Project description:Graft compatibility is the ability of two plants to form cohesive vascular connections. Tomato and pepper grafts are incompatible but the underlying causes of this phenomenon remain unknown. We utilzied a broad array of techniques to profile graft compatibility including viability, biophysical stability, and growth. Cell death in the junction was quantified using trypan blue and TUNNEL assays. Transcriptomic analysis of cell death in the junction was preformed using RNA-sequncing. Finally a meta-transcriptomic analysis was conducted with published datasets to further explore the genetic signature of graft incompatibility.We found that all varieties of pepper tested across two species were incompatible with tomato. Tomato and pepper graft incompatibility is characterized by stem instability, reduced growth, and persistent cell death in the graft junction. We showed that tomato and pepper heterografts have prolonger transcriptional activity, with defense processes highly enrched. We identfied a large subset of NLRs and genes involved in programmed cell death which were upregulated in incompatible tissue. We also identified a set of genes with orthologs in both tomato and pepper which are upregulated in incompatible grafts including biosythesis of steroidal glycoalkaloids. Finally we utilized various biological stressors to explore the genetic signature of grafting. We found a significant overlap in the genetic profile of grafting and plant parsitism. We also identified over 1000 genes uniquely upregulated in incompatible grafting including genes in involved in DNA-damage repair. Based on the broad upregulation of NLRs and genes involved with programmed cell death, prolonged cell death in the junction, and DNA damage, we have determined that tomato and pepper graft incompatibility is likely caused by a form of genetic incompatibility which triggers an autoimmune-like response.

Project description:Phytophthora blight is a highly destructive soil borne disease caused by Phytophthora capsici Leonian, which seriously threatens global pepper production. Grafting is one of the important means to improve plant disease resistance and prevent soil borne diseases in vegetable production. However, the molecular mechanism by which grafting enhances the resistance of pepper to Phytophthora blight is still unclear. This study used Phytophthora capsici resistant strain ‘ZCM334’ and susceptible strain ‘Early Calwonder’ as rootstocks, and ‘Early Calwonder’ as scions for grafting. Phenotypic observation and cytological analysis showed that compared with the ‘Early Calwonder’ self rooted plants, the ‘ZCM334’ grafted plants had a later onset of disease, stronger resistance, and less damage to leaf cells, indicating that grafting can significantly improve the resistance of pepper to Phytophthora capsici. This study identified differentially expressed proteins (DEPs) in the leaves and roots of ‘ZCM334’ grafted plants and ‘Early Calwonder’ self rooted plants through proteomic analysis based on iTRAQ technology, and 478 and 349 DEPs were identified between their leaves and roots, respectively. These DEPs were mainly involved in metabolic process, cellular process, response to stimulus and catalytic activity processes. We identified and screened 12 DEPs with consistent expression trends in the leaves and roots of ‘ZCM334’ grafted plants and ‘Early Calwonder’ self rooted plants, including seven DEPs related to Phytophthora capsici resistance (CA01g31060, CA02g15780, CA02g30850, CA01g11410, CA05g12260, CA03g35150, and CA03g36980) and five proteins with unknown functions (CA01g26190, CA11g10620, CA12g02730, CA01g20890 and CA02g11340). Through qRT-PCR analysis, a significant correlation was observed between the protein and transcript levels of these 12 DEPs, and their expression characteristics were analyzed in the roots and leaves of ‘ZCM334’ grafted plants and ‘Early Calwonder’ self rooted plants at different stages (0h, 12h, 24h, and 36h) after inoculation with Phytophthora capsici. This study provides valuable information for exploring the molecular mechanisms by which grafting enhances the resistance of pepper to Phytophthora blight and the excavation of these key genes provides research ideas for studying the regulatory network of pepper resistance to Phytophthora capsici.

Project description:Purpose: To investigate the influence of grafting on adipose tissues, samples were collected from the inguinal fat tissues of mice before grafting and 1 week after grafting. Methods: The RNA-seq analysis was performed by Novogene (Beijing, China). Total RNA was drawn from the tissues with TRIzol reagent (Absin, Shanghai, China). Transcriptome sequencing were performed by using the Illumina HiSeq X Ten (Novogene Bioinformatics Technology Co., Ltd., Beijing, China). Mapping of 150-bp paired-end reads to the genes was undertaken. Result: In fat tissues one week after grafting, 28145 genes were detected. And 3271 genes were found to be upregulated, while 3745 genes were downregulated (padj < 0.05 and log2(FC) > 1), when compared with that in the natural fat tissues. Conclusions: Our study firstly reported the differentially expressed genes in adipose tissues after grafting, providind a valuable reference for fat grafting studies.
