Project description:Purpose: Betaine-Homocysteine Methyltransferase (BHMT) catalyzes the conversion of betaine to dimethylglycine and homocysteine to methionine. We were interested in both overexpressing and knocking out BHMT and examining the global effects of RNA transcription. We did this in both in vivo and in vitro models. Methods: Tumors: 1,000,000 HepG2 BHMT OE or BHMT KO cells were injected to the left and right flank of nude mice (n = 5), respectively, as a xenograft. Once tumors reached the appropriate size, they were harvested. The tissue was homogenized and RNA was extracted. Cells: HepG2 BHMT OE and KO cells (n = 3/group) were grown in Human Plasma-Like Medium (HPLM) for 48 hours. Then, cells were harvested and RNA was extracted. Both: Samples were submitted to the University of Utah sequencing core, who performed library prep and sequencing. Total RNA was taken from samples and run on an illumina sequencer. Note: OE2 and KO2 were large outliers, and therefore remove from the data analysis. They came from the same mouse.

Project description:Gene expression in satellite cell-derived primary myoblasts islolated from Gata4-loxP mice. Myoblasts were infected with nLacZ for control (Wt), Cre recombinase to knockout GATA4 (KO), or GATA4 expression vector to overexpress GATA4 (OE). Infected myoblasts were cultured in growth medium (day 0) then differentiated into myotubes in differentiation medium for 3 days (day 3). Total 18 samples. Three replicates in each myoblast; GATA4-Wt, -KO, and -OE myoblasts at day 0 and day 3.

Project description:To investigate the altered glucose and lipid metabolism genes after STIM1 knockout (STIM1 KO) or STIM1 knockout plus Snail1 OE (STIM1 KO+Snail1 OE) in SMMC7721 cells. STIM1 KO-SMMC7721 cells were constructed by CRISPR/Cas9, and STIM1 KO+Snail1 OE-SMMC7721 cells was established via lentiviral Infection of Snail1 overexpression in STIM1 KO-SMMC7721 cells. The cells mentioned above were cultured in DMEM supplemented with 10% (v/v) FBS at 37 °C in 5% CO2. When the cells grow to 60~70% confluent, total RNA was extracted by using Trizol (TAKARA, Japan). Reverse transcription was performed from 1μg total RNA using PrimeScript™ RT reagent Kit with gDNA Eraser (TAKARA), Quantitative real-time PCR were performed (CFX386, BioRad) with 40 cycles at 95 °C for 10 seconds, 59 °C for 20 seconds and 72 °C for 30 seconds.
