Project description:A cDNA microarray composed of 9065 uniEST probes was constructed and applied to detect the gene expression profile of protoscoleces of E. granulosus treated with albendazole and artemisinin in vitro, respectively. Compared with the untreated parasite, 7 genes were up-regulated and 38 genes were down-regulated in protoscoleces treated with albendazole and 100 genes were up-regulated and 6 genes were down-regulated in protoscoleces treated with artemisinin. The differential expression genes in protoscoleces induced by albendazole were clustered into energy metabolism, cell cycle and assembly of cell structure. And the differential expression genes affected by artemisinin were clustered into genetic information, the transduction of environmental signals, metabolism. Under transmission electron microscope observation, albendazole intervention damaged the cell structure, and form the heterochromatin in protoscoleces cells was mainly increased in the artemisinin group. Isolated E. granulosus protoscoleces were treated with albendazole and artemisinin respectively and was extracted of total RNA using the TRIZOL Reagent according to the manufacturer's instructions. Gene-expression profiling was performed for each RNA sample separately on the GeneChip of Echinococcus granulosus Genome Array (Capitalbio) at CapitalBio Corporation (Beijing, China).

Project description:TGF-β is a crucial cytokine participate in the interplay between the intermediate host and helminthes. TGF-β receptors were discovered in many cestode, and could bind the human TGF-β. However, the function of host TGF-β on the Echinococcus is still not elucidated, and this paper aim to explore the question at transcription level. Microarray analysis was used to investigate differential expression genes in protoscolices of Echinococcus granulosus cultured in the presence or absence of human TGF-β at different time points (4h, 8h and 24h) in vitro. A total of 523 genes were up- or down-regulated in response to TGF-β, compared with control group, 390 genes were up-regulated and 47 genes were down-regulated at 8h, and 376 genes were up-regulated and 19 genes were down-regulated at 12h, including 310 differential genes were regulated at both time point. Only 1 gene down-regulated at 4 h. Gene ontology (GO) analysis showed that the biological process of the up-regulated genes in protoscolex were predominantly involved in DNA packaging, nucleosome assembly, chromatin assembly, etc. And the cellular component gene were located in cell nucleus. TGF-β appeared to promote growth or development of the protoscolex by up-regulated the gene related with mitosis. In addition, the study also indicated that TGF-β has a multiple influence on the protoscolex, as reflected in the increased stimulation of gene expression of the ErbB signaling pathway, MAPK signaling pathway, Notch signaling pathway and VEGF signaling pathway. Project description:Echinococcosis is a disease resulting from infection by flatworms of the Echinococcus genus. The vast majority of cystic echinococcosis cases in Southern Brazil are caused by Echinococcus granulosus and Echinococcus ortleppi. Here, we surveyed the protein contents of the hydatid fluid compartment of E. granulosus and E. ortleppi pulmonary bovine cysts, in an attempt to compare their molecular arsenal in this host-parasite interface. We detected some molecular players equally in E. granulosus and E. ortleppi, such as proteolytic enzymes and development signaling molecules, while some activities were more evident in one species, such as apoptosis in E. ortleppi, and cysteine protease activity in E. granulosus.

Project description:Parasitic infection improves metabolic homeostasis in “western diet” induced-obesity, which partially results from the regulation of adipogenesis. However, the underlying mechanism remains largely unknown. This study investigated the long non-coding RNA（lncRNA）and mRNA profiles of subcutaneous adipose tissues in mice infected by the protoscoleces of Echinococcus granulosus (EgPSC) using a microarray analysis. In comparison with control mice, the infected mice showed decreased mass of adipose tissues and shrunken size of adipocytes. The indirect calorimetry found the alternation of energy metabolism post infection, which was characterized by less CO2 production and O2 consumption, sharp decline of carbohydrate oxidation and a little increase of fat oxidation. A total of 1,052 mRNAs and 220 lncRNAs were differentially expressed in infected subcutaneous adipose tissues (fold change ≥2, P＜0.05). Of them, 541 mRNAs were upregulated and 511 mRNAs were downregulated; 126 lncRNAs were upregulated and 94 lncRNAs were downregulated. The Gene Ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis showed that the parasitic infection reprograms a complex metabolic network. Notably, “lipoxygenase” and “Arginine and proline metabolism” pathways were significantly upregulated while “glycolysis”, “Tricarboxylic acid (TCA) cycle”, “denovo lipogenesis” and “lipid droplet” pathways were dramatically downregulated. In addition, several key lncRNAs were linked with insulin resistance and adipocyte differentiation. Overall, this study suggests that E. granulosus infection inhibits the adipogenesis, which provides a novel insight for metabolic homeostasis mediated by parasitic infection, and hypertrophic adipocytes in obesity.

