Project description:Untargeted proteomics from a 5,000 km+ transect across the central Pacific Ocean from Hawaii to Tahiti. The expedition crossed multiple biogeochemical provinces, inlcuding the oligotrophic North Pacific Subtropical Gyre, the extremety of the Eastern Tropical North Pacific Oxygen Deficient Zone, and the relatively productive equatorial region associated with upwelling. This dataset focuses on the microbial fraction (0.2-3.0 micrometer filter size) and the microbial community dynamics across these biogeochemical provinces, from the surface oceance to the mesopelagic (1,250 m depth maximum).

Project description:SPARC is a matricellular glycoprotein involved in regulation of the extracellular matrix, growth factors, adhesion, and migration. SPARC-null mice have altered basement membranes and develop posterior sub-capsular cataracts with cell swelling and equatorial vacuoles. Exchange of fluid, nutrients, and waste products in the avascular lens is driven by a unique circulating ion current. Here we demonstrate that SPARC-null mouse lenses exhibit abnormal circulation of fluid, ion, and small molecules which leads to altered fluorescein distribution in vivo, loss of resting membrane polarization, and altered distribution of small molecules. Microarray analysis of SPARC-null lenses showed changes in gene expression of ion channels and receptors, matrix and adhesion genes, cytoskeleton, immune response genes, and cell signaling molecules. Our results demonstrate that the regulation of SPARC on cell-capsular matrix interactions can influence the circulation of fluid and ions in the lens, and the phenotype in the SPARC-null mouse lens is the result of multiple intersecting pathways.

Experiment Overall Design: Lens epithelial cells from 7 lenses of littermate mice were isolated by laser capture microdissection. 3 wild-type lenses from 3 different mice and 4 knock-out lenses from 3 different mice were used as biological replicates.

Project description:The mitotic kinesin-like Protein 2 (MKLP2, KIF20A) is essential for the proper execution of cytokinesis and required to ‘passage’ the chromosomal passenger complex (CPC) from the chromosomes in (pro)metaphase to the spindle midzone and equatorial cortex in anaphase. Together with MKLP1 (KIF23) and MPP1 (KIF20B), MKLP2 forms the kinesin-6 subfamily of motor proteins. Whilst MKLP1 and MPP1 are both plus-end directed processive motors, the typical structure of the MKLP2 motor domain along with the extended neck-linker region, suggested that MKLP2 might not be able to function as a transport motor but is more likely to act as a microtubule bundler. This notion contradicts the prevailing hypothesis that MKLP2 can transport the CPC in anaphase, but appears to be in line with in vitro and in cellulo studies showing that recombinant MKLP2 efficiently bundles microtubules, and that knock-down of MKLP2, disturbs the organization of the anaphase spindle midzone. Using TIRF microscopy and purified fluorescent proteins, we here demonstrate that MKLP2 is a plus-end directed processive motor that can transport the CPC along microtubules in vitro. Live imaging of MKLP2::GFP and INCENP::GFP in early anaphase cells revealed that a fraction of both MKLP2 and INCENP displays directed movements towards the (equatorial) cortex. In line, inhibition of MKLP2 ATPase activity at the start of anaphase disturbed the localization of MKLP2 and CPC at the equatorial cortex. Our data suggest that MKLP2 is a processive microtubule-based motor that transports itself and the CPC to the equatorial cortex.

Project description:Local adaptation is often a product of environmental variations in geographical space and has implications for biodiversity conservation. We investigated the role of latitudinal heterogeneity in climate on the organization of genetic and phenotypic variation in the dominant coastal tree Avicennia schaueriana. In a common garden experiment, samples from an equatorial region, with pronounced seasonality in precipitation, accumulated less biomass, and showed lower stomatal conductance and transpiration, narrower xylem vessels, smaller leaves and higher reflectance of long wavelengths by the stem epidermis than samples from a subtropical region, with seasonality in temperature and no dry season. Transcriptomic differences identified between trees sampled under field conditions at equatorial and subtropical sites, were enriched in functional categories such as responses to temperature, solar radiation, water deficit, photosynthesis and cell wall biosynthesis. Remarkably, the diversity based on genome-wide SNPs revealed a north-south genetic structure and signatures of selection were identified for loci associated with photosynthesis, anthocyanin accumulation and the responses to osmotic and hypoxia stresses. Our results suggest the existence of divergence in key resource-use characteristics, likely driven by seasonality in water deficit and solar radiation. These findings provide a basis for conservation plans and for predicting coastal plants responses to climate change.
