Project description:Ovarian cancer is a heterogeneous disease that constitutes great menace to female health and poses tremendous clinical challenge. Based on unsupervised classification of whole-genome gene expression profiles, four molecular subtypes of ovarian cancer were recently identified. However, currently it is still unelucidated on the immune infiltration pattern of different molecular subtypes of ovarian cancer. we used a computational approach (CIBERSORT) to bulk gene expression profiles of ovarian cancer datasets to infer the proportions of 22 subsets of immune cells in the four subtypes. We found that plasma cells are enriched in the mesenchymal subtype of ovarian cancer and the content of plasma cells was correlated with the density of mesenchymal cells in clinical specimens of ovarian cancer. Co-culture of non-mesenchymal subtype ovarian cancer cell lines with plasma cells could induce a mesenchymal phenotype and enhance the metastatic potential of ovarian cancer cells. Mechanistically, this phenotypic switch was mediated by the transfer of miR-330-3p-containing exosomes to the non-mesenchymal ovarian cancer cells.miR-330-3p, which subsequently activate gene transcription of junctional adhesion molecule B(JAM2) epigenetically as an enhancer trigger. Collectively, our work identifies an immune-associated cellular, molecular, and clinical network that highlights the defining role of plasma cells in the mesenchymal identify of ovarian cancer.

Plasma cells shape the mesenchymal identity of ovarian cancers through transfer of exosome-derived microRNAs.

Project description:Ovarian cancer represents a highly lethal disease that poses a substantial burden for females, with four main molecular subtypes carrying distinct clinical outcomes. Here, we demonstrated that plasma cells, a subset of antibody-producing B cells, were enriched in the mesenchymal subtype of high-grade serous ovarian cancers (HGSCs). Plasma cell abundance correlated with the density of mesenchymal cells in clinical specimens of HGSCs. Coculture of nonmesenchymal ovarian cancer cells and plasma cells induced a mesenchymal phenotype of tumor cells in vitro and in vivo. Phenotypic switch was mediated by the transfer of plasma cell-derived exosomes containing miR-330-3p into nonmesenchymal ovarian cancer cells. Exosome-derived miR-330-3p increased expression of junctional adhesion molecule B in a noncanonical fashion. Depletion of plasma cells by bortezomib reversed the mesenchymal characteristics of ovarian cancer and inhibited in vivo tumor growth. Collectively, our work suggests targeting plasma cells may be a novel approach for ovarian cancer therapy.

Integrated analysis identifies key determinants of embryonic stem cell identity and homeostasis

Project description:Despite RNAi-based screens to uncover genes controlling embryonic stem cell (ESC) identity, the pluripotency network remains poorly characterized, as does the precise molecular mechanisms underlying the balance between self-renewal and differentiation. Here we carried out a systematic meta-analysis of published gene expression data to rank-order genes based on their likelihood of regulating ESC identity. Not only did our analysis correctly rank known pluripotency-associated genes atop the list, but it also helped unearth many novel determinants of ESC identity including several components of functionally distinct complexes, as determined using RNAi. We focus on our top-hit Nucleolin, and characterize its mechanistic role in the maintenance of ESC homeostasis by shielding from differentiation-inducing redox imbalance-induced oxidative stress. Notably, we identify a conceptually novel mechanism involving a Nucleolin-dependent bistable switch regulating the homeostatic balance between self-renewal and differentiation in ESCs. Our gene ranks represent a rich and valuable resource for uncovering novel ESC regulators. Single cell RNA sequencing of thymic dendritic cells reveals molecular determinants of antigen transfer in the thymus

Project description:The presentation of self-antigens in the thymus by medullary thymic epithelial cells (mTECs) and dendritic cells (DCs) is crucial for the establishment of central tolerance. While mTECs produce and present self-antigens in an autonomous manner, DCs acquire them from mTECs by Cooperative Antigen Transfer (CAT). Although CAT represents a key process of central tolerance, molecular determinants which drive CAT are largely unknown. To reveal these determinants, we compared the transcriptomes of CAT-experienced and CAT-inexperienced DCs via scRNAseq. Specifically, we sequenced TdTOMATO+ and TdTOMATO– CD11c+ and CD11b+ cells from the thymus of Foxn1CreR26TdTOMATO mice. In this model, fluorescent TdTOMATO protein is exclusively produced by thymic epithelium, thus TdTOMATO expression in DCs (CD11c+ and CD11b+ cells) mark their previous experience of CAT. Using this approach we unraveled several candidate determinants of CAT. We focused on the gene determinant, Cldn1, encoding the tight junction protein Claudin 1, that was found to be highly expressed among the CAT-experienced type 1 lineage DCs. We performed further experiments that confirmed its role in driving CAT. In addition, this scRNAseq approach allowed us to redefine the heterogeneity and maturation states of thymic DCs.
