Project description:Our previous study found that acupuncture with low frequency electrical stimulation (Acu/LFES) prevents muscle atrophy by attenuation of protein degradation in mice. The current study examines the impact of Acu/LFES on protein synthesis. C57/BL6 mice received Acu/LFES treatment on hindlimb for 30 minutes once. Acupuncture points were selected by WHO Standard Acupuncture Nomenclature and electric stimulation applied using an SDZ-II Electronic acupuncture instrument. Muscle protein synthesis was measured by the surface-sensing of translation (SUnSET) assay. Exosomes were isolated using serial centrifugation and concentration and size of the collected exosomes were measured using a NanoSight instrument. The mature microRNA library in serum exosomes was validated using a High Sensitivity DNA chip. Protein synthesis was enhanced in the both hindlimb and forelimb muscles. Blocking exosome secretion with GW4869 decreased the Acu/LFES-induced increases in protein synthesis. MicroRNA-deep sequencing demonstrated that four members of the Let-7 miRNA family were significantly decreased in serum exosomes. Real time qPCR further verified Acu/LFES-mediated decreases of let-7c-5p in serum exosomes and skeletal muscles. In cultured C2C12 myotubes, inhibition of let-7c not only increased protein synthesis, but also enhanced protein abundance of Igf1, Igf1 receptor. Using a luciferase reporter assay, we demonstrated that let-7 directly inhibits Igf1. Conclusion: Acu/LFES on hindlimb decreases let-7-5p leading to upregulation of the Igf1 signaling and increasing protein synthesis in skeletal muscles. This provides a new understanding of how the electrical acupuncture treatment can positively influence muscle health.

Project description:Interventions: Transcutaneous electrical acupoint stimulation group :Transcutaneous electrical stimulation was given at the bilateral Hegu, Neiguan, Feet Three Miles, Upper Juxu, Lower Juxu, and Sanyinjiao points 1 day before, during, and 1, 2, and 3 days after surgery, respectively ;False stimulus group :The electrode pads were selected to be attached at the same acupuncture points as in group A, but not energized, i.e., no current stimulation was given to the patients.

Project description:Acupuncture stimulations at GB34 and LR3 inhibit the reduction of tyrosine hydroxylase in the nigrostriatal dopaminergic neurons in the parkinsonism animal models. Especially, behavioral tests showed that acupuncture stimulations improved the motor dysfunction in a previous study. The thalamus is a crucial area for the motor circuit and has been identified as one of the most markedly damaged areas in Parkinson’s disease (PD), so acupuncture stimulations might also have an effect on the thalamic damage. We investigated gene expression profile changes in the thalamic region of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced parkinsonism models after acupuncture at the acupoints GB34 and LR3 C57BL/6 mice were divided into four experimental groups; ① C: Control, ② M: MPTP-treatment only, ③ MA: MPTP- and acupuncture-treatment at acupoints GB34 and LR3, ④ MNA: MPTP- and acupuncture-treatment at non-acupoints. Project description:Introduction: In our previous study, we demonstrated that acupuncture improved gait disturbances, such as hypometric gait, in patients with Parkinson’s disease (PD). To investigate specific genes that indicate a therapeutic response to acupuncture, we analyzed transcriptome alterations following acupuncture using blood samples collected in our previous clinical trial. Methods: We reanalyzed the clinical data of patients and selected a representative patient with PD for transcriptomic analysis following acupuncture. We examined the expression changes of PD biomarker genes known to be consistently dysregulated in both the brain and blood in patients with PD. We validated these gene expression changes using quantitative real-time polymerase chain reaction (qPCR) in the blood of five other patients with PD who received acupuncture treatment. Results: After acupuncture treatment, the transcriptomic alterations in the representative patient were similar to those induced by dopaminergic therapy. Among PD biomarkers, ankyrin repeat domain 22 (ANKRD22), which is upregulated after dopaminergic therapy, and synapsin 1 (SYN1), which is the common gene to indicate synaptic dysfunction in PD, were upregulated following acupuncture. These alterations correlated with changes of gait parameters in patients with PD.alterations following acupuncture using blood samples collected in our previous clinical trial. Conclusion: This is the first report on gene expression changes and improvement of gait disturbance in patients with PD following acupuncture.
