Project description:Ectodermal patterning is required for the establishment of multiple components of the vertebrate body plan. Previous studies have demonstrated that precise combinations of extracellular signals determine the induction of neural or neural crest progenitors. Yet, we still have a limited understanding of how the response to inductive signals is optimized to generate the proper transcriptional output in target cells. Here we show that post-transcriptional attenuation of signaling gradients is essential for the formation of the neural crest. We found that neural crest cells display enhanced expression of Dicer, which promotes the maturation of a set of cell-type-specific miRNAs. These miRNAs target multiple components of the FGF signaling pathway, a central player in the process of neural induction in avian embryos. Loss of neural crest miRNAs prevented the attenuation of this neuralizing signal, leading to the expansion of the neural plate at the expense of neural crest cells. Thus, the post-transcriptional attenuation of FGF signaling is pre-requisite for neural crest specification. These findings demonstrate how post-transcriptional repression may reshape signaling gradients to set the boundaries between distinct spatial domains of progenitor cells.

Here we show that posttranscriptional attenuation of fibroblast growth factor (FGF) signaling is essential for the establishment of the neural crest territory. We found that neural crest progenitors display elevated expression of DICER, which promotes enhanced maturation of a set of cell-type-specific miRNAs. These miRNAs collectively target components of the FGF signaling pathway, a central player in the process of neural induction in amniotes. Inactivation of this posttranscriptional circuit results in a fate switch, in which neural crest cells are converted into progenitors of the central nervous system. Thus, the posttranscriptional attenuation of signaling systems is a prerequisite for proper segregation of ectodermal cell types. These findings demonstrate how posttranscriptional repression may alter the activity of signaling systems to generate distinct spatial domains of progenitor cells.

Project description:Neural cest cells are a transient stem cell-like population appearing during vertebrate embryonic development. Generation of the cranial neural crest is known to require a balanced combination of FGF and BMP levels. However, it is poorly understood how the functions of such growth factors are controlled in the extracellular spaces. Anosmin is an extracellular matrix protein implicated in FGF signaling and mutated in Kallmann syndrome. Here, we demonstrate that anosmin (Gga.14976.1.S1_at, clone ChEST132d10) is synthesized locally in the cranial neural crest of chicken embryos and is essential for cranial neural crest formation. Anosmin upregulates FGF8 and BMP5 gene expression; it also enhances FGF8 activity while inhibiting BMP5 and WNT3a signaling. Taken together, our data establish that the matrix protein anosmin is required for cranial neural crest formation, with funtional modulation of FGF, BMP, and WNT.

Project description:The complex interplay between signaling systems, transcription factors, and cis-regulatory elements required for development are encoded within gene regulatory networks (GRN). Although GRNs have greatly expanded our understanding of cell fate decisions during embryogenesis, they are typically assembled via a candidate-based approach, which fails to consider the tissue specific functions more broadly expressed TFs may employ. To address this, we combined a TF-focused CRISPR screen with epigenomic and transcriptional profiling of human neural crest cells. This approach unbiasedly identified several key regulators of human neural crest cell development, including the ubiquitous AP-1 transcription factor complex. Functional characterization of AP-1 demonstrated it is a major regulator of neural crest identity, working with the cell type specific TF, TFAP2A, to shape the neural crest epigenome. Interestingly, overexpression of AP-1 and TFAP2A in human embryonic stem cells is sufficient to drive neural crest identity, suggesting a critical role for AP-1 in the transition from pluripotency to more specialized stem cell states. Furthermore, we demonstrate that AP-1 serves as a nuclear effector of FGF/MAPK signaling in human neural crest cells, a previously missing link between FGF signaling and neural crest development. Our results demonstrate signaling effectors carry out cell-type specific enhancer selection and may contribute to our efforts in engineering neural crest cells for the purpose of tissue repair and regeneration.

Project description:During embryonic development, cellular identity relies upon the complex interplay between signaling systems, transcription factors, and cis-regulatory elements. This is epitomized in the neural crest, whose formation is governed by a comprehensive gene regulatory network (GRN) jumpstarted by several signaling systems including Wnt, BMP, and FGF. Although there is substantial knowledge regarding the nuclear effectors and downstream target genes of Wnt and BMP signaling in neural crest formation; the effectors of FGF signaling remain unclear. Recent work from our group has demonstrated a decisive role for the FGF/MAPK/AP-1 axis in modulating the human neural crest epigenome. Nevertheless, whether this regulation remains conserved across vertebrate species remains unknown. To address this, we examined the activity and requirement of the AP-1 transcription factor complex in avian neural crest cell formation. Functional characterization of AP-1 demonstrated it is a major regulator of avian neural crest identity, directly regulating the expression of several GRN components. Furthermore, we demonstrate that AP-1 serves as a nuclear effector of FGF signaling in avian neural crest cells, consistent with our previous results observed in human neural crest. Together, these results demonstrate the conserved activity of AP-1 required for neural crest specification and can facilitate its incorporation into the GRN.

Project description:We report that FGF exposure of sensory progenitors (pPPR) leads to rapid deposition of active chromatin marks H3K27ac near hundreds of FGF-responsive, otic-epibranchial progenitor (OEP) genes, while H3K27ac is depleted in the vicinity of non-otic genes. Genomic regions that gain H3K27ac act as cis-regulatory elements controlling OEP gene expression in time and space and define a unique transcription factor signature likely to control their activity. Thus, during ear induction FGF signalling modifies the chromatin landscape to promote enhancer activation and chromatin accessibility.

Project description:Assessment of differentiation potential is a basic requirement to obtain qualified human pluripotent stem cells (hPSCs). Here, we report a simple differentiation method using fetal bovine serum (FBS) to estimate differentiation potential and propensity of hPSCs. PluriTest using RNA-sequencing showed that cells differentiated after treatment with 5% FBS. Expression patterns of three germ layer markers revealed that cells cultured in Knockout Serum Replacement-containing medium (KSR) with mouse feeder cells had higher differentiation potential than cells cultured in a chemically defined medium (E8) with recombinant matrix proteins, especially into the mesoderm and endoderm lineages. Analysis of differentially expressed genes between KSR and E8 identified DUSP6 as a marker for where cells had been cultured. Expression of DUSP6 correlated with FGF-ERK signaling activity. Fine-tuning of FGF-ERK signaling activity to a range that can shut down DUSP6 transcription but sustain NANOG transcription partially increased the differentiation potential. Our data suggest that differentiation with 5% FBS is good to estimate differentiation potential and propensity at the early stage, and that DUSP6 is an excellent marker to monitor ERK signaling activity.
