Project description:We developed a post-menopausal orthotopic ER+ breast cancer model that we used to investigate mechanisms of obesity mediated lung metastases. Obesity altered the immune cell composition and function in the primary tumor microenvironment. To identify tumor-derived genes associated with obesity mediated lung metastases we performed RNASeq on tumors grown in lean vs obese mice. From these results we identified a hepatocyte growth factor (HGF)-c-Met (HGF receptor)-CCL2 loop recruiting metastasis-promoting cells.

Project description:Obesity is thought to contribute to worse disease outcome in breast cancer as a result of increased levels of adipocyte-secreted endocrine factors, insulin, and insulin-like growth factors (IGFs) that accelerate tumor cell proliferation and impair treatment response. We examined the effects of patient obesity on primary breast tumor gene expression, by profiling transcription of a set of tumors for which the patients’ body mass index (BMI) was ascertained. Sample profiles were stratified according to patients’ obesity phenotype defined as normal (BMI <25), overweight (BMI 25-29.9), or obese (BMI>30). Widespread alterations in gene expression were evident in breast tumors from obese patients as compared to tumors from other patients, allowing us to define an obesity-associated cancer transcriptional signature of 662 genes. Project description:Lysine residues undergo diverse and reversible post-translational modifications including acetylation. Acetylation of lysine residues have traditionally been studied as epigenetic modifiers of histone tails within chromatin that provides an important mechanism for regulating gene expression. In the heart, histone acetylation acts as a key regulator of cardiac remodeling and function. However, recent studies have shown that thousands of proteins (~4,500) can be acetylated at multiple acetylation sites (~15,000 sites). These data suggest that the acetylome rivals phosphorylation in prevalence as a post-translational modification. Based on this, we examined the impact of obesity on the regulation of lysine acetylation in the left ventricle of male c57BL/6J mice. We report that obesity contributed to a significant increase in heart enlargement and fibrosis. Of interest, immunoblot analysis demonstrated that lysine acetylation was markedly altered in response to diet-induced obesity and that this phenomena was cardiac tissue specific. Mass spectral analysis was performed in which 3264 proteins were identified in the left ventricle. Of these, 254 proteins were acetylated, 16 of which were significantly impacted by obesity. Ingenuity Pathway Analysis identified the Cardiovascular Disease network as significantly regulated by obesity, 54 of the 254 acetylated proteins impact this pathway. This network includes LIM domain-binding protein 3 (LDB3), aconitate hydratase (ACO2), and dihydrolipoyl dehydrogenase (DLD), which are all significantly impacted by obesity and known to regulate cardiac function. Combined, these findings suggest a critical role for the cardiac acetylome in obesity-mediated remodeling and ultimately have the potential to elucidate novel targets that regulate cardiac pathology.

Project description:Obesity is linked to an increased risk of atrial fibrillation (AF) via increased oxidative stress. While NADPH oxidase II (NOX2), a major source of oxidative stress and reactive oxygen species (ROS) in the heart predisposes to AF, the underlying mechanisms remain unclear. Here, we studied NOX2-mediated ROS production in obesity-mediated AF using Nox2-knock-out (KO) mice and mature human induced pluripotent stem cell-derived atrial cardiomyocytes (hiPSC-aCMs). Diet-induced obesity (DIO) mice and hiPSC-aCMs treated with palmitic acid (PA) were infused with a NOX blocker (apocynin) and a NOX2-specific inhibitor, respectively. We showed that NOX2 inhibition normalized atrial action potential duration and abrogated obesity-mediated ion channel remodeling with reduced AF burden. Unbiased transcriptomics analysis revealed that NOX2 mediates atrial remodeling in obesity-mediated AF in DIO mice, PA-treated hiPSC-aCMs, and human atrial tissue from obese individuals by upregulation of paired-like homeodomain transcription factor 2 (PITX2). Furthermore, hiPSC-aCMs treated with hydrogen peroxide, a NOX2 surrogate, displayed increased PITX2 expression, establishing a mechanistic link between increased NOX2-mediated ROS production and modulation of PITX2. Our findings offer insights into possible mechanisms through which obesity triggers AF and support NOX2 inhibition as a potential novel prophylactic or adjunctive therapy for patients with obesity-mediated AF.

