Project description:Background: Dendrobium officinale, an endangered Chinese herb, has extensive therapeutic effects and contains bioactive ingredients including a large number of polysaccharides and alkaloids, and minimal flavonoids. Firstly, this study attempts to obtain the protocorm-like bodies of this plant through tissue culture to produce the main secondary metabolites whose distribution in each organelle and protocorm like bodies is analyzed. Then, analysis of the correlation between comparative transcriptome sequence and the metabolite content in different organs enables the discovery of putative genes encoding enzymes involved in the biosynthesis of polysaccharides and alkaloids, and flavonoids. Results: The optimum condition for protocorm-like bodies (PLBs) induction and propagation of D. officinale is established. For protocorm induction, we use the seed as the explant, and the optimum medium formula for PLBs propagation is 1/2 MS + α-NAA 0.5 mg·L-1 +6-BA 1.0 mg·L-1 + 2, 4-D 1.5-2.0 mg·L-1 + potato juice 100 g·L-1. The distribution of polysaccharides, alkaloids and flavonoids in D. officinale organs was clarified. Stems, PLBs and leaves have the highest content of polysaccharides, alkaloids and flavonoids, respectively. PLBs replace organs to produce alkaloids in D. officinale, and naringenin was only produced in stem. Hot water extraction (HWE) method was found outperforming the ultrasound-assisted extraction (UAE) method for polysaccharides from D. officinale. A comparative transcriptome analysis of the protocorm-like bodies and leaves of D. officinale showed genes encoding enzymes involved in polysaccharides, alkaloids and flavonoids biosynthetic pathway were differentially expressed. Putative genes encoding enzymes involved in polysaccharides, alkaloids and flavonoids synthetic pathway were identified. Notably, genes encoding enzymes of strictosidine beta-glucosidase, geissoschizine synthase and vinorine synthase in alkaloids biosynthesis of D. officinale are first reported. Conclusions: Our works, especially the identification of candidate genes encoding enzymes involved in metabolites biosynthesis will help to explore and protect the endangered genetic resources and will also facilitate further analysis of the molecular mechanism of secondary metabolites’ biosynthesis in D. officinale.

Project description:Licorice (Glycyrrhiza uralensis Fisch) flavonoids have many pharmacological effects, as the main chemical component of licorice, its content directly affects the quality of licorice. Methyl jasmine (MeJA) is an important signaling molecule in the secondary metabolic pathway of plants, but the biological mechanisms that stimulating the production of licorice flavonoids and the related changes in transcriptome are still less studied. In this research, the expression of two key enzyme genes: Chalcone synthase (CHS) and Cinnamate 4-hydroxylase (C4H) in the biosynthesis pathway of licorice flavonoids was determined, and it was significantly different after 9 hours of MeJA induction. The transcriptome profiles of licorice cells at 9 hours after MeJA treatment were analyzed to investigate the transcriptional alterations of licorice cell in response to MeJA elicitation by “RNA-seq”. 151, 529 transcripts (200 bp in length) of cDNA from the samples were generated, and 116, 907 unigenes were found. MeJA appeared to stimulate a large number of genes involved in several relevant functional categories, such as carbohydrate metabolism and encoding transcription factors, 11 MYB transcription factors expressed significant differences were screened. This comprehensive description of gene expression information could help elucidate the molecular mechanism of MeJA-mediated biosynthesis of licorice flavonoids and MeJA-regulated network formation.

