Molecular characterization of pyrethroid resistance in the olive fruit fly Bactrocera oleae

Project description:Α reduction of pyrethroid efficacy has been recently recorded in the olive fruit fly Bactrocera oleae, the most destructive insect pest of olives worldwide. We analyzed the transcriptomic differences between two highly pyrethroid resistant populations versus a relatively susceptible field population and two laboratory strains to gain more insight into the molecular mechanism of resistance. A large number of genes was found to be significantly differentially transcribed across the pairwise comparisons between resistant and susceptible insect populations. Interestingly, gene set analysis revealed that genes of the ‘electron carrier activity’ GO group were enriched in one specific pairwise transcriptomic comparison. As P450 monooxygenase enzymes are typically associated with this Molecular Function GO-group, this might reflect a P450-mediated resistance mechanism. These results suggest that transcriptional induction of the CYP6 P450s might be an important mechanism of pyrethroid resistance in B. oleae and pave the way for the development of synergists and molecular diagnostics for insecticide resistance management.

New insights into the reproductive transcriptome profile of the Mediterranean fruit fly Ceratitis capitata

Project description:Purpose: Use of RNAseq to identify crucial genes expressed in the reproductive tract of C. capitata The medfly, Ceratitis capitata continues to pose a major threat to the agricultural sector in almost every continent. Promiscuous female medflies can oviposit up to 1000 eggs throughout their lifetime causing remarkable losses in crop viability. Nowadays, there is an increasing demand for finding new improved solutions to counteract its invasive spreading capacity. To these regards, research that aims in understanding the reproductive biology of fruit fly pests by obtaining its genomic profiling can be of great contribution to this field. In this study we have generated a De-novo assembly of the medfly reproductive tract transcripts. We used Transcriptomics via deep RNA-sequencing to identify the transcripts present in the sex organs (spermatheca, uterus and accessory glands) of sexually matured virgin and 24hr-freshly mated females as well as testes, and male accessory glands of C. capitata. In addition, differential expression changes between virgin and mated females could help elucidate crucial candidate genes that participate in the female physiological response to the male seminal input. Furthermore, in order to ensure pure and significant results of the transcriptomic analysis, we have included a negative control that includes gut and abdominal adipose tissue. The newly annotated genes from this investigation could furthermore contribute to the ongoing field of biological and genetic population control.

Decoding the Reproductive System of the Olive Fruit Fly, Bactrocera oleae.

Project description:In most diploid organisms, mating is a prerequisite for reproduction and, thus, critical to the maintenance of their population and the perpetuation of the species. Besides the importance of understanding the fundamentals of reproduction, targeting the reproductive success of a pest insect is also a promising method for its control, as a possible manipulation of the reproductive system could affect its destructive activity. Here, we used an integrated approach for the elucidation of the reproductive system and mating procedures of the olive fruit fly, Bactrocera oleae. Initially, we performed a RNAseq analysis in reproductive tissues of virgin and mated insects. A comparison of the transcriptomes resulted in the identification of genes that are differentially expressed after mating. Functional annotation of the genes showed an alteration in the metabolic, catalytic, and cellular processes after mating. Moreover, a functional analysis through RNAi silencing of two differentially expressed genes, yellow-g and troponin C, resulted in a significantly reduced oviposition rate. This study provided a foundation for future investigations into the olive fruit fly's reproductive biology to the development of new exploitable tools for its control.

Proteomics of extra virgin olive oil (EVOO)

Project description:Proteins and peptides are minor components of vegetal oils. The presence of these compounds in virgin olive oil was first reported in 2001, but the nature of the olive oil proteome is still a puzzling question for food science researchers. In this project, we have compiled for a first time a comprehensive proteomic dataset of olive fruit and fungal proteins that are present at low but measurable concentrations in a vegetable oil from a crop of great agronomical relevance as olive (Olea europaea L.). Accurate mass nLC-MS data were collected in high definition direct data analysis (HD-DDA) mode using the ion mobility separation step. Protein identification was performed using the Mascot Server v2.2.07 software (Matrix Science) against an ad hoc database made of olive protein entries. Starting from this proteomic record, the impact of these proteins on olive oil stability and quality could be tested. Moreover, the effect of olive oil proteins on human health and their potential use as functional food components could be also evaluated. In addition, this dataset provides a resource for use in further functional comparisons across other vegetable oils, and also expands the proteomic resources to non-model species, thus also allowing further comparative inter-species studies.

Microarray analysis on fruit fly (Drosophila melanogaster) larvae fed with either a semi-purified diet only or a semi-purified diet supplemented with palm fruit juice (PFJ)

Project description:PFJ (4 ml for a final concentration of 19,000 mg gallic acid equivalent (GAE) per kg diet or 0.86 mg GAE per kcal diet) was supplemented to larvae of fruit flies (Drosophila melanogaster) given a semi-purified diet to observe for possible effects on energy metabolism and lifespan. Larvae were used five days since the egg stage for gene expression studies. Results from the microarray data analysis carried out show that fruit fly larvae given PFJ had up-regulated transport and metabolic processes, while development and morphogenesis processes were down-regulated.

Integration of tomato reproductive developmental landmarks and expression profiles, and the effect of SUN on fruit shape

Project description:Universally accepted landmark stages are necessary to highlight key events in tomato reproductive development. In this study, we provide a description of floral and fruit development in a red-fruited closely related wild relative of tomato, Solanum pimpinellifolium accession LA1589. We use established and propose new landmarks as the framework for the characterization of the tomato fruit shape gene SUN in fruit development. SUN controls fruit shape predominantly after fertilization and its effect reaches a maximum at 8 days post anthesis coinciding with fruit landmark 4 representing the globular embryo stage of seed development. We also analyzed gene expression profiles of floral buds 10 days before anthesis (floral landmark 7), anthesis-stage flowers (floral landmark 10 and fruit landmark 1), and 5 days post anthesis fruit (fruit landmark 3). The expression profiles of the NILs that differ at sun showed that 34 genes were differentially expressed and most of them at a less than 2-fold difference. However, many genes were differentially expressed between the developmental times points, including many genes involved in phytohormone biosynthesis or signaling as well as organ identity and patterning of tomato fruit.
