Project description:Photomorphogenesis is a critical developmental process bridging light-regulated transcriptional reprogramming with morphological changes in organisms. Strikingly, the chromatin-based transcriptional control of photomorphogenesis remains poorly understood. Here, we show that both light-induced genes and dark-induced genes were affected in the atino80 mutant. Genome-wide occupancy of the H2A.Z histone variant and levels of histone H3 were reduced in atino80.

Project description:We observed the expression profile of the total mRNA of litR (TTHB100) deletion mutant of T. thermophilus HB8 strain grown in a rich (TM) medium at 60 M-BM-0C under the dark condition compared with that of wild-type. Three wild-type strain of T. thermophilus HB8 were pre-cultured at 60 M-BM-0C for 16 hours in 100 mL of TM medium containing 0.8% polypeptone, 0.4% yeast extract, 0.2% NaCl, 0.4 mM CaCl2, and 0.4 mM MgCl2, which was adjusted to pH 7.5 with NaOH. The cells (1 mL) were inoculated into 100 mL of the synthetic medium and then cultivated at 60M-BM-0C for 8 hours under dark condition. Then the 1.0 mM of ZnSO4 was added into the medium, and continued cultivation. Cells were collected, and then, the expression of each mRNA was analyzed.

Project description:The transcriptome of the three atino80 allelic mutants was compared to that of wild-type and 50B Arabidopsis plants (see Fritsch et al. 2004). Since the transcriptomes of 50B and wild-type plants were found to be identical, we compared expression in the mutant with 50B and with wild-type without distinction. Therefore, we had four replicates of the wild type condition (50B line, wild-type) and two replicates for each of the mutant alleles (atino80-1, atino80-2 and atino80-3), all ecotype Columbia. All lines were profiled in duplicate (grown independently at 2-week-intervals).
