Project description:The mechanisms underlying Roux-en-Y gastric bypass (RYGB) surgery-induced weight loss and the immediate postoperative beneficial metabolic effects associated with the operation remain uncertain. We aimed to identify novel gut-derived peptides with therapeutic potential in obesity and/or diabetes by determining genome-wide expression patterns in isolated human small intestinal enteroendocrine cells (EECs) obtained from 20 obese subjects undergoing RYGB and again three months later by upper enteroscopy. RYGB increased expression levels of the inverse ghrelin receptor agonist, liver-enriched antimicrobial peptide 2 (LEAP2), and a secreted endogenous LEAP2 fragment (LEAP238-47) demonstrated insulinotropic properties in vitro, promoting an increase in insulin release comparable to the gut hormone glucagon-like peptide 1. LEAP238-47 showed reciprocal effects on growth hormone secretagogue receptor (GHSR) activity, suggesting that the insulinotropic action of the peptide may be directly linked to attenuation of tonic GHSR activity. The fragment was infused to healthy human individuals, but compared to placebo, no glucoregulatory effect was observed in the chosen dose. In conclusion, small intestinal LEAP2 expression was upregulated after RYGB; and the fragment LEAP238-47 showed a strong in vitro insulinotropic effect, but failed to elicit glucoregulatory effects when infused in healthy human subjects.

Identification of two novel candidate genes for insulin secretion by comparative genomics of multiple backcross populations

Project description:To identify novel disease genes for type 2 diabetes (T2D) we generated two backcross populations of obese and diabetes-susceptible New Zealand Obese (NZO) with the two lean mouse strains 129P2 and C3H/FeJ. Subsequent whole-genome linkage scan revealed 36 novel quantitative trait loci (QTL) for T2D-associated traits. The strongest association with blood glucose (12 cM, LOD 13.3) and plasma insulin (17 cM, LOD 4.8) was detected on proximal chromosome 7 (designated Cdp7prox) exclusively in the NZOxC3H crossbreeding, suggesting that the causal gene is unique for the C3H genome. Introgression of the critical C3H fragment into the genetic NZO background by generating recombinant congenic strains (RCS) and metabolic phenotyping validated the phenotype. For the detection of candidate genes in the critical region (30-46 Mb) we used a combined approach of haplotype- and gene expression analysis to search for C3H-specific gene variants in the pancreatic islets, which appeared as the most likely target tissue for the QTL. Only the two genes Potassium-transporting ATPase alpha chain 1 (Atp4a) and Ribonuclease P protein subunit p29 (Pop4) fulfilled the criteria from our candidate gene approaches. The knockdown of both genes in MIN6 cells led to a decreased glucose-stimulated insulin secretion (GSIS), indicating a stimulating role of both genes in insulin secretion, thereby likely contributing to the phenotype linked to Cdp7prox. In conclusion, our combined- and comparative-cross analysis approach has successfully led to the identification of two novel diabetes susceptibility genes and thus has proven to be a powerful tool for the discovery of novel disease genes.

Silencing of mt-tRF-LeuTAA fragment in pancreatic rat islets and investigation by RNA-sequencing of the downstream mechanisms activated by this fragment

Project description:In this study, we discovered cytosolic and mitochondrial fragments resulting from tRNA and mt-tRNA cleavage, which may act as new regulators of cellular and metabolic functions. We selected the mt-tRF-LeuTAA fragment for further investigation, as its level is altered in the islets diabetes-susceptible animal models, while being abundant in ß-cells. mt-tRF-LeuTAA fragment is derived from the cleavage of tRNA-LeuTAA encoded by the mitochondrial genome. We demonstrated that mt-tRF-LeuTAA acts as a key regulator of mitochondrial OXPHOS functions, mitochondrial membrane potential, the insulin secretory capacity of ß-cells, and the insulin sensitivity of myotube muscle cells. To gain a comprehensive understanding, we conducted transcriptomic and proteomic analyses on rat islets with silenced mt-tRF-LeuTAA for 72 hours. We transfected rat islet cells with an antisense oligonucleotides (antisense oligonucleotides to reduce mt-tRF-LeuTAA levels versus control oligos). These oligos specifically decreased the levels of the fragment (anti-tRF-LeuTAA) by more than 95% without affecting the host full-length tRNA mt-tRNA-LeuTAA. Inhibiting mt-tRF-LeuTAA led to significant differential expression of 4843 mRNA transcripts, cut-off adjusted P ≤ 0.05. To further investigate the cellular rearrangement associated with the inhibition of mt-tRF-LeuTAA, we conducted enrichment analysis using Gene Ontology Molecular Function terms on RNA-sequencing. At the transcriptomic level, we observed enrichment of pathways related to ion transport and various enzymatic activities, including mitochondrial oxidoreductase activity, when the mitochondrial fragment was repressed in rat islets.

Study of the tRNA-derived fragment expression profile in rat pancreatic islets of diabetes-susceptible low-protein (LP) exposed rats

Project description:In this study, we discovered cytosolic and mitochondrial fragments resulting from tRNA and mt-tRNA cleavage, which may act as new regulators of cellular and metabolic functions. The so-called LP rat model consists of 22-day-old rats whose mothers have been subjected to a low protein diet during gestation and lactation. These offspring have impaired pancreatic development with reduced ß-cell mass, glucose-stimulated insulin secretion defect and are more prone to develop diabetes in adulthood. We identified hundreds of these fragments as being changed in the pancreatic islets of LP rats and compared them to chow diet (CD) exposed rats. In our analysis, we identified 6164 tRFs in the islets of LP rats, among which 256 exhibited significant changes (≥ 2 fold; adjusted p value ≤ 0.05) compared to controls. Of these, 113 tRFs showed increased levels, while 143 tRFs showed decreased levels in the LP rats. We focused our investigations on mitochondrial tRFs (mt-tRFs) that show significant alterations in both, adult pre-diabetic mice (db/db) and LP rats. We selected the mt-tRF-LeuTAA fragment for further investigation, as its level is reduced in the islets of db/db mice and LP rats, while being abundant in ß-cells. mt-tRF-LeuTAA fragment is derived from the cleavage of tRNA-LeuTAA encoded by the mitochondrial genome. We demonstrated that mt-tRF-LeuTAA acts as a key regulator of mitochondrial OXPHOS functions, mitochondrial membrane potential, the insulin secretory capacity of ß-cells, and the insulin sensitivity of myotube muscle cells.

Development of a physiological insulin resistance model in human stem cell-derived adipocytes

Project description:Adipocytes are key regulators of human metabolism, and their dysfunction in insulin signaling is central to metabolic diseases including type II diabetes mellitus (T2D). However, the progression of insulin resistance into T2D is still poorly understood. This limited understanding is due, in part, to the dearth of suitable models of insulin signaling in human adipocytes. Traditionally, adipocyte models fail to recapitulate in vivo insulin signaling, possibly due to exposure to supraphysiological nutrient and hormone conditions. We developed a protocol for hPSC-derived adipocytes that uses physiological nutrient conditions to produce a potent insulin response comparable to in vivo adipocytes. After systematic optimization, this protocol allows robust insulin-stimulated glucose uptake and transcriptional insulin response. Furthermore, exposure of sensitized adipocytes to physiological hyperinsulinemia dampens insulin-stimulated glucose uptake and dysregulates insulin-responsive transcription. Overall, our methodology provides a novel platform for the mechanistic study of insulin signaling and resistance using human pluripotent stem cell-derived adipocytes.
