Strain NaphS2, an anaerobic sulfate reducing marine delta-proteobacterium is capable of using naphthalene and the aromatic compound benzoate, as well as pyruvate, as an electron donors in the presence of sulfate. In order to identify genes involved in the naphthalene degradation pathway, we compared gene expression in NaphS2 during growth on benzoate vs. pyruvate, naphthalene vs. pyruvate, and naphthalene vs benzoate.

Project description:The success of bottom-up proteomic analysis frequently depends on the efficient removal of contaminants from protein or peptide samples before LC-MS/MS. For a peptide clean-up workflow, the single-pot solid-phase-enhanced peptide sample preparation on carboxylate-modified paramagnetic beads (termed SP2) was evaluated for sodium dodecyl sulfate or polyethylene glycol removal from Arabidopsis thaliana tryptic peptides. The robust and efficient 40-min SP2 protocol, tested for a 10 ng, 250ng and 10µg peptide sample, was proposed and benchmarked thoroughly against the ethyl acetate extraction protocol. The SP2 protocol on carboxylated magnetic beads proved to be the robust approach even for simultaneous removal of massive sodium dodecyl sulfate (SDS) and polyethylene glycol (PEG) contaminations from AT peptide samples in respect of the LC-MS/MS data outperforming ethyl acetate extraction.

Project description:BackgroundBioelectrochemical methane oxidation catalysed by anaerobic methanotrophic archaea (ANME) is constrained by limited methane bioavailability as well as by slow kinetics of extracellular electron transfer (EET) of ANME. In this study, we tested a combination of two strategies to improve the performance of methane-driven bioelectrochemical systems that includes (1) the use of hollow fibre membranes (HFMs) for efficient methane delivery to the ANME organisms and (2) the amendment of ferricyanide, an effective soluble redox mediator, to the liquid medium to enable electrochemical bridging between the ANME organisms and the anode, as well as to promote EET kinetics of ANME.ResultsThe combined use of HFMs and the soluble mediator increased the performance of ANME-based bioelectrochemical methane oxidation, enabling the delivery of up to 196 mA m-2, thereby outperforming the control system by 244 times when HFMs were pressurized at 1.6 bar.ConclusionsImproving methane delivery and EET are critical to enhance the performance of bioelectrochemical methane oxidation. This work demonstrates that by process engineering optimization, energy recovery from methane through its direct oxidation at relevant rates is feasible.

Project description:Electro-bioremediation offers a promising approach for eliminating persistent pollutants from groundwater since allows the stimulation of biological dechlorinating activity, utilizing renewable electricity for process operation and avoiding the injection of chemicals into aquifers. In this study, a two-chamber microbial electrolysis cell has been utilized to achieve both reductive and oxidative degradation of tetrachloroethane (TeCA). By polarizing the graphite granules cathodic chamber at -650 mV vs the standard hydrogen electrode and employing a mixed metal oxide (MMO) counter electrode for oxygen production, the reductive and oxidative environment necessary for TeCA removal has been established. Continuous experiments were conducted using two feeding solutions: an optimized mineral medium for dechlorinating microorganisms, and synthetic groundwater containing sulphate and nitrate anions to investigate potential side reactions. The bioelectrochemical process efficiently reduced TeCA to a mixture of trans-dichloroethylene, vinyl chloride, and ethylene, which were subsequently oxidized in the anodic chamber with removal efficiencies of 37 ± 2%, 100 ± 4%, and 100 ± 5%, respectively. The introduction of synthetic groundwater with nitrate and sulphate stimulated reductions in these ions in the cathodic chamber, leading to a 17% decrease in the reductive dechlorination rate and the appearance of other chlorinated by-products, including cis-dichloroethylene and 1,2-dichloroethane (1,2-DCA), in the cathode effluent. Notably, despite the lower reductive dechlorination rate during synthetic groundwater operation, aerobic dechlorinating microorganisms within the anodic chamber completely removed VC and 1,2-DCA. This study represents the first demonstration of a sequential reductive and oxidative bioelectrochemical process for TeCA mineralization in a synthetic solution simulating contaminated groundwater.

