STING activation promotes robust immune response and NK cell- mediated tumor regression in glioblastoma models

Project description:Immunotherapy has had a tremendous impact on cancer treatment in the past decade, with hitherto unseen responses at advanced and metastatic stages of the disease. However, the aggressive brain tumor glioblastoma (GBM) is highly immunosuppressive and remains largely refractory to current immunotherapeutic approaches. The cGAS-STING cytoplasmic double stranded DNA (dsDNA) sensing pathway has emerged as a next-generation immunotherapy target with potent local immune stimulatory properties. Here, we investigated the status of the STING pathway in GBM and the modulation of the brain tumor microenvironment (TME) with the STING agonist ADU-S100. Our data reveal the presence of STING in human GBM specimens, where it stains strongly in the tumor vasculature. We show that human GBM explants can respond to STING agonist treatment by secretion of inflammatory cytokines. In murine GBM models, we show a profound shift in the tumor immune landscape after STING agonist treatment, with massive infiltration of the tumor-bearing hemisphere with innate immune cells including inflammatory macrophages, neutrophils and NK populations. Treatment of established murine intracranial GL261 and CT-2A tumors by biodegradable ADU-S100-loaded intracranial implants demonstrated a significant increase in survival in both models and long-term survival with immune memory in GL261. Responses to treatment were abolished by NK cell depletion. This study reveals therapeutic potential and deep remodeling of the TME by STING activation in GBM and warrants the further examination of STING agonists alone or in combination with other immunotherapies such as cancer vaccines, CAR T cells, NK therapies or immune checkpoint blockade.

BJ cyto-nuclear translocation upon STING activation

Project description:The goal of this project is to identify changes of cyto-nuclear import upon STING activation by cGAMP. BJ cells stably expressing TurboID-NES were treated with biotin to label cytoplasmic proteins, after which cells were further treated with or without cGAMP to activate STING signaling. Cells were then fractionated to collect the nuclei. The biotinylated proteins in the nuclear pellets were purified by streptavidin beads for mass spectrometry analysis.

The effect of myeloid STING knockout after tumor formation on gene expression in AOM/DSS-induced CAC mice

Project description:Myeloid-derived STING has been recognized to play a vital role in mediating the development of colitis-associated carcinoma (CAC). We here report that myeloid-specific knockout of STING after tumor formation enhanced tumor growth by modifying the tumor microenvironment to a more immunologically inactive state. Pathways related to antigen presentation, macrophage and DC activation, T cell chemotaxis and activation, T cell-mediated cytotoxicity and other immune responses to tumor cells were all inhibited by lateral myeloid STING kncckout after tumor formation.

Context-dependent activation of STING-interferon signaling by CD11b agonists enhances anti-tumor immunity

Project description:Chronic activation of inflammatory pathways and suppressed interferon are hallmarks of immunosuppressive tumors. Previous studies have shown that CD11b integrin agonists could enhance anti-tumor immunity through myeloid reprograming, but the underlying mechanisms remain unclear. Herein we find that CD11b agonists alter tumor-associated macrophage (TAM) phenotypes by repressing NF-κB signaling and activating interferon gene expression simultaneously. Repression of NF-κB signaling involves degradation of p65 protein and is context independent. In contrast, CD11b agonism induces STING/STAT1 pathway-mediated interferon gene expression through FAK-mediated mitochondrial dysfunction, with the magnitude of induction dependent on the tumor microenvironment and amplified by cytotoxic therapies. Using tissues from phase I clinical studies, we demonstrate that GB1275 treatment activates STING and STAT1 signaling in TAMs in human tumors. These findings suggest potential mechanism-based therapeutic strategies for CD11b agonists and identify patient populations more likely to benefit.

Project description:Bendamustine plus rituximab (BR) therapy has demonstrated favorable clinical outcomes in the treatment of DLBCL patients who are intolerant to R-CHOP therapy. In vitro, BR therapy exhibits a synergistic cytotoxic effect on DLBCL cell lines. Mechanistically, BR induces pyroptosis in DLBCL cells by activating the cGAS-STING pathway, leading to the release of inflammatory factors and the formation of an "immunologically hot" microenvironment. Additionally, BR therapy upregulates MHC molecules on the tumor cell surface, thereby augmenting T cell activation and function.
