Project description:In rainbow trout (Oncorhynchus mykiss), the effect of a paternal and a maternal high carbohydrate/low protein diet was assessed on progeny. To this purpose, two-year old males and females rainbow trout were fed either a control diet or a high carbohydrate/low protein diet for an entire reproductive cycle for females and for 5 months for males. Crossed-fertilizations were carried out in order to obtain 4 groups of offspring. Before the first feeding, whole fry transcriptomes were compared to detect any impact of the parental diet on offspring metabolism.

Project description:It is now recognized that parental diets could highly affect offspring metabolism and growth. Studies in fish are, however, lacking. In particular, the effect of a parental diet high in carbohydrate (HC) and low in protein (LP) on progeny has never been examined in higher trophic level teleost fish. Thus, two-year old male and female rainbow trout (Oncorhynchus mykiss) were fed either a control diet (0% carbohydrate and 63.89% protein) or a diet containing 35% carbohydrate and 42.96% protein (HC/LP) for a complete reproductive cycle for females and over a 5-month period for males. Cross-fertilizations were then carried out. To evaluate the effect of the parental diet on their offspring, different phenotypic and metabolic traits were recorded for offspring before their first feeding and again three weeks later. When considering the paternal and maternal HC/LP nutrition independently, fry phenotypes and transcriptomes were only slightly affected. The combination of the maternal and paternal HC/LP diets altered the energy metabolism and mitochondrial dynamics of their progeny, demonstrating the existence of a synergistic effect. The global DNA methylation of whole fry was also highly affected by the HC/LP parental diet, indicating that it could be one of the fundamental mechanisms responsible for the effects of nutritional programming.

Project description:Sex steroids play a key role in triggering sec differentiation in fish and the use of exogenous hormone treatment leads to partial or complete sex reversal. This phenomenon has attracted attention since the discovery taht even low environmental doses of exogenous steroids can adversely affect gonad morphology (ovotestis development) and induce reproductive failure. Modern genomic-based technologies have enhanced opportunities to find mechanisms of action (MOA) and dentify biomarkers for the toxic action of a compound. The goal of this study are to improve the understanding of feminization in fish by analyzing gene expression patterns in the gonads of rainbow trout fry after a chronic exposure to several doses (0.01, 0.1, 1 and 10 ?g/L) of ethynylestradiol (EE2) and to offer target genes as potential biomarkers of ovotestis development. An all-male population of Rainbow trout fry was exposed during 76 days (from 60 to 136 days post-fertilization (dpf)) to five nominal concentrations of 17?-ethynylestradiol (0-solvent control-, 0.01, 0.1, 1 and 10 ?g EE2/L of water), using 3 tanks per condition. In total, 30 samples were analyzed independantly: 6 samples per concentration tested (two samples per tank, three tanks per concentration), each sample being a pool of 10 pairs of gonads.

Project description:Newly hatched rainbow trout were exposed to the PAHs retene and fluoranthene semistatically for 1, 3, 7 and 14 days. Fry were exposed to the PAHS alone or as a binary mixture. At sample, hearts were excised, pooled and prepared for microarray analysis. We also investigated body burden of the two PAHs and how the whole organisms respond biometrically following exposure. Transcriptomic responses were confirmed using qPCR.

Project description:We investigate mechanisms behind the response to a plant based diet at short term, by analyzing the transcriptome of 3 isogenic lines of rainbow trout (R23, AB1, A22). These families represent an interesting genetic material to work on as they have similar responses when fed marine based-diet (M diet) but highly different responses when fed a plant-based diet (V diet). Transcriptomic analyses of whole fry revealed 1740, 1834 and 246 probes differentially expressed among the three isogenic lines. Gene ontology enrichment analysis revealed different enriched pathways. Among them, some were related to immunity and perception. Among ontologies related to metabolism only the methionine metabolism pathway was found to be significantly enriched. First, the use of these isogenic lines of rainbow trout led to find new pathways related to plant-based diets utilization which had not been described previously. Secondly, difference in expression of genes related to immunity suggest that this function play an important role but specific effect on immune status was difficult to assess, and further investigations are needed. Finally, results suggested that mechanisms underlying the use of plant-based diets is more linked to feed intake rather than to metabolism. Research on plants components affecting feed intake should be further explored to continue to improve feed formulation and reduce the reliance of aquaculture on marine resources without affecting fish performance.

Project description:The rainbow trout, Oncorhynchus mykiss, is relatively sensitive to hypoxia and does not survive deep hypoxia or anoxia. Given this lack of hypoxia-tolerance in the whole animal, the aim of this experiment was to investigate the in vitro responses of cultured rainbow trout cells following anoxia exposure. Rainbow trout hypodermal fibroblast (RTHDF) cells were exposed to anoxia for 12 and 24 h, whilst control cells were held under normoxic conditions for 24 h. Differential gene expression as a consequence of the treatments was analysed using an oligoarray composed of approximately 21,500 BLAST-identified sequences fabricated on the Agilent microarray platform. 57 genes were found to display significant responses to oxygen deprivation and these genes were assigned Gene Ontology terms and IDs. Of the 57 differentially expressed genes, 6 genes associated with carbohydrate metabolism were up-regulated following anoxia, these included phosphoglucomutase (PGM) and glycogen phosphorylase, consistent with glycogen serving as an important energy source during anoxia. Other genes up-regulated in response to anoxia included a number of putative targets of hypoxia inducible factor 1 alpha (HIF1A), namely phosphoglycerate kinase, triosephosphate isomerase and the glucose transporter, GLUT2. Egl9 homolog 3, the proline hydroxylase responsible for HIF1A regulation, was also induced. This analysis indicates that cultured trout cells have the capacity for adaptive gene responses when subjected to oxygen levels that are lethal to the whole organism.
