A single SNP in the M1UK tmRNA gene ssrA drives enhanced SpeA superantigen expression as a result of ssrA terminator readthrough in the M1UK lineage. This represents a new paradigm of toxin expression and urges enhanced international surveillance.

Project description:Kashin-Beck disease (KBD) is an endemic and chronic osteochondropathy with unknown etiology. The disease mostly occurs in children between the ages of 3 and 13 in a diagonal belt-like area ranging from Northeast to Southwest China. We carried out this microarray analysis to investigate the differences in gene expression levels between KBD patients and healthy controls. Total RNA was isolated from peripheral blood mononuclear cells (PBMCs).

Project description:With its 2.5 Mb DNA genome packed in amphora-shaped particles of bacterium-like dimension (1.2 µm in length, 0.5 µm in diameter), the Acanthamoeba-infecting Pandoravirus salinus remained the most spectacular and intriguing virus since its description in 2013. Following its isolation from shallow marine sediment off the coast of central Chile, that of its relative Pandoravirus dulcis from a fresh water pond near Melbourne, Australia, suggested that they were the first representatives of an emerging worldwide-distributed family of giant viruses. This was further suggested when P. inopinatum discovered in Germany, was sequenced in 2015. We now report the isolation and genome sequencing of three new strains (P. quercus, P.neocaledonia, P. macleodensis) from France, New Caledonia, and Australia. Using a combination of transcriptomic, proteomic, and bioinformatic analyses, we found that these six viruses share enough distinctive features to justify their classification in a new family, the Pandoraviridae, distinct from that of other large DNA viruses.

Project description:O. viverrini infection is recognised by the WHO as a group 1 biological carcinogen because of its extremely strong link with cholangiocarcinoma (CCA), cancer of the bile ducts. The incidence of CCA associated with liver fluke infection, calculated as an age‐standardized rate, varies by geographical region and other risk factors but exceeds 100 per 100,000 in men and 40 per 100,000 in women in hotspots in northeast Thailand. Any attempts to control liver fluke infection, whether through mass drug administration or ultimately through vaccination requires specific and sensitive diagnostic tools that are readily deployable in the field and easy to use. It is imperative that methods to detect infection are appropriately sensitive and rapid in order to diagnose new cases, assess effectiveness of elimination measures and be applicable to large-scale disease surveillance. Indeed, the WHO recently highlighted the low sensitivity of current diagnostic methods as a barrier to controlling liver fluke infection. Herein, we have leveraged the O. viverrini soluble secreted proteome to help create the first O. viverrini protein microarray.

Project description:The goal of this study was to examine commonalities in the molecular basis of learning in mice and humans. In our previous work on mouse learning we suppressed activity in the anterior cingulate cortex (ACC) and hippocampus (HC) and found the stages for which each area was critical in performing a 2-choice visuospatial discrimination task. Our current study began by examining gene expression changes in mouse ACC and HC as a result of learning a new skill. Genes upregulated in both brain areas were used as candidates to examine commonalities between genes upregulated in mouse and human blood. We used microarrays to identify candidate genes and real-time PCR to compare the mouse results with two forms of human learning. One form involved training in a working memory task (network training), the other a generalized training shown to change many networks (meditation). We identified two genes that were upregulated in both mice and humans following training. We believe these genes act to regulate pathways that influence NF-κB, a factor previously found to be related to enhanced synaptic function and learning.
