Project description:Nucleic acids in wastewater provide a rich source of data for detection and surveillance of microbes. We have longitudinally collected 116 RNA samples from a wastewater treatment plant in Berlin/Germany, from March 2021 to July 2022, and 24 DNA samples from May to July 2022. We tracked human astroviruses, enteroviruses, noroviruses and adenoviruses over time to the level of strains or even individual nucleotide variations, showing how detailed human pathogens can be observed using wastewater. For respiratory pathogens, a broad enrichment panel enabled us to detect waves of RSV, influenza, or common cold coronaviruses in high agreement with clinical data. By applying a profile Hidden Markov Model-based search for novel viruses, we identified more than 100 thousand novel transcript assemblies likely not belonging to known virus species, thus substantially expanding our knowledge of virus diversity. Phylogenetic analysis is shown for bunyaviruses and parvoviruses. Finally, we identify Hundreds of novel protein sequences for CRISPR-associated proteins such as Transposase B, a class of small RNA-guided DNA editing enzymes. Taken together, we present a longitudinal and deep investigation into wastewater-derived genomic sequencing data that underlines the value of sewage surveillance for public health, planetary virome research, and biotechnological potential.

Project description:Industrial activities produce a variety of pollutants that may not be easily treated using centralized wastewater treatment systems based on a single treatment unit. The variability of the pollutants brings the importance of industrial-specific integrated wastewater treatment plants such as integrated membrane filtration systems. However, the performance of a membrane filtration process can be highly affected by the presence of high amounts of suspended particles in the raw wastewater. Therefore, proper selection of a pre-treatment unit prior to a membrane filtration wastewater treatment system is a key aspect of its performance. This study investigated the performance of an integrated membrane filtration treatment system connected to an electrochemical process (pre-treatment) on the purification of a poultry slaughterhouse wastewater toward achieving a high-quality effluent. The industrial-scale treatment plant installed at the Izhevsk Production Corporative (PC) poultry farm in Kazakhstan is composed of an electrochemical, ultrafiltration (UF), and reverse osmosis (RO) as the main treatment units. From the analysis results, the electrochemical pre-treatment unit was observed to be highly effective for the removal of some physicochemical parameters such as turbidity, color, total suspended solids, total iron, aluminum, chemical oxygen demand, and biochemical oxygen demand; with removal efficiency ranging from 71 to 85%. The low removal efficiency of the pre-treatment system was also observed from free and total chlorine, nitrites, nitrates, phosphates, and ammonium nitrogen; with removal efficiency ranging from 4 to 45%. While in general, the overall treatment train was observed to be highly efficient for some physicochemical parameters such as turbidity, color, total suspended solids, as well as chemical and biochemical oxygen demand; maintaining almost 100% removal efficiency throughout the study period. Also, the high removal efficiency of the electrochemical pre-treatment processes led to a relatively low rate of cake formation on the membrane filters.

Project description:Effluents from sewage treatment plants contain a mixture of micropollutants with the potential of harming aquatic organisms. Thus, addition of advanced treatment techniques to complement existing conventional methods has been proposed. Some of the advanced techniques could, however, potentially produce additional compounds affecting exposed organisms by unknown modes of action. In the present study the aim was to improve our understanding of how exposure to different sewage effluents affects fish. This was achieved by explorative microarray and quantitative PCR analyses of hepatic gene expression, as well as relative organ sizes of rainbow trout exposed to different sewage effluents (conventionally treated, granular activated carbon, ozonation (5 or 15 mg/L), 5 mg/L ozone plus a moving bed biofilm reactor, or UV-light treatment in combination with hydrogen peroxide). Exposure to the conventionally treated effluent caused a significant increase in liver and heart somatic indexes, an effect removed by all other treatments. Genes connected to xenobiotic metabolism, including cytochrome p450 1A, were differentially expressed in the fish exposed to the conventionally treated effluents, though only effluent treatment with granular activated carbon or ozone at 15 mg/L completely removed this response. The mRNA expression of heat shock protein 70 kDa was induced in all three groups exposed to ozone-treated effluents, suggesting some form of added stress in these fish. The induction of estrogen-responsive genes in the fish exposed to the conventionally treated effluent was effectively reduced by all investigated advanced treatment technologies, although the moving bed biofilm reactor was least efficient. Taken together, granular activated carbon showed the highest potential of reducing responses in fish induced by exposure to sewage effluents.
