Identifying heterogeneity of Schwann cell like cells via scRNA-seq.

Project description:Previously we have reported that Schwann cell like cells (SCLCs) induced from human amniotic mesenchymal stem cells (hAMSCs) have better peripheral nerve regeneration ability, but the heterogeneity of SCLCs is still poorly understood. Here, based on single-cell RNA sequencing, we investigated the heterogeneity of hAMSCs and SCLCs, and identified the transition from hAMSCs to SCLCs. Totally 6,008 and 5,140 cells generated from hAMSCs and SCLCs were profiled and used for deep analysis. A subpopulation of SCLCs highly expressed Schwann cell markers, JUN, JUND, and NRG1, was identified. Then, cell cycle analysis distinguished the proliferation state of each cluster, and single cells of the subpopulation in SCLCs mainly remained in G0/G1 phase. Pseudotime analysis illustrated how hAMSCs transformed into SCLCs, hAMSCs showed more possibility transformed into other cell types. Through marker gene identification, pathway enrichment, transcription factor analysis, and secreted protein validation, genes such as NRG1, PTGS2, PITX1, VEGFA, and FGF2 were highly expressed in single cells. And also, pathways associated with nerve regeneration and immune control were enriched, such as angiogenesis or TNF signaling.

Project description:The extent of transcriptional diversity in mouse NPCs is likely to be influenced by a variety of unexamined factors that include programmed cell death, genomic mosaicism as well as a variety of âenvironmentalâ influences such as changes in exposure to signaling lipids. We therefore used scRNA-seq to assess a cohort of cortical NPCs from an embryonic mouse. We demonstrate that PAGODA (Pathway And Geneset OverDispersion Analysis) effectively recovers the known neuroanatomical and functional organization of NPCs, identifying multiple aspects of transcriptional heterogeneity within the developing mouse cortex that are difficult to discern by the existing heterogeneity analysis approaches. Schwann Cells Shape Tumor Cells and Cancer-Associated Fibroblasts in the Pancreatic Ductal Adenocarcinoma Microenvironment [scRNA-seq]

Project description:Neuropathy is a feature more frequently observed in pancreatic ductal adenocarcinoma (PDAC) than other tumors. Schwann cells, the most prevalent cell in peripheral nerves, migrate toward tumor cells and associate with poor prognosis in PDAC. To unveil the effects of Schwann cells on the neuro-stroma niche, single-cell RNA-sequencing and microarray-based spatial transcriptome analysis of PDAC tissues were performed. Results suggested that Schwann cells may drive tumor cells and cancer-associated fibroblasts (CAFs) to more malignant subtypes: basal-like and inflammatory CAFs (iCAFs), respectively. Moreover, in vitro and in vivo assays demonstrated that Schwann cells enhanced the proliferation and migration of PDAC cells via Midkine and promoted the switch of CAFs to iCAFs via interleukin-1a. By Schwann cells condition medium, tumor cells together with iCAFs accelerate PDAC progression. Thus, we revealed, for the first time, that Schwann cells induce malignant subtypes of tumor cells and CAFs in the PDAC milieu.

In this study, we performed single cell RNA-sequencing (scRNA-seq) analysis of Schwann cells exist in both dorsal root ganglion(DRG) and sciatic nerve.We categorized DRG and sciatic nerve Schwann cells into different subtypes,and found common subtypes and different subtypes.In addition, we discovered the proliferation and migration ability of Schwann cells were distinct in different tissues.Our current study revealed the distinctive characteristics of Schwann cells in DRG and sciatic nerve.
