Project description:In this study, the mitochondrial genomes (mitogenomes) of two walnut leaf insect pests, Gastrolina depressa depressa and G. depressa thoracica, were sequenced by Sanger sequencing technology. The mitogenome of G. depressa thoracica was complete at 16,109 bp in length, while the mitogenome of G. depressa depressa (14,277 bp) was partial. The genomic analyses indicated that both mitogenomes have the typical gene content and arrangement. The formerly identified elements, 'TAGTA' between trnSer(UCN) and nad2, and 'ATGATAA' between atp8 and atp6, were more conserved than that between nad4L and nad4, which was 'ATGTTAA' in Coleoptera excluding Polyphaga. Phylogenetic analyses of the 13 protein-coding genes from 36 coleopteran species well supported a close affinity between the subfamily Chrysomelinae including G. depressa thoracica and G. depressa depressa and Galerucinae, as well as a sister relationship of ((Eumolpinae + Cryptocephalinae) + Cassidinae) within Chrysomelidae.

Project description:Investigation of the South African plant Urginea depressa Baker (Asparagaceae Juss.) for antiproliferative activity against the A2780 ovarian cancer cell line led to the isolation of the six new homoisoflavonoids urgineanins A-F (1-6), the two known bufatrienolides 7 and 9, and the new bufatrienolides urginins B and C (8 and 10). Structures were elucidated based on analysis of their 1D and 2D NMR spectra, electronic circular dichroism, and mass spectrometric data. Five of the six new homoisoflavonoids had good antiproliferative activity against the A2780 ovarian cancer, A2058 melanoma, and H522-T1 human non-small-cell lung cancer cells, and urgineanin A (1) had submicromolar activity against all three cell lines. The four bufatrienolides 7-10 had strong antiproliferative activity against the same cell line, with IC50 values of 24.1, 11.2, 111, and 40.6 nM, respectively.

Project description:The use of micro-computed tomography (?CT) provides a unique opportunity to look inside the shells of larger benthic foraminifera to investigate their structure by measuring linear and volumetric parameters. For this study, gamonts/schizonts and agamonts of the species Heterostegina depressa d'Orbigny were examined by ?CT; each single chamber's volume was digitally measured. This approach enables cell growth to be recognised in terms of chamber volume sequence, which progressively increases until reproduction occurs. This sequence represents the ontogeny of the foraminiferal cell and has been used here to investigate controlling factors potentially affecting the process of chamber formation. This is manifested as instantaneous or periodic deviations of the realised chamber volumes derived from modelled growth functions. The results obtained on naturally grown specimens show oscillations in chamber volumes which can be modelled by sums of sinusoidal functions. A set of functions with similar periods in all investigated specimens points to lunar and tidal cycles. To determine whether such cyclic signals are genuine and not the effects of a theoretical model, the same analysis was conducted on specimens held in a closed laboratory facility, as they should not be affected by natural environmental effects. Surprisingly, similar cyclicities were observed in such samples. However, a solely genetic origin of these cycles couldn't be verified either. Therefore, detailed analysis on the phase equality of these growth oscillations have been done. This approach is pivotal for proving that the oscillatory patterns discovered in LBF are indeed genuine signals, and on how chamber growth might be influenced by tidal currents or lunar months.

Project description:In this study, 19 octadecanoid derivatives-four pairs of enantiomers (1⁻8), two racemic/scalemic mixtures (9⁻10), and nine biosynthetically related analogues-were obtained from the ethanolic extract of a Chinese medicinal plant, Plantago depressa Willd. Their structures were elucidated on the basis of detailed spectroscopic analyses, with the absolute configurations of the new compounds assigned by time-dependent density functional theory (TD-DFT)-based electronic circular dichroism (ECD) calculations. Six of them (1, 3⁻6, and 9) were reported for the first time, while 2, 7, and 8 have been previously described as derivatives and are currently obtained as natural products. Our bioassays have established that selective compounds show in vitro anti-inflammatory activity by inhibiting lipopolysaccharide-induced nitric oxide (NO) production in mouse macrophage RAW 264.7 cells.
