Both fractions were sequenced.

Project description:We collected dorsal root ganglion (DRG), sciatic nerve (SN) and cochlea tissues from each E14, E18 and P14 plp-GFP+ mice. This allowed for a comparison of plp-GFP+ glia from lumbar (DRG+SN) and cochlea tissues within the same mice. The transcriptional composition of all samples was assessed using single-cell RNA-seq (scRNA-seq).

Single-Cell RNA-Seq Uncovers a Robust Transcriptional Response to Morphine by Glia.

Project description:Molecular and behavioral responses to opioids are thought to be primarily mediated by neurons, although there is accumulating evidence that other cell types play a prominent role in drug addiction. To investigate cell-type-specific opioid responses, we performed single-cell RNA sequencing (scRNA-seq) of the nucleus accumbens of mice following acute morphine treatment. Differential expression analysis uncovered unique morphine-dependent transcriptional responses by oligodendrocytes and astrocytes. We examined the expression of selected genes, including Cdkn1a and Sgk1, by FISH, confirming their induction by morphine in oligodendrocytes. Further analysis using RNA-seq of FACS-purified oligodendrocytes revealed a large cohort of morphine-regulated genes. The affected genes are enriched for roles in cellular pathways intimately linked to oligodendrocyte maturation and myelination, including the unfolded protein response. Altogether, our data illuminate the morphine-dependent transcriptional response by oligodendrocytes and offer mechanistic insights into myelination defects associated with opioid abuse.

Ensheathing glia promote healthy brain aging

Project description:Drosophila ensheathing glia decline in relative numbers during normal aging. Using p35, we inhibit apoptosis specifically in ensheathing glia, and show that this is sufficient to rescue the decline in ensheathing glia, improve neuromotor performance of aged flies, and increase lifespan. Remaining ensheathing glia from aged brains exhibit transcriptomes exhibiting signs of lipid metabolism and apoptosis dysregulation. Expanding ensheathing glia with p35 also prevented the accumulation of amyloid plaques and delayed premature death in a fly model of Alzheimer's disease.

Single cell RNA-seq of adult human neural retina

Project description:To comprehensively profile cell types in the human retina, we performed single cell RNA-sequencing on 20,009 cells obtained post-mortem from three donors and compiled a reference transcriptome atlas. Using unsupervised clustering analysis, we identified 18 transcriptionally distinct clusters representing all known retinal cells: rod photoreceptors, cone photoreceptors, Müller glia cells, bipolar cells, amacrine cells, retinal ganglion cells, horizontal cells, retinal astrocytes and microglia.
