Project description:Bacterial cell envelope is the first and the major line of defense against threats from the environment. Because of its crucial roles in bacterial cell life, cell envelope is a prime target for numerous antibiotics. In this study, by treating Gram-positive model strain Bacillus subtilis with numbers of cell wall targeted antibiotics, we aimed to obtain a global comprehensive transcriptional profile of cell envelope stress response in B. subtilis via transcriptomic study using high-throughput RNA sequencing approach. This knowledge will then be subject to a series of comparative genomics analyses to get detailed information of the distribution and conservation of cell envelope stress-sensing regulatory systems in B. subtilis.

Genome-wide mapping of H3K27me3 in Daxx-depleted cells

Project description:We discovered Daxx can negatively regulated H3K27me3 level via facilitating long non-coding RNA HOTAIR degradation. To further explore the biological consequence and search for affected genes, we used high throughput sequencing technology to demonstrate the enrichment of H3K27me3 on the human genome and target genes repression after depletion of Daxx. This study revealed a novel role of Daxx in regulating histone modifications via modulates long non-coding RNA stability.

Project description:The goal of these experiments were to test the on-target and target-adjacent editing efficiencies of different single-nucleobase editing systems. Previous studies have shown that tethering DNA mutating enzymes to Cas9-nickase-UGI complexes results in editing of chromosomal DNA. However, these editing events encompass undesirable target-adjacent nucleobase edits. Here, we characterize a novel approach that reduces the frequency of target-adjacent editing while maintaining a high level of on-target editing.
