Project description:This project focused on the identification of membrane proteins involved in salinity tolerance in the halophyte Salicornia bigelovii. Salicornia bigelovii plants were treated with 0, 50, 200, and 600 mM NaCl for 6 weeks. Membrane preparations for each treatment were obtained in triplicates and were separated into 12 fractions per sample through a continuous sucrose gradient. Based on protein relative abundances per fraction, organellar membrane proteomes were generated. Protein abundances were also compared across treatments.

Project description:The root-colonizing fungal endophyte Serendipita indica, formerly known as Piriformospora indica, is well known to promote plant biomass production and stress tolerance of its host plants. Previous studies highlighted an important role of calcium Ca2+ signaling in the establishment of the plant–fungus interaction. We here report the comparative analysis of the effect of a mock- and S. indica-infection on both wild-type Arabidopsis plants (Col-3) and cbl7 knockout mutants. Our data provide evidence for the involement of the Ca2+ sensor CBL7 in the control of potassium distribution in the plant and in adjusting plant defense responses to allow the establishment of the plant–fungus symbiosis. The impairment of CBL7 was shown to translate into increased induction of plant defense-related genes.

Project description:Plant species posses a special set of genes functional only in arbuscular mycorrhizal symbiosis. So, the model plant Medicago truncatula (Jemalong 5) was used for transcriptome comparative analysis while infected with compatible rhizobia Sinorhizobium meliloti (strain 10) and with or without arbuscular mycorrhizal fungus Rhizophagus irregularis (SYM5). Whole shoot and whole root were used for RNA isolation and processed via one of the European certified Affymetrix core labs (http://core.img.cas.cz).

Project description:Numerous Trichoderma strains are beneficial for plants, promote their growth and confer stress tolerance. A recently described novel Trichoderma strain strongly promotes growth of Arabidopsis thaliana seedlings on media with 50 mM NaCl, while 150 mM NaCl strongly stimulated root colonization and induced salt-stress tolerance in the host without growth promotion. To understand the dynamics of plant-fungus interaction, we examined the secretome from both sides, and revealed a substantial change under different salt regimes, and during co-cultivation. Stress-related proteins, such as fungal Kp4-, WSC- and CFEM-domain-containing proteins, the plant calreticulin and cell-wall modifying enzymes, disappear when the two symbionts are co-cultured under high salt concentrations. More proteins involved in plant and fungal cell wall modifications and the battle of root colonization are found in the co-cultures under salt stress, while the number of plant antioxidant proteins decreased. We identified symbiosis- and salt concentration-specific proteins for both partners. The Arabidopsis PYK10 and a fungal prenylcysteine lyase are only found in the co-culture which promoted plant growth. The comparative analysis of the secretomes suggests that both partners profit from the interaction under salt stress but have to invest more in balancing the symbiosis. We discuss the role of the identified stage- and symbiosis-specific fungal and plant proteins for salt-stress and conditions promoting root colonization and plant growth.

Project description:High ambient temperature regulated the plant systemic response to the beneficial endophytic fungus Serendipita indica. Most plants in nature establish symbiotic associations with endophytic fungi in soil. Beneficial endophytic fungi induce a systemic response in the aboveground parts of the host plant, thus promoting the growth and fitness of host plants. Meanwhile, temperature elevation from climate change widely affects global plant biodiversity as well as crop quality and yield. Over the past decades, great progresses have been made in the response of plants to high ambient temperature and to symbiosis with endophytic fungi. However, little is known about their synergistic effect on host plants. The endophytic fungus Serendipita indica colonizes the roots of a wide range of plants, including Arabidopsis. Based on the Arabidopsis-S. indica symbiosis experimental system, we analyzed the synergistic effect of high ambient temperature and endophytic fungal symbiosis on host plants. By transcriptome analysis, we found that DNA replication-related genes were significantly upregulated during the systemic response of Arabidopsis aboveground parts to S. indica colonization. Plant hormones, such as jasmonic acid (JA) and ethylene (ET), play important roles in plant growth and systemic responses. We found that high ambient temperature repressed the JA and ET signaling pathways of Arabidopsis aboveground parts during the systemic response to S. indica colonization in roots. Meanwhile, PIF4 is the central hub transcription factor controlling plant thermosensory growth under high ambient temperature in Arabidopsis. PIF4 is also involving JA and/or ET signaling pathway. We found that PIF4 target genes overlapped with many differentially expressed genes (DEGs) during the systemic response, and further showed that the growth promotion efficiency of S. indica on the pif4 mutant was higher than that on the wild type plants.

Project description:The early phase of the interaction between tree roots and ectomycorrhizal (ECM) fungi, prior to symbiosis establishment, is accompanied by a stimulation of lateral root (LR) development. We set out to identify gene networks that regulate LR development during the early signal exchanges between Populus tremula x Populus alba (hereafter called poplar) and the ECM fungus Laccaria bicolor. A sandwich culture system was developed in order to bring plant and fungus into an indirect contact, which permits signal molecule exchange and LR stimulation (emergence after 4-5 days of contact) but prohibits root colonization. A NimbleGen full genome poplar oligo-array was used to investigate transcript profiles at three days of indirect poplar/L. bicolor contact, referring to the time point of LR initiation in response to the fungus.

Project description:During plant life, roots support beneficial associations with soil inhabiting microbes, mainly fungi and bacteria. Increasing evidence suggests that plant-associated microorganisms can promote plant growth and expand immune functions of the plant host. The beneficial entomopathogenic fungus Beauveria bassiana colonizes plant tissues in an asymptomatic way, triggering poorly characterized plant metabolic changes, which negatively affect both pest insects and plant pathogens. B. bassiana has an extremely broad host spectrum, including tomato (Solanum lycopersicum L.). In order to obtain a great overall snapshot of molecular events regulating tomato-B. bassiana interaction, plant proteome and hormonome changes induced over the time by the fungus have been in-depth analyzed by using a combination of high throughput profiling techniques and bioinformatics tools. The proteomics data highlighted interesting molecular pathways affected by B. bassiana related to primary and secondary metabolism and plant growth. Additionally, downregulation of a member of the endochitinase family and upregulation of calcium channel and transporter proteins suggested well-established plant-fungus symbiosis. The molecular pathways linked to protein/amino acids turn-over and to the biosynthesis of energy compounds shed some light on the strategies exploited by the plant to get the most out of the beneficial interaction in improving growth and development.