Project description:Erythrina velutina is widely used in traditional medicine, being a source of specialized metabolites with anti-inflammatory and central nervous system modulatory activities. To date, the specialized metabolic network of the genus Erythrina and its regulation are missing, in part due to the lack of genomic information. In an effort to fill this knowledge gap, leaves and seeds had their metabolic and proteomic profiles (LC/MS) analyzed and integrated with transcriptome (RNA-seq) data. These data sets were obtained from plants growing in their harsh natural habitat and examined with emphasis on differentially expressed transcripts, transcription factors, and genes putatively involved in the biosynthesis of flavonoids, the most abundant metabolite class annotated. Of the 96 metabolites distributed in different classes found in this study, 28 and 30 were exclusively found in seeds and leaves, respectively, whereas 38 metabolites occurred in both structures. It was possible to assemble a set of 17,822 transcripts, of which 3,937 transcripts were differentially expressed (2,815 down- and 1,122 up-regulated). Candidate transcripts involved in flavonoid accumulation and transcription factors amounted to 28 and 394, respectively. Expression patterns of transcripts involved in flavonoid biosynthesis and its regulation, as well as those involved in responses to biotic and abiotic stresses, are discussed in detail. Hopefully, these findings will provide a solid base to further the understanding of the molecular and metabolic responses in this important medicinal plant species, as well as identify key regulatory targets for future studies aiming at bioactive metabolite production.

Project description:We cloned and characterized a subgroup S4 R2R3 Myb transcription factor, PgMyb308-like. Overexpressing PgMyb308-like in pomegranate hairy roots. Transcriptome analysis of PgMyb308-like-overexpressing hairy roots revealed reprogramming of cell wall-related genes, while overexpression of PgMyb308-like in Arabidopsis thaliana plants uncovered its distinct role in a different genetic and metabolic background. The results suggest that PgMyb308-like activates genes in the shikimate pathway and lignin biosynthesis, but suppresses those involved in the production of HTs and flavonoids.

Project description:Growth inhibition of the pathogen Staphylococcus aureus with currently available antibiotics is problematic in part due to bacterial biofilm protection. While Although the recently characterized natural products, including 3´,4´,5-trihydroxy-6,7-dimethoxy-flavone [1], 3´,4´,5,6,7-pentahydroxy-flavone [2] and 5-hydroxy-4´,7-dimethoxy-flavone [3] exhibit both antibiotic and biofilm inhibitory activities, the mode of action of such hydroxylated flavonoids with respect to S. aureus inhibition has is yet to be characterized. Enzymatic digestion and high- resolution MS mass spectral analysis of differentially expressed proteins from S. aureus, with and without exposure to antibiotic flavonoids (1-3) has allowed for the characterization of global -protein alterations induced by metabolite treatment. A total of 56, 92, and 110 proteins are were differentially expressed with bacterial exposure to 1, 2 or 3, respectively. Connectivity of identified proteins is characterized using a search tool for the retrieval of interacting genes/proteins (STRING) with multi-targeted S. aureus inhibition of energy metabolism and biosynthesis by the assayed flavonoids observed. Identifying the mode of action of natural products as antibacterial agents is expected to provide insight into the potential use of flavonoids alone or in combination with known therapeutic agents to effectively control S. aureus infection.

Project description:Rubus chingii Hu, indigenous to China, is very rich in flavonoids. With the exception of anthocyanin, fruit flavonoids were much higher than most berries including other raspberry species, which partly contributed to its high phenolics and antioxidant capability. In contrast to other raspberries, anthocyanin and other flavonoids decreased as fruit matured. We investigated four typical phases of fruit maturation using transcriptomics, proteomics and metabolomics. The results indicate that the down-regulation of phenylpropanoid, flavonoid and anthocyanin biosynthesis are responsible for the metabolic decrease. The down-regulation of RcCHS (CL6140.Contig5) and RcCHI (Unigene14858 and Unigene22344) at gene and protein levels were associated with decreases of naringenin/naringenin chalcone respectively. Down-regulation of RcLAR (CL9527.Contig3) at gene and protein levels was consistent with decreases of afzelechin/epiafzelechin, catechin/epicatechin, and gallocatechin/epigallocatechin. However, multiple genes/proteins involved in the enzymatic pathways were divergent and differently regulated, e.g. Rc4CL genes/proteins were down-regulated while Rc4CL-like was maintained at constantly low levels.

Fagopyrum tataricum (Tartary buckwheat) itself contains more flavonoids, so exploring the regulation of Tartary buckwheat flavonoid synthesis can provide a solid theoretical basis for the cultivation of new Tartary buckwheat varieties with high nutrition. In this paper, we studied the role of FtMYBs in regulating the biosynthesis of flavonoids in Tartary buckwheat.
