Project description:Primary objectives: The primary objective is to investigate circulating tumor DNA (ctDNA) via deep sequencing for mutation detection and by whole genome sequencing for copy number analyses before start (baseline) with regorafenib and at defined time points during administration of regorafenib for treatment efficacy in colorectal cancer patients in terms of overall survival (OS). Expression analysis of a full SlrV locus 1 knockout in Haemophilus influenzae strain PittII

Project description:Investigation of whole genome gene expression differences between a full (four-gene) SlrV locus 1 deletion compared to the WT PittII strain. Removal of the four SlrV genes (aka macrophage survival factors : ?msf) results in a competetive defect with the WT strain in in vitro biofilms and in the otitis media chinchilla model. It also results in an inability to persist within macrophages long-term in vitro and to escape from the middle-ear space and infect other sites in the chinchilla model. WT and ?msf strains were grown in a chemically defined media (CDM) to early stationary phase (OD ~1.0). RNA was extracted from two separate cultures (biological replicates). 3-13 probes represent each subcluster(gene) and two replicate probe sets are included on each custom NTHi supragenome chip (technical replicates) thus under each sampling condition 4 separate values are obtained per strain and used for WT:Msf-KO comparisons.

Clinicopathologic, immunohistochemical, and ultrastructural features of histiocytic sarcoma in a chinchilla (Chinchilla lanigera).

Project description:A 9-year-old intact female chinchilla (Chinchilla lanigera) was presented to a referring veterinarian due to small, multiple cervical nodules that had been rapidly increasing in size and number. Cytology of the nodules revealed sheets of pleomorphic round cells that were morphologically most compatible with histiocytic sarcoma. Histologically, the nodules were fairly demarcated, partially infiltrative, densely cellular neoplasm, and was composed of pleomorphic large round cells arranged in sheets. Special stains for bacteria (Gram stain and Ziehl-Neelsen stain) and fungi (periodic acid-Schiff stain) were all negative. On immunohistochemistry, the neoplastic cells showed strong cytoplasmic positivity for Iba-1 and CD204, but were negative for CD3 and CD20. Transmission electron microscopy failed to detect Birbeck's granules in the cytoplasm of the neoplastic histiocytes. The chinchilla received chemotherapy with lomustine but died spontaneously on day 62 despite treatment. Autopsy with histopathologic examination revealed disseminated histiocytic sarcoma involving the bone marrow, bronchial lymph nodes, nasal cavity, lung, heart, stomach, pancreas, pancreatic lymph nodes, liver, spleen, and kidney. To the best of our knowledge, this is the first report of disseminated histiocytic sarcoma in chinchillas.

Prospective Collection of Surplus Surgical Tumor Tissues and Pre-surgical Blood Samples

Project description:The study is intended to collect specimens to support the application of genome analysis technologies, including large-scale genome sequencing. This study will ultimately provide cancer researchers with specimens that they can use to develop comprehensive catalogs of genomic information on at least 50 types of human cancer. The study will create a resource available to the worldwide research community that could be used to identify and accelerate the development of new diagnostic and prognostic markers, new targets for pharmaceutical interventions, and new cancer prevention and treatment strategies. This study will be a competitive enrollment study conducted at multiple institutions.

Occurrence of Giardia duodenalis assemblages in farmed long-tailed chinchillas Chinchilla lanigera (Rodentia) from Romania.

Project description:BACKGROUND:Giardia duodenalis is a parasitic protist that infects a large number of species, being localized in the small intestine. Two of the eight recognized assemblages have zoonotic potential, but studies regarding their distribution in less important pet or farm species are scarce. Of these species, the long-tailed chinchilla is a host for Giardia spp., although data on the spread of infection and assemblages involved are confined. The present work aimed to determine the prevalence of Giardia infection and assemblage identification in farmed chinchillas in Romania. A total of 341 fecal samples were collected from 5 farms and microscopically examined using flotation test based on saturated sodium chloride solution. DNA from all positive samples was extracted and identified by PCR targeting the gdh gene. RESULTS:The overall prevalence of Giardia infection was 55.7% (190/341); there was no statistically significant difference (P = 0.25) in prevalence between young animals (58.8%) and adults (52.6%). Assemblages B (151/190), D (33/190) and E (6/190) were identified. Among assemblage B, sub-assemblages BIII (6/151) and BIV (145/151) were determined. CONCLUSIONS:This study demonstrates that Giardia spp. infection is highly prevalent in farmed chinchillas from Romania, and the sub-assemblages identified are potentially zoonotic.
