Project description:We examined the role of cytolytic strength on T cell fate decisions made by functionally competent CTLs under non-inflammatory priming conditions. Employing the HY-specific miHAg system in conjunction with non-inflammatory priming conditions, we demonstrate a higher cytolytic efficacy despite unchanged effector frequencies in the primary response. In Ebag9-/- mice, an expanded memory population was obtained for anti HY- and anti SV40 T antigen-specific CD8+ T cells.

Bach Factors Operate Erythro-Myeloid Differentiation by Responding to Infection

Project description:Hematopoietic cell differentiation should be tightly regulated in accordance with environmental changes to keep homeostasis. Infection is one of the conditions that induce myelopoiesis to exclude pathogens and repress erythropoiesis, which might be beneficial for the restriction of nutritional iron for pathogens. While several transcription factors (TFs), including C/EBP family and Gata1, play central roles in erythro-myeloid differentiation, the precise mechanism which controls the differentiation for the adaptation to infection remains obscure. In this study, it was revealed that Bach factors induce erythropoiesis and repress myelopoiesis in the erythro-myeloid bifurcation step, and their expressions per se were suppressed by infectious stimuli. Hence, they work as balancers of the erythro-myeloid differentiation between steady state and infectious state. These findings give us new insight into the machinery which regulates the fate of hematopoietic cells by responding to surrounding environments.

Gene expression profiles of neonatal cardiomyocytes and immortalized heart muscle cells exposed to inflammatory stimuli

Project description:We used a transcriptomic approach to systematically compare the response to inflammatory stimuli for two murine cardiomyocyte models. Our results indicate that murine neonatal cardiomyocytes (NNC) are able to exhibit a pronounced response upon inflammatory stimuli, while HL-1 cells were predominantly recruiting transcripts corresponding to Jak-Stat-signalling. RNA from murine HL-1 cardiomyocytes and isolated neonatal cardiomyocytes (NNC) stimulated by infectious (PCI) serum or LPS or a cytokine mix (CM) were extracted (n=4 each group) and subjected to microarray analysis for comparison of transcriptomic responses.

Project description:We used microarrays to detail the global programme of gene expression in human macrophages infected in vitro with HIV-1 and then subjected to phagocytic stimuli; we identified distinct classes of differentially regulated genes during this process Primary human macrophages from 3 different donors were infected with HIV-1ADA WT or mock-infected for 8 days. They were incubated or not (basal) with IgG-SRBCs (FcR) or Salmonella Typhimurium (4/74 strain) during 1 hour at 37Â°C, then lysed and RNA of each sample was analyzed with microarrays.

Gene expression profiling of grape-bud response to two alternative dormancy-release stimuli.

Project description:A grape-bud-oriented genomic platform was produced for a large-scale comparative analysis of bud responses to two stimuli of grape-bud dormancy release, hydrogen cyanamide (HC) and heat shock (HS). The results suggested considerable similarity in bud response to the stimuli, both in the repertoire of responding genes and in the temporary nature of the transcriptome reprogramming. Nevertheless, the bud response to HC was slower, more condensed and stronger, as reflected by a higher number of regulated genes and a higher intensity of regulation compared to the response to HS.
