Project description:Branched-chain aminotransferases (BCAT) are enzymes that initiate the catabolism of branched-chain amino acids (BCAA), such as leucine, thereby providing macromolecule precursors. In order to study the effect of BCAT1 inhibitor (ERG240) on LPS-polarized macrophage transcriptome, we stimulated monocyte derived macrophages (MDMs) with LPS for 8 hours, with or without the presence of ERG240. We then performed whole-genome transcriptome sequencing by RNA-sequencing (RNA-seq).

Project description:We have employed whole genome microarray expression profiling to identify differently expressed genes following stimulation of wild-type bone-marrow derived macrophages with Escherichia coli lipopolysaccharide (LPS). Macrophages were stimulated with LPS (100 ng/ml) for 6 hours and a signature was identified that distinguished between infected and control samples. Expression of several genes from this signature was quantified in the same RNA samples by real-time PCR, confirming the predicted macrophage response pattern.

Project description:Ureteral obstruction is marked by disappearance of the vasopressin-dependent water channel aquaporin-2 (AQP2) in the renal collecting duct and polyuria upon reversal. Most studies of unilateral ureteral obstruction (UUO) models have examined late time points, obscuring the early signals that trigger loss of AQP2. Here, we performed RNA-Seq on microdissected rat cortical collecting ducts (CCDs) to identify early signaling pathways post-UUO.
