Niche differentiation of arsenic-transforming microbial groups in rice rhizosphere compartments as impacted by water management and soil-As concentrations

Project description:Arsenic (As) bioavailability in the rice rhizosphere is influenced by many microbial interactions, particularly by metal-transforming functional groups at the root-soil interface. This study was conducted to examine As-transforming microbes and As-speciation in the rice rhizosphere compartments, in response to two different water management practices (continuous and intermittently flooded), established on fields with high to low soil-As concentration. Microbial functional gene composition in the rhizosphere and root-plaque compartments were characterized using the GeoChip 4.0 microarray. Arsenic speciation and concentrations were analyzed in the rhizosphere soil, root-plaque, porewater and grain samples. Results indicated that intermittent flooding significantly altered As-speciation in the rhizosphere, and reduced methyl-As and AsIII concentrations in the pore water, root-plaque and rice grain. Ordination and taxonomic analysis of detected gene-probes indicated that root-plaque and rhizosphere assembled significantly different metal-transforming functional groups. Taxonomic non-redundancy was evident, suggesting that As-reduction, -oxidation and -methylation processes were performed by different microbial groups. As-transformation was coupled to different biogeochemical cycling processes establishing functional non-redundancy of rice-rhizosphere microbiome in response to both rhizosphere compartmentalization and experimental treatments. This study confirmed diverse As-biotransformation at root-soil interface and provided novel insights on their responses to water management, which can be applied for mitigating As-bioavailability and accumulation in rice grains.

Overexpression of the HRD gene in Arabidopsis

Project description:Freshwater is a limited and dwindling global resource; therefore, efficient water use is required for food crops that have high water demands, such as rice, or for the production of sustainable energy biomass. We show here that expression of the Arabidopsis HARDY (HRD) gene in rice improves water use efficiency, the ratio of biomass produced to the water used, by enhancing photosynthetic assimilation and reducing transpiration. These drought tolerant low-water-consuming rice plants exhibit increased shoot biomass under well irrigated conditions and an adaptive increase in root biomass under drought stress. The HRD gene, an AP2/ERF-like transcription factor, identified by a gain-of-function Arabidopsis mutant hrd-D having roots with enhanced strength, branching, and cortical cells, exhibits drought resistance and salt tolerance, accompanied by an enhancement in the expression of abiotic stress associated genes. Although HRD overexpression in Arabidopsis produces thicker leaves with more chloroplast-bearing mesophyll cells, in rice there is an increase in leaf biomass and bundle sheath cells that probably contribute to the enhanced photosynthesis assimilation and efficiency. HRD overexpression was also studied for clues of molecular mechanisms involved using microarrays. The results exemplify application of a gene identified from the model plant Arabidopsis for the improvement of water use efficiency coincident with drought resistance in the crop plant rice. Succinylome analyses in the germinating rice seed

Nipponbare) seeds were washed with distilled water for three times and then imbibed with distilled water in dark growth chamber at 26 °C and 70% relative humidity. About 100 embryos (50 mg) of rice seeds were sliced manually and collected at intervals of 24h after imbibitions (HAI), respectively. After frozen with liquid nitrogen, the samples were stored in −80 °C until used for protein extraction.

Oxidative stress-triggered interactions between the succinyl- and acetyl-proteomes of rice leaves

Project description:Protein lysine acylation, such as succinylation and acetylation are important posttranslational modification (PTM) mechanisms, with key roles in the regulation of diverse metabolic and cellular processes. Antibody-based affinity enrichment, high-resolution LC-MS/MS analysis and integrated bioinformatics analysis, were used to characterize the lysine succinylome (Ksuc) and acetylome (Kace) in rice leaves. Rice (Oryza sativa., Wuyunjing 7) was kindly provided byJiangsu Academy of Agricultural Sciences, Jiangsu Province, China. Rice seeds were surface-sterilized with 5% NaClO for 20 min, washed extensively with distilled water and then germinated in distilled water at 28ºC for 2 days. Germinated seeds were cultivated on half-strength ammonium-free Murashige and Skoog (MS) medium. For oxidative stress treatments, 14-days-old rice seedlings were transferred to 1/2 MS medium in the absence or presence of 1 mM H2O2 for 6 days. Rice seedlings were then harvested for protein extraction.
