Proteome dataset of peripheral blood mononuclear cells in postpartum dairy cows supplemented with different sources of omega-3 fatty acids

Project description:This article contains raw and processed data related to research published by Kra et al. [1]. There is a scarce knowledge on the proteome of peripheral blood mononuclear cells (PBMC) during the transition period in dairy cows. In human research, proteomics PBMC is used in order to gain insight into inflammatory diseases and syndromes. Dietary fats, and specifically omega-3 (n-3) FA, can moderate the immune fluctuation caused by parturition through improvements of the immune function [2]. Therefore, this study aim was to characterize the changes that may occur in proteome of PBMC during transition, as influenced by different n-3 FA supplementation. Proteomics data of PBMC was obtained from postpartum dairy cows supplemented peripartum with either encapsulated saturated fat (CTL), encapsulated flaxseed oil that is enriched with ALA (α-linolenic acid; FLX) or encapsulated fish oil that is enriched with EPA (eicosapentaenoic acid) and DHA (docosahexaenoic acid; FO).The analysis was done by liquid chromatography-mass spectrometry from PBMCs protein extraction. The cells were collected from six cows per treatment during the 1st week postpartum. Quantification of differential abundance between groups was done using MS1 intensity based label-free. Label-free quantitative shotgun proteomics was used for characterization. This novel dataset of proteomics data from PBMC contains 3807 proteins; 44, 42 and 65 were differently abundant (P ≤ 0.05 and FC ± 1.5), in FLX vs. CTL, FO vs. CTL and FLX vs. FO, respectively; these findings are discussed in our recent research article [1]. The present dataset of PBMC proteome adds new information regarding the effects of n-3 FA on the immune system, while providing reference for PBMC proteome in postpartum dairy cows.

Here, we show that PERK-mediated suppression of miRNAs by sildenafil is vital to keep mitochondrial homeostasis, using cardiac-specific PERK knockout (cko) mice.

The effect of underfeeding on gene expression in the bovine tract during postpartum

Project description:This study aimed to investigate the effect of a mild underfeeding on global gene expression in the oviduct, the endometrium and the corpus luteum between 4 and 15 days post-ovulation in postpartum (PP) Holstein cows. Four control cows (C) received 100% of energy and protein requirements after calving when 4 underfed (U) received 80% of the control diet. Oestrus synchronization treatment was applied to induce ovulation on D80 PP. Oviduct, ovaries and the anterior part of each uterine horn were recovered chirurgically 4, 8, 12 and 15 days after ovulation. Gene expression was assessed on a dedicated 10K array and statistically analyzed on pools across the oestrus cycle days. Tissues appeared divergently "diet-responsive" as no differentially expressed gene (DEG) in the corpus luteum between U and C were highlighted, conversely to 293 DEGs in the oviduct vs. 1 in endometrium under a False discovery rate (FDR)<0.11, and 3830 DEGs vs. 28 respectively under a FDR<0.30. Pathways involved immunity, lipid catabolism and cell proliferation. However, once correlated with dedicated statistics (regularized canonical correlation analysis) to the evolution of 6 plasmatic markers measured weekly over 80 days, IGF-1, insulin, beta-Hydroxybutyrate and genes potentially implied in tissue repair mechanisms (e.g. FABP5/CRABP2, ELF1, TCEA1, NF-kB, TGF-beta1, HNF4A) became relevant to dairy cow physiology during PP in the oviduct and the endometrium. NP-18 vs. NP-18AR transcriptional profile upon treatment with TK/GCV

Project description:Pancreatic cancer-derived cells NP-18 underwent four rounds of treatment with increasing doses of an adenoviral vector encoding TK enzyme and GCV. Surviving cells were termed NP-18AR and displayed decreased sensitivity to treatment. This experiment analyses the transcriptomic effect of TK/GCV treatment on NP-18AR as compared to that of NP-18 cells. Two-condition experiment, where response to treatment in naïve cells (NP-18) was compared to response to treatment in previously treated resistant cells (NP-18AR). Biological replicates: samples from 3 independently generated and independently treated NP-18AR lines (NP-18AR1, NP-18AR2 and NP-18AR3) were hibridized with 3 independently treated samples of NP-18 cells. A total of 9 hybridizations were performed, 2 for NP-18AR1 (including 1 dye-swap), 4 for NP-18AR2 (including 2 dye swaps) and 3 for NP-18AR3 (including 1 dye-swap).
