Expression profiling of pitavastatin in a tumor-immune model in vitro

Project description:We developed a phenotypic screening platform that identified statins as enhancers of immune cell-induced cancer cell death. Statins are currently gaining interest as repurposing candidates for use in oncology as increasing evidence supports that statins exert anti-cancer effects. Considering also our findings, the clinical benefits described for cancer patients receiving statins might be due to a combined effect on both cancer- and immune cells. Studying gene expression profiles is a well established approach for investigating a compound's mode of action. We performed transcriptome-wide gene expression profiling on the human colorectal cancer cell line HCT116-GFP and peripheral blood mononuclear cells (PBMCs). Monocultured and co-cultured cells were treated with DMSO vehicle or pitavastatin (1µM or 10µM) for 24 hours before gene expression was analyzed using a Human Clariom S array.

Combined pitavastatin and capmatinib inhibits tumor growth though completed shutdown of MET signal

Project description:To reveal the function of pitavastatin in cancer, we performed several experiments. In the present study, we show that pitavastatin inhibits the tumor growth through the inactivation of ERK and AKT signals by MET immaturation, and combination of pitavastatin and capmatinib, a MET inhibitor, might be useful cancer therapy in OSCC and ESCC. It could be reduced the dose of pitavastatin by using this combination therapy, resulting in decreasing the risk of adverse events such as myopathy. Taken together, the mevalonate pathway regulates the processes of MET maturation and promotes the cell growth in cancer. Moreover, the expression of GGPS1, an enzyme that synthesizes geranylgeranyl pyrophosphate (GGPP) from farnesyl pyrophosphate (FPP), associates with the sensitivity of pitavastatin, suggesting that GGPS1 could be a biomarker in cancer therapy with pitavastatin. Deep sequencing of an AML cell line treated with vehicle control, BETi, MDM2i, or a drug combination

Project description:Gene expression changes in human OCI-AML3 cells treated with DMSO (vehicle control), the BET inhibitor CPI-203, the MDM2 inhibitor Nutlin-3a, or a combination of both drugs, were compared. Method: poly-A mRNA from the OCI-AML3 cell line treated for 24 hrs with either DMSO, CPI-203, Nutlin-3a or a combination of both drugs was extracted, in triplicate, then sequenced with an Illumina NextSeq500 sequencer. Sequence reads passing the quality control filters were aligned using Tophat2 and then analysed with Cufflinks.
