Project description:We perfomed a microRNA microarray on to assess differentially expressed miRNAs in bleomycin-induced lung fibrosis in mice. To induce pulmonary fibrosis, belomycine (Sigma-Aldrich, St Louis, MO) was dissolved and administred intratracheally at a dose of 1.5U/kg body weight. Control animals received saline only. 21 days post bleomycin treatment, mice were sacrificed and lung tissue were collected for RNA extraction and microRNA microarray.

Project description:For the subcutaneous model, 5x105 KPC-Luc cells were injected subcutaneously near the right flank of female C57BL/6 mice. 6-7 days after tumor implantation mice were randomized into 4 treatment groups and treatedwith VIP-R antagonist and/or anti-PD-1. While scram+IgG control mice received scrambled peptide and isotype IgG, the VIP-R antagonist, anti-PD-1 and VIP-R antagonist and anti-PD-1 groups received VIP-R antagonist and IgG; scrambled peptide and anti-PD-1; and VIP-R antagonist and anti-PD-1, respectively. The treatment regimen involved administering 10μg of scrambled or VIP-R antagonist: ANT008, subcutaneously every day and 200μg of IgG or anti-PD-1 intraperitoneally once every three days, for a total of 10 days.

Project description:Simple and efficient delivery of CRISPR genome editing systems in primary cells remains a major challenge. Here, we describe an engineered Peptide-Assisted Genome Editing (PAGE) CRISPR-Cas system for rapid and robust editing of primary cells. PAGE couples a cell-penetrating Cas protein with a cell-penetrating endosomal escape peptide in a 30-minute incubation that yields up to ~98% editing efficiency in primary human and mouse T cells. PAGE provides a broadly generalizable platform for next generation genome engineering in primary cells. Project description:Chromogranin A (CHGA) is elevated in plasma of patients with renal failure and several CHGA polymorphisms are associated with hypertensive renal disease. Therefore, we investigated the role of CHGA in kidney disease using the partial nephrectomy model of kidney failure. Mice under surgery to remove 2/3rds of the left kidney and the entire right kidney. Control mice received the sham surgery. Wild-type and CHGA-/- mice used. To model the changes in vitro, CRL-1927 kidney cells were treated with or without CHGA in vitro.

Project description:After an electrically induced seizure, mice (NMRI) received electrical stimulation (current intensity 44 mA, 0.2 ms monopolar pulses at 6 Hz frequency for 3 s). RNA was extracted from the hippocampi at 0, 3, 6, 24 and 72 h after the seizure. microRNA expressions were measured via microarray technology using Exiqon's miCURY™ LNA Arrays.

Project description:The aim of this study was to analyse alterations in circulating microRNA expression during breast tumour progression in a murine model, and to apply significantly altered miRNAs to patient samples to identify novel circulating microRNA markers or tumour progression. Athymic nude mice (n=10) received an injection of 2 x 105 MDA-MB-231 cells. Tumour volume was monitored weekly and blood sampling performed at weeks 1 and 6 following tumour induction. A microRNA array was performed comparing circulating 384 microRNAs in animals with early (week 1, n=5) versus late (week 6, n=5) disease.

Project description:CD44 wild-type and knockout C57 BL/6 mice were immunized by subcutaneous injection of MOG35-55 peptide. CD4 T cells were isolated from spleens of the mice on day 15 of the MOG35-55 peptide immunization. Peripheral blood lymphocytes (PBLs) were prepared from multiple sclerosis patients and normal individuals. Total RNA was extracted and subjected to microRNA high-throughput array with Affymetrix platform.