Project description:Acetate as the dominant fraction of volatile fatty acids (VFAs) is an important intermediate in metabolic pathways of methanogenesis, which could reflect the stability status of anaerobic digestion (AD) process. Bioelectrochemical sensors for environmental or bioprocess monitoring have become increasingly attractive in recent years. Although it was more favorable, several challenges still need to be addressed for acetate detection, including large electrode spacing, low stability, biofouling at the cathode and low detection range. In this study, an innovative biosensor on the basis of a three-chamber microbial electrochemical system was proposed to monitor the acetate during the AD process. In such a system, acetate was first transferred from sample chamber through the anion exchange membrane (AEM) to anode due to the driven force of concentration difference and then oxidized by anodic biofilm as a substrate for the current generation. With such design, the influence of waste properties fluctuation in the cathodic reaction could be avoided. The response of current density to different acetate concentrations was investigated. The selectivity, the influence of the sample temperature and the external resistance were also evaluated. The correlation (R 2 > 0.99) between the current densities and acetate concentrations (up to 160 mM) was established at specific reaction time (from 2 to 5 h). Current densities after 5 h reaction were improving about 20% when the sample temperature was high (e.g., 37 and 55°C). The detection range increased along with the decrease of external resistance. The acetate concentrations of AD effluents as determined by the biosensor where within 24.2% of the ones determined by gas chromatography. Nevertheless, the application of the biosensor for monitoring acetate in environmental samples could still be promising.

Project description:A cylindrical bioelectrochemical reactor (BER) containing carbon fiber textiles (CFT; BER + CFT) has characteristics of bioelectrochemical and packed-bed systems. In this study, utility of a cylindrical BER + CFT for degradation of a garbage slurry and recovery of biogas was investigated by applying 10% dog food slurry. The working electrode potential was electrochemically regulated at -0.8 V (vs. Ag/AgCl). Stable methane production of 9.37 L-CH4 · L-1 · day-1 and dichromate chemical oxygen demand (CODcr) removal of 62.5% were observed, even at a high organic loading rate (OLR) of 89.3 g-CODcr · L-1 · day-1. Given energy as methane (372.6 kJ · L-1 · day-1) was much higher than input electric energy to the working electrode (0.6 kJ · L-1 · day-1) at this OLR. Methanogens were highly retained in CFT by direct attachment to the cathodic working electrodes (52.3%; ratio of methanogens to prokaryotes), compared with the suspended fraction (31.2%), probably contributing to the acceleration of organic material degradation and removal of organic acids. These results provide insight into the application of cylindrical BER + CFT in efficient methane production from garbage waste including a high percentage of solid fraction.

Project description:Nitrogen and arsenic contaminants often coexist in groundwater, and microbes show the potential for simultaneous removal of nitrogen and arsenic. Here, we reported that Hydrogenophaga sp. H7 was heterotrophic nitrification and aerobic denitrification (HNAD) and arsenite [As(III)] oxidation bacterium. Strain H7 presented efficient capacities for simultaneous NH4+-N, NO3--N, or NO2--N removal with As(III) oxidation during aerobic cultivation. Strikingly, the bacterial ability to remove nitrogen and oxidize As(III) has remained high across a wide range of temperatures, pH values, and shaking speeds, exceeding that of the most commonly reported HNAD bacteria. Additionally, the previous HNAD strains exhibited a high denitrification efficiency, but a suboptimal concentration of nitrogen remained in the wastewater. Here, strain H7 combined with FeCl3 efficiently removed 96.14% of NH4+-N, 99.08% of NO3--N, and 94.68% of total nitrogen (TN), and it oxidized 100% of As(III), even at a low nitrogen concentration (35 mg/L). The residues in the wastewater still met the Surface Water Environmental Quality Standard of China after five continuous wastewater treatment cycles. Furthermore, genome and proteomic analyses led us to propose that the shortcut nitrification-denitrification pathway and As(III) oxidase AioBA are the key pathways that participate in simultaneous nitrogen removal and As(III) oxidation.

Project description:A study was initiated to construct a micro-reactor for protein digestion based on trypsin-coated fused-silica capillaries. Initially, surface plasmon resonance was used both for optimization of the surface chemistry applied in the preparation and for monitoring the amount of enzyme that was immobilized. The highest amount of trypsin was immobilized on dextran-coated SPR surfaces which allowed the covalent coupling of 11 ng mm(-2) trypsin. Fused-silica capillaries were modified in a similar manner and the resulting open-tubular trypsin-reactors having a pH optimum of pH 8.5, display a high activity when operated at 37 degrees C and are stable for at least two weeks when used continuously. Trypsin auto-digestion fragments, sample carry-over, and loss of signal due to adsorption of the protein were not observed. On-line digestion without prior protein denaturation, followed by micro-LC separation and photodiode array detection, was tested with horse-heart cytochrome C and horse skeletal-muscle myoglobin. The complete digestion of 20 pmol microL(-1) horse cytochrome C was observed when the average residence time of the protein sample in a 140 cm x 50 microm capillary immobilized enzyme reactor (IMER) was 165 s. Mass spectrometric identification of the injected protein on the basis of the tryptic peptides proved possible. Protein digestion was favorable with respect to reaction time and fragments formed when compared with other on-line and off-line procedures. These results and the easy preparation of this micro-reactor provide possibilities for miniaturized enzyme-reactors for on-line peptide mapping and inhibitor screening.