Project description:This study investigates the impact of hydatid antigens on the miRNA expression profiles within extracellular vesicles (EVs) derived from mesenchymal stem cells (MSCs). By stimulating MSCs with echinococcus granulosus protoscoleces (ESPs), hydatid cyst fluid (HCF), and particles from the laminated layer (pLL), we aim to uncover the changes in miRNA expression and their potential roles in modulating immune responses and osteogenic differentiation. Through high-throughput sequencing, differential expression analysis, and subsequent bioinformatics analyses, we identify key miRNAs and their target genes involved in these processes. Our findings provide insights into the complex interplay between parasitic infections and host cell responses, highlighting the therapeutic potential of MSC-derived EVs in treating hydatid disease.

Project description:microRNAs (miRNAs), a class of small non-coding RNAs, are key regulators of gene expression at post-transcriptional level and play essential roles in fundamental biological processes such as development and metabolism. Here, we perform a comprehensive analysis of miRNAs in the zoonotic parasite E. canadensis G7, one of the causative agents of the neglected disease cystic echinococcosis. Small RNA libraries from protoscoleces and cyst walls of E. canadensis G7 and protoscoleces of E. granulosus sensu stricto G1 were sequenced using Illumina technology. As a result, we found transcriptional evidence of 37 miRNAs thus expanding the miRNA repertoire of E. canadensis G7. Differential expression analysis showed significant regulated miRNAs between life cycle stages of E. canadensis G7. We confirmed the remarkable loss of conserved miRNA families in E. canadensis, reflecting their low morphological complexity and high adaptation to parasitism. This study will provide valuable information for better understanding the complex biology of this parasite and could help to find new potential targets for therapy and/or diagnosis. Small RNA libraries from protoscoleces and cyst walls of E. canadensis G7 and protoscoleces of E. granulosus sensu stricto G1 were sequenced using Illumina technology. For each sample type, two libraries were constructed from two independent samples in order to have biological replicates.

Project description:Cystic echinococcosis (CE) is a common zoonotic parasitic disease that seriously impacts public health. However, the full spectrum of immune cell changes in Echinococcus granulosus (E. granulosus) infection, especially the negative immune regulation of subpopulations of regulatory T cells (Treg), are not yet well understood. In this study, we aimed to identify the panoramic landscape of changes in immune cells and molecules and the distinct alterations in Treg subpopulations in mice infected with E. granulosus. We used single-cell RNA sequencing and single-cell T/B cell receptor sequencing to analyze 53,298 cells from the spleen and peripheral blood mononuclear cells (PBMCs) of healthy and E. granulosus-infected mice. We used immunofluorescence combined with RNA fluorescence in situ hybridization and quantitative real-time polymerase chain reaction to verify the sequencing results. Our results showed organ-specific immune system alterations in mice infected with E. granulosus. E. granulosus-infected mice induced a subpopulation of CD4+ cells with type I interferon production potential. Furthermore, there were seven different Treg cell subpopulations in vivo at three stages of differentiation, and Treg subpopulations of different classes and different stages of differentiation showed tissue specificity. After infection, the Lag3hi Treg and Gpr83+Igfbp4+ naïve Treg subpopulations were specifically induced in PBMCs and spleen, respectively. Furthermore, T follicular helper 2(Tfh2) cells with high expression of Cxxc5 and Spock2 were found in E. granulosus-infected mice. Additionally, combined single-cell immunome repertoire sequencing analysis revealed that gene fragment expression of multiple VJ-paired combinations of T cell receptor β chains were downregulated after infection. Our data uncovered changes in the full spectrum of immune cells in mice following E. granulosus infection, including subpopulations of cells that have not been emphasized in previous studies. These results further enrich the study of the bidirectional immunomodulatory mechanism and offer a different perspective for subsequent studies of E. granulosus infection.
