesity, their role in diabetes has only recently been appreciated. We conducted studies by using DNA microarrays to identify differences in gene expression in adipose tissue from lean, obese, and obese-diabetic mice. The expression level of over 11,000 transcripts was analyzed, and 214 transcripts showed significant differences between lean and obese mice. Surprisingly, the expression of genes normally associated with adipocyte differentiation were down-regulated in obesity. Not all obese individuals will become diabetic; many remain normoglycemic despite profound obesity. Understanding the transition to obesity with concomitant diabetes will provide important clues to the pathogenesis of type 2 diabetes. Therefore, we examined the levels of gene expression in adipose tissue from five groups of obese mice with varying degrees of hyperglycemia, and we identified 88 genes whose expression strongly correlated with diabetes severity. This group included many genes that are known to be involved in signal transduction and energy metabolism as well as genes not previously examined in the context of diabetes. Our data show that a decrease in expression of genes normally involved in adipogenesis is associated with obesity, and we further identify genes important for subsequent development of type 2 diabetes mellitus. Experiment Overall Design: For obesity study, lean samples include C57BL/6J lean, (C57BL/6J X BTBR) F1 and BTBR lean, obese sampples incluse C57BL/6J-ob/ob, (C57BL/6J X BTBR) F2-ob/ob and BTBR-ob/ob.