Project description:ABSTRACT: Obesity is responsible for decreased overall survival for breast cancer patients. Here, we describe the generation, characterization and application of a novel murine mammary tumor initiating cell model (M-Wnt) that recapitulates the claudin-low subtype of human breast cancer and permits the study of TIC’s in wild-type, immunocompetent mice. M-Wnt cells readily form mammospheres in suspension culture, express markers consistent with epithelial-to-mesenchymal transition (EMT), and generate claudin-low mammary tumors when as few as 50 cells are orthotopically injected. Using the M-Wnt cell lines in tandem with a more basal-like epithelial breast cancer cell line, E-Wnt, we found that diet induced obesity significantly downregulates epithelial markers, such as E-cadherin, and upregulates mesenchymal markers including fibronectin, N-cadherin, SNAIL, Oct-4, and TGF-b. This reveals a previously unidentified link between energy balance and EMT. The ability of calorie restriction (CR) to reverse EMT, upregulate epithelial markers and downregulate mesenchymal markers indicates the plasticity of the TICs, as well as the potential importance of lifestyle modifications as cancer prevention strategies. 28 array samples

Project description:Obesity has long been considered one of the most important risk factors for a number of cancer types, including breast cancer and remains one the more severe disease concerns. However, the functional importance of noncoding RNA elements remains elusive. Here, we report the upregulation of SNORD46 in obesity serum. The expression status of SNORD46 is correlated with donor’s body mass index (BMI). The expression of SNORD46 in adipocytes was regulated in a CEBP-beta-dependent manner. Mechanically, adipocyte-expressed SNORD46 bound with interleukin-15 (IL-15) in serum, by which Snord46 G11A mutant exhibited enhanced interaction with IL-15. A knockin mouse strain harboring a Snord46 G11A mutation (referred as Snord46mut/mut mice) exhibited obesity and obesity associated complexities compared to wild type or heterozygous littermates. The SNORD46-IL-15 interaction blocked IL-15-dependent non-canonical signaling events, which contained the tyrosine-protein kinase Fer-dependent phosphorylation of platelet glycoprotein 4 (CD36) and monoglyceride lipase (MGLL). Consequently, the obesity-expressed SNORD46 suppressed the IL-15 triggered, Fer-dependent phosphorylation of CD36 and MGLL, leading to inhibited lipase of adipocytes. SNORD46 locked nucleic acids (LNAs) effectively conquered the inhibitory effect of SNORD46 and exhibited anti-obesity effects. Hence, our findings demonstrated the functional importance of snoRNAs in obesity and the efficacy of snoRNA LNAs for antagonizing obesity.

Project description:Immune checkpoint blockade (ICB) has improved outcomes in some cancers. A major limitation of ICB is that a majority of patients fail to respond, which is partly attributable to immunosuppression. Obesity appears to improve immune checkpoint therapies in some cancers but impacts on breast cancer (BC) remain unknown. In obese mice fed an obesogenic or lean controls on a low fat diet, mice are injected with BC cells andtreated with anti-PD-1 or control. Obesity leads to immunosuppression systemically in tumor-free mice that is exacerbated in tumor-bearing mice. Obesity augments tumor incidence and progression. Anti-PD-1 induces regression in lean mice and potently abrogates progression in obese mice. Anti PD-1 significantly reduces immunosuppressive cells and elevates anti-tumor immune cells. Lamb2, downregulated by anti-PD1, significantly predict patient survival. Last, we identify a microbial signature associated with anti-PD-1 efficacy. In sum, anti-PD-1 is highly efficacious in obese mice by reinvigorating durable antitumor immunity.
