Project description:We detected different deregulated RNAs in LEVs and SEVs from patients with Alzheimer's, Parkinson's Disease, Amyotrophic Lateral Sclerosis, ALS. MiRNAs resulted to be the most interesting subpopulation of transcripts transported by plasma derived SEVs since they appeared to discriminate all NDs disease from CTRLs and they can provide a signature for each NDs. Common enriched pathways for SEVs were mainly linked to ubiquitin mediated proteolysis and Toll-like receptor signaling pathways and for LEVs to neurotrophin signaling and Glycosphingolipid biosynthesis pathway.

Location and function of TDP-43 in platelets: Alterations in neurodegenerative diseases and arising considerations for current plasma biobank protocols

Project description:The TAR DNA Binding Protein (TDP-43) has been implicated in the pathogenesis of human neurodegenerative diseases and exhibits hallmark neuropathology in amyotrophic lateral sclerosis (ALS). Here, we explore its tractability as a plasma biomarker of disease and describe its localization and possible functions in the cytosol of platelets. Novel TDP-43 immunoassays were developed on three different technical platforms and qualified for specificity, signal-noise ratio, detection range, variation, spike recovery and dilution linearity in human plasma samples. Fractionation studies revealed that >95% of plasma TDP-43 protein [RL1] was located within the platelet cytosol, together with numerous RNAs. Platelet-derived TDP-43 exhibits TDP-43 proteoforms detected in neurodegenerative diseases, TARDBP RNA splice variants and TDP-43 RNA targets found in the central nervous system (CNS). We propose that TDP-43 serves similar functional roles in platelets and synapses, suggesting that the study of platelet TDP-43 might provide a window into TDP-43 proteinopathies within the CNS. The restricted compartmentalization of plasma TDP-43 in platelets provides a highly concentrated substrate for further biochemical analyses. Moreover, our results suggest that current plasma biobanking protocols are subject to considerable heterogeneity in platelet recovery and measurements of TDP-43 in plasma.

Project description:We have analyzed six different extracelular vesicles isolation protocols from plasma of patients coinfected with human immunodeficiency virus and hepatitis C virus (HIV/HCV). In addition, two different RNA isolation kit were used for each protocol, one phenol-based and another pehenol-free kit. Same library kit was used for all samples. Small RNA, including miRNAs, piwiRNA, and sncRNAs were detected.

Different miRNA Profiles in Plasma Derived Small and Large Extracellular Vesicles from Patients with Neurodegenerative Diseases.

Project description:Identifying biomarkers is essential for early diagnosis of neurodegenerative diseases (NDs). Large (LEVs) and small extracellular vesicles (SEVs) are extracellular vesicles (EVs) of different sizes and biological functions transported in blood and they may be valid biomarkers for NDs. The aim of our study was to investigate common and different miRNA signatures in plasma derived LEVs and SEVs of Alzheimer's disease (AD), Parkinson's disease (PD), Amyotrophic Lateral Sclerosis (ALS) and Fronto-Temporal Dementia (FTD) patients. LEVs and SEVs were isolated from plasma of patients and healthy volunteers (CTR) by filtration and differential centrifugation and RNA was extracted. Small RNAs libraries were carried out by Next Generation Sequencing (NGS). MiRNAs discriminate all NDs diseases from CTRs and they can provide a signature for each NDs. Common enriched pathways for SEVs were instead linked to ubiquitin mediated proteolysis and Toll-like receptor signaling pathways and for LEVs to neurotrophin signaling and Glycosphingolipid biosynthesis pathway. LEVs and SEVs are involved in different pathways and this might give a specificity to their role in the spreading of the disease. The study of common and different miRNAs transported by LEVs and SEVs can be of great interest for biomarker discovery and for pathogenesis studies in neurodegeneration.

Circulating total extracellular vesicles cargo are associated with age-related oxidative stress and susceptibility to cardiovascular diseases: exploratory results from microarray data

Project description:Aging is a risk factor for many non-communicable diseases such as cardiovascular and neurodegenerative diseases. Aging could impact the extracellular vesicles and particles (EVPs) miRNA profile and impair redox homeostasis, contributing to chronic age-related diseases. We aimed to investigate the microRNA profiles of circulating total EVPs from aged and young adult animals. Plasma from 3- and 21-month-old male Wistar rats was collected and circulating total EVPs were isolated. MicroRNA isolation and microarray expression analysis were performed on EVPs to determine the predicted regulation of targeted mRNAs. 31 mature miRNAs in circulating EVPs were impacted by age and predicted to target molecules in canonical pathways directly related to cardiovascular diseases and oxidative status. Our data show that circulating total EVP cargo, specifically microRNAs, are involved in redox imbalance in the aging process and can potentially drive cardiovascular aging and consequently cardiac disease.
