Project description:We overexpressed the brassinolide synthase DET2 in 84K poplar and obtained an overexpression material.It was found that the brassinolide content of the overexpression material increased significantly.And it was found that the plant height, diameter, and number of internodes of the overexpression material increased significantly.However,the mechanism of this phenotype is unknown.We collected two-month-old DET2 overexpression material and 84K wild-type material in three parts, apex,cambium,and xylem,and conducted high-throughput sequencing.The sequencing results were mined, and the difference genes that were significantly changed in the DET2 overexpression material and the 84K wild-type material were found.Find out how the increase in brassinolide content promotes the development of wood.

Project description:We investigated the transcriptional response of hybrid poplar (Populus trichocarpa x deltoides) source and sink leaves to simulated herbivory (mechanical wounding plus the application of Malacosoma disstria oral secretions) over a 24 hour time course. Experiments were conducted using clonal trees under greenhouse conditions at the University of British Columbia. We used the 15.5K poplar cDNA microarray platform previously described by Ralph et al. (Molecular Ecology 2006, 15:1275-1297). Differentially expressed genes were determined using three criteria: fold-change between treated and untreated control leaves > 1.5-fold, P value < 0.05 and Q value < 0.05. This study identified > 3,000 differentially expressed genes in response to simulated insect feeding damage, which possess distinct source/sink and treated/systemic patterns.

To reveal identify the enzymes that contributeing to lignocellulose degraredauction and its degradation mechanisms, we evaluated transcriptomic how study theof T. versicolor transcriptome was changes during evaluated growthing on the poplar wood relative to growth on glucose medium. 853 genes were differentially expressed;, 360 genes were up-regulated on poplar wood, and 493 genes were down-regulated on poplar wood. Notably, most genes relative involved into lignin degradation were up-regulated, including eight lignin peroxidase (LiP) genes, and two manganese peroxidase (MnP) genes etc. Genes encoding cellulose and hemicelluloses degrading-enzymesation were mostly down-regulated, including six endo-β-1, 4-glucanase genes, three cellobiohydrolase I genes, and one cellobiohydrolase II gene, etc. MeanwhileAdditionally, expression of more significant expansion of P450s in T. versicolor genome, along with differences in carbohydrate- and lignin-degrading enzymes, could bewere correlated withto poplar wood degradation.
