Project description:To better understand the regulatory mechanisms of water stress response in wheat, the transcript profiles in roots of two wheat genotypes, namely, drought tolerant 'Luohan No.2' (LH) and drought susceptible 'Chinese Spring' (CS) under water-stress were comparatively analyzed by using the Affymetrix wheat GeneChip®. A total of 3831 transcripts displayed 2-fold or more expression changes, 1593 transcripts were induced compared with 2238 transcripts were repressed, in LH under water-stress; Relatively fewer transcripts were drought responsive in CS, 1404 transcripts were induced and 1493 were repressed. Comparatively, 569 transcripts were commonly induced and 424 transcripts commonly repressed in LH and CS under water-stress. 689 transcripts (757 probe sets) identified from LH and 537 transcripts (575 probe sets) from CS were annotated and classified into 10 functional categories, and 74 transcripts derived from 80 probe sets displayed the change ratios no less than 16 in LH or CS. Several kinds of candidate genes were differentially expressed between the LH and CS, which could be responsible for the difference in drought tolerance of the two genotypes.

Project description:The use of cereal microgreens is increasing because of increased consumer's interest in healthier products. Chlorophyll (Chl) and Carotenoids (Car) are suggested to correlate with health promoting components like phenolics and antioxidant potential of the plant-part. They also play role against clinical conditions like thalassemia and hemolytic anemia and reduce the risk of some chronic diseases, such as cancer, cardiovascular diseases, skin diseases and age-related eye diseases. This study was carried out for the comprehensive profiling of Chl and Car in wheat (Triticum aestivum L.), and barley (Hordeum vulgare L.) micro-greens between 7 and 16 days on dry basis. Chl and Car content strongly correlated with the number of days of growth. Significantly high correlations existed among Chl a, Chl b, total Chl and total Car with concomitant Chl a/b and Chl/Car ratios. The peaks for the rate of accumulation of pigments were between 7-10 days on wheat and 10-13 days on barley. The maximum content of Chl and Car were 616.63?±?18.45 mg/100 g DM and 54.80?±?1.72 mg/100 g DM on day 16 and statistically not significant to variety of grain. The Chl level was slightly lower than Chl rich vegetables like kale and comparable to spinach and Car level was comparable to carrots, which is higher than most of the daily consumable fruits and vegetables. Further cell-based or in vivo studies of cereal microgreens could be considered to draw more valuable information related to human health.

Project description:The Green Revolution dwarfing genes, Rht-B1b and Rht-D1b, encode mutant forms of DELLA proteins and are present in most modern wheat varieties. DELLA proteins have been implicated in the response to biotic stress in the model plant, Arabidopsis thaliana. Using defined wheat Rht near-isogenic lines and barley Sln1 gain of function (GoF) and loss of function (LoF) lines, the role of DELLA in response to biotic stress was investigated in pathosystems representing contrasting trophic styles (biotrophic, hemibiotrophic, and necrotrophic). GoF mutant alleles in wheat and barley confer a resistance trade-off with increased susceptibility to biotrophic pathogens and increased resistance to necrotrophic pathogens whilst the converse was conferred by a LoF mutant allele. The polyploid nature of the wheat genome buffered the effect of single Rht GoF mutations relative to barley (diploid), particularly in respect of increased susceptibility to biotrophic pathogens. A role for DELLA in controlling cell death responses is proposed. Similar to Arabidopsis, a resistance trade-off to pathogens with contrasting pathogenic lifestyles has been identified in monocotyledonous cereal species. Appreciation of the pleiotropic role of DELLA in biotic stress responses in cereals has implications for plant breeding.

Two common wheat (Triticum aestivum L.) cultivars, Luohan No.2 (LH) and Chinese Spring (CS), were used for this study. Seedlings at the two leaf stage were stressed by cultured in PEG solutions for 6h, and some other seedlings were cultured in tap water as control. Root samples of LH and CS at 6h after the stress treatment and untreated control were prepared for microarray analysis.
