Transcription profiling of mouse pancreatic islets from wild type and DcR3 transgenic mice to investigate primary non-function (PNF) in islet transplantation

Project description:The islet primary non-function (PNF) is a serious problem in islet transplantation. In this study, we investigated whether DcR3-secreting transgenic (Tg) islets could reduce PNF. We generated transgenic mice expressing human DcR3. The transgenically expressed DcR3 protected islets from IFN-gama plus IL-1beta, or TNF-alpha plus IL-1beta-induced dysfunction and apoptosis in vitro. The Tg islets presented significantly reduced PNF after transplantation. <br><br> Three independent batches of islet isolation were carried out. For each batch, islets were obtained from 4 Tg and 4 WT mice, and pooled respectively. The pooled islets (Tg or WT) were then divided into 4 groups: 2 were cultured in the presence of IFN-gamma (0.5 ug/ml) plus IL-1beta (0.5 ng/ml) with 1 harvested at 24 h and 1 harvested at 48 h; 2 were cultured in the presence of TNF-alpha (100 ng/ml) plus IL-1beta (0.5 ng/ml) with 1 harvested at 24 h and 1 harvested at 48 h. Each treatment employed 3 chips using RNA from 3 different batches of islet isolation. For each treatment, genes with a mean signal strength difference above 2-fold between Tg and WT islets were selected for reverse PCR confirmation.

Project description:We investigated the underlying molecular mechanism of Brsk2's effects by comparing gene expression of whole primary islets isolated from Brsk2 TG mice and their control littermates by RNA sequencing.We chosed a four-day Dox administration and studied gene-expression differences resulting in hyperglycemia. A total of 23318 gene transcripts was captured,and 1020 genes showed significant expression differences (p-value<0.05 and >2-fold change) between the two group.Gene set-enrichment ananlysis was performed using Kyoto Encyclopedia of Genomes algorithms and MSigDB software.The most enriched pathways were linked to pathyways associated with olfactory tranduction, ribosomes, and pancreatic secretion. We focused on olfactory transduction because 1122 dysregulated genes are involved in that process.

RNA-seq of mice islets from wt and miR-29a/b-1 pancreatic beta cell specific overexpression transgenic mice

Project description:We investigated the mechanism associated with miR-29 effects by comparing gene expression of whole primary islets isolated from miR-29a/b-1 pancreatic beta cell specific overexpression transgenic mice (βTG mice) and their control littermates by RNA sequencing. Ten-week-old mice were chosen to avoid secondarily affected genes. We identified 346 genes that showed significant differences (p-value < 0.05 and >2-fold change) between the two groups. We then used gene ontology (GO) algorithms to perform functional annotations of these differential genes. The top enriched pathways were linked primarily to inflammatory response, secretory vesicle, and ion channel activity. Islets from the βTG mice showed an enhanced inflammatory response based on an assessment of both the upregulated and the downregulated genes , which suggested a likely positive regulation of phagocytosis (Ahsg), regulation of blood vessel diameter (P2rx1/Kng1/Nos2), and monocyte chemotaxis (Cxcl10/Cxcl17).

Gene expression data of islets from WT and βRapKO mice

Project description:The mechanistic target of rapamycin complex 1 (mTORC1) regulates beta cell growth and mass; yet it remains unclear whether it also directs beta cell functional maturation. To understand the global molecular basis of the phenotype caused by the loss of Raptor in beta cells, we isolated pancreatic islets from 8-week-old βRapKO and WT mice. We compared gene-expression profile by Affymetrix microarray of islets, which revealed that a number of mRNAs were dys-regulated in Raptor-deficient islets.
