Expression Analysis of Geobacter sulfurreducens Current-Consuming Biofilm versus No-Current Biofilm

Project description:There is a wide diversity of potential applications for direct electron transfer from electrodes to microorganisms, which might be better optimized if the mechanisms for this novel electrode-biofilm interaction were further understood. Geobacter sulfurreducens is one of the few microorganisms available in pure culture that is known to be capable of directly accepting electrons from a negatively poised electrode. Gene transcript abundance in cells of G. sulfurreducens using electrons delivered from a graphite electrode as the sole electron donor for fumarate reduction was compared with transcript abundance in cells growing on the same graphite material, but without an electrical connection and acetate as the electron donor.

Function of Periplasmic Hydrogenases in the Sulfate-Reducing Bacterium Desulfovibrio vulgaris Hildenborough

Project description:The sulfate-reducing bacterium Desulfovibrio vulgaris Hildenborough possesses four periplasmic hydrogenases to facilitate the oxidation of molecular hydrogen. These include an [Fe], a [NiFeSe] and two [NiFe] hydrogenases encoded by the hyd, hys, hyn1 and hyn2 genes, respectively. In order to understand their cellular functions the expression levels of these hydrogenases, along with the growth rate analysis of mutant strains, was determined during growth on defined media under 3 different conditions. These conditions incuded lactate or hydrogen at either 5% or 50% (vol/vol) used as the sole electron donor for sulfate reduction. Keywords: Electron donor change

The Pd (II) reduction mechanisms in Bacillus megaterium Y-4 revealed by proteomic analysis

Project description:Many studies have been conducted on microbial reduction of Pd (II) to palladium nanoparticles (PdNPs) due to the environmental friendliness, low cost and the decreased toxicity of Pd (II) ions. In this study, we investigate the reduction mechanism of Pd (II) by Bacillus megaterium Y-4 through proteomics. Our results revealed that B. megaterium Y-4 may use the endogenous electron donor (NAD(P)H) generated by nirB, tdh and fabG and reductase to reduce Pd (II) to PdNPs. The expression levels of fabG, tdh, gudB and rocG that generates NAD(P)H were further increased, and the amount of reduced PdNPs was further increased in the presence of exogenous electron donor sodium formate. Endogenous electron mediators such as quinones and flavins in B. megaterium Y-4, can further enhance Pd (II) reduction. The findings provided invaluable information regarding the reduction mechanism of Pd (II) by B. megaterium Y-4 at the proteome level.

Nitrate reduction by Geobacter sulfurreducens and the catalytic role of palladium on the reduction pathway

Project description:Geobacter sulfurreducens is a widely explored microorganism recognized by its metabolic versatility able to reduce a number of external electron acceptors. In the present study the capacity of this strain to reduce nitrate was evaluated along with its transcriptomic profile under nitrate-reducing conditions and the catalytic role of Pd nanoparticles on the reductive pathway. Results demonstrated that G. sulfurreducens was able to reduce nitrate and important kinetic differences related to the time response were found among the electron donors used (acetate and hydrogen). When using acetate, a delay response on nitrate reduction of 4 days and reduction of 94% of nitrate was achieved, while nitrite was not detected, and all the nitrogen was recovered as ammonium (79.6 ± 5.7 %). The use of hydrogen as electron donor increased 2-fold the maximum rate of nitrate reduction, leading to 93% reduction of nitrate during the first 20 h with recovery of 45% as ammonium, while nitrite was not detected. In addition, transcriptome profiling analysis of G. sulfurreducens under nitrate-reducing conditions using hydrogen or acetate as an electron donor at 2 and 6 days reveals that a core of 146 genes (69 upregulated and 77 downregulated) are differentially expressed in all conditions. Genes related to nitrogen metabolism, such as nrfA and nrfH, gdhA, and amtB, were upregulated in the incubations and RT-qPCR data confirmed upregulations of these genes. Experiments performed with biologically synthesized Pd (Bio-Pd) + G. sulfurreducens cells demonstrated synergistic input of Bio-Pd and the metabolic capacity of G. sulfurreducens. These results expand the metabolic versatility of G. sulfurreducens, which may have important implications in nitrogen cycling in natural environments and engineered systems.

Arabidopsis transcriptional profile in response to chromate (K2CrO4)

Project description:Transcriptional profiling of Arabidopsis thalina seedlings in response to high (140 M-BM-5M) and low (20 M-BM-5M) concentrations of chromate (K2CrO4). Low concentrations of chromate (e.g. 40 M-BM-5M) promoted primary root growth, while high concentrations (e.g. 140 M-BM-5M) repressed growth and increased formation of root hairs, lateral roots and adventitious roots. Three-condition experiment, high (140 M-BM-5M) and low (20 M-BM-5M) concentrations of chromate (K2CrO4) vs non-chromate added (Control). Four biological replicates, one replicate per array.
