Project description:Salinization poses a significant challenge in agriculture, exacerbated by anthropogenic global warming. Biostimulants, derived from living microorganisms or natural extracts, have emerged as valuable tools for conventional and organic agriculture. However, our understanding of the molecular mechanisms underlying the effects of biostimulants is very limited, especially in crops under real cultivation conditions. In this study, we adopted an integrative approach to investigate the effectiveness of the combined application of plant growth-promoting bacterium (Bacillus megaterium strain BM08) and a non-microbial biostimulant under control conditions (normal watering) and salt stress. After confirming the yield increase under both conditions, we investigated the molecular mechanisms underlying the observed effect by measuring a number of physiological parameters (i.e., lipid peroxidation, antioxidants, chlorophylls, total phenolics and phytohormone content), as well as RNA sequencing and primary metabolite analyses. Our findings reveal that the combined effect of the microbial and non-microbial biostimulants led to a decrease in the antioxidant response and an up-regulation of genes involved in cytokinin biosynthesis under salt stress conditions. This, in turn, resulted in a higher concentration of the bioactive cytokinin, isopentenyladenosine, in roots and leaves and an increase in γ-aminobutyric acid, a non-proteic amino acid related to abiotic stress responses. In addition, we observed a decrease in malic acid, along with an abscisic acid (ABA)-independent up-regulation of SR-kinases, a family of protein kinases associated with abiotic stress responses. Furthermore, we observed that the single application of the non-microbial biostimulant triggers an ABA-dependent response under salt stress; however, when combined with the microbial biostimulant, it potentiated the mechanisms triggered by the BM08 bacterial strain. This comprehensive investigation shows that the combination of two biostimulants is able to elicit a cytokinin-dependent response that may explain the observed yield increase under salt stress conditions.

Project description:The Fusarium incarnatum strain K23, originally isolated from a habit-adapted wild plant Thapsia species, colonized the roots and shoots of tomato seedlings and protected them against salt stress. Comparison of expression and metabolite profile changes uncovered that the fungus completely reprogramed the tomato response to salt stress. Barely any overlap was observed among the genes and metabolites which are regulated by salt stress in uncolonized and colonized tomato seedlings. In colonized seedlings exposed to salt stress, less stress- related genes are activated than in un-colonized seedlings. Furthermore, K23 produced gibberellin and gibberellin-responsive genes were detected in all RNA samples. Our analysis demonstrates that K23 colonisation completely alters the salt-responsive gene and metabolite profiles in tomato seedlings.

Project description:The aim of this study was to examine the transcriptional changes that govern the primary nitrogen (N) limitation response in roots and the long-term effect of N limitation in the basal nodes, which may be associated with tiller suppression by N supply. The effect of biostimulant application on N limitation response was also examined by including plant treated and untreated with microalgae extract-based biostimulant. For this purpose, RNA-seq was conducted in roots of 15 days-old hydroponically grown wheat plants (treated and untreated with biostimulants) 24 h after the introduction of the plants to the N limitation and in basal nodes of 32 days-old wheat plants (treated and untreated with biostimulants) exposed to N limitation for 18 days. The two different N treatments were: High N (10 mM N) and Low N (0.1 mM N). In total, three biological replicates were included per treatment. RNA-seq was conducted in total RNA extracted from 3 basal node samples pooled together per biological replicate. In this study, the basal node is defined as the 0.5 cm of the main shoot base, which includes the apical meristem, lateral buds, leaf meristems etc.

Project description:Salinity stress is a major problem affecting plant growth and crop productivity. While plant biostimulants have been reported to be an effective solution to tackle salinity stress in different crops, the key genes and metabolic pathways involved in these tolerance processes remain unclear. This study focused on integrating phenotypic, physiological, biochemical and transcriptome data obtained from different tissues of Solanum lycopersicum L. plants (cv. Micro-Tom) subjected to a saline irrigation water program for 61 days (EC: 5.8 dS/m) and treated with a combination of protein hydrolysate and Ascophyllum nodosum-derived biostimulant, namely PSI-475. The biostimulant application was associated with the maintenance of higher K+/Na+ ratios in both young leaf and root tissue and the overexpression of transporter genes related to ion homeostasis (e.g., NHX4, HKT1;2). A more efficient osmotic adjustment was characterized by a significant increase in relative water content (RWC), which most likely was associated with osmolyte accumulation and upregulation of genes related to aquaporins (e.g., PIP2.1, TIP2.1). A higher content of photosynthetic pigments (+19.8% to +27.5%), increased expression of genes involved in photosynthetic efficiency and chlorophyll biosynthesis (e.g., LHC, PORC) and enhanced primary carbon and nitrogen metabolic mechanisms were observed, leading to a higher fruit yield and fruit number (47.5% and 32.5%, respectively). Overall, it can be concluded that the precision engineered PSI-475 biostimulant can provide long-term protective effects on salinity stressed tomato plants through a well-defined mode of action in different plant tissues.

Project description:The stress proteasome in the animal kingdom facilitates faster conversion of oxidized proteins during stress conditions by incorporating different catalytic β subunits. Plants deal with similar kind of stresses and also carry multiple paralogous genes encoding for each of the three catalytic β subunits. Here we investigated the existence of stress proteasomes upon salt stress in tomato roots. In contrast to Arabidopsis thaliana, tomato has a simplified proteasome gene set with single genes encoding each β subunit except for two genes encoding β2. Using proteasome activity profiling on tomato roots during salt stress we discovered a transient modification of the catalytic subunits of the proteasome coinciding with a loss of viability. This stress-induced proteasome disappears at later time points and coincides with the need to degrade oxidized proteins during salt stress. Subunit-selective proteasome probes and MS analysis of fluorescent 2D gels demonstrated that the stress-induced proteasome is not caused by altered subunit assembly, but involves an increased molecular weight (MW) of both β2 and β5 subunits, and an additional acidic pI shift for β5, whilst β1 remains mostly unchanged. Although the underlying subunit modification is unknown, we can rule out ubiquitination, phosphorylation and glycosylation as underlying molecular mechanisms. This stress-induced proteasome may play an important role in PCD during abiotic stress.

Project description:Several fungi have been reported to form intimate bonds with plants. These close mutual relationships can provide physiological benefits to the interacting organisms, including the improvement of nutrient assimilation or enhanced stress tolerance. The root-colonizing fungus Fusarium sp. strain K-23 is known to successfully colonize tomato roots. Previous studies highlighted the promotion of plant biomass production and stress tolerance of tomato plants infected with the K-23. However, up to data, nothing is known about the effect of this fungus on the model plant Arabidopsis thaliana. We here report the comparative analysis of the effect of mock- and K-23-infected wild-type Arabidopsis plants (Col-0) in the abscence and presence of moderate salt stress conditions (50 mM NaCl). Our data provide evidence for a profound effect of K-23 on root hair elongation through the activation of a regulatory network that includes several plant hormones. Intriguingly, the fungus suppresses gibberellic acid biosynthesis which, in turn, promotes root hair elongation.
