Project description:Abstract: Ammonia is one of the most prominent air pollutants in poultry houses. High levels of ammonia have adverse effects on respiratory health, growth performance, meat production of broilers, and breast meat growth and yield are critical important in the broiler industry. To date, studies focus on the negative relationship of ammonia exposure and breast muscle tissue are still very limited, and the underlying molecular mechanisms remain poorly understood. In this study, high concentrations of atmospheric ammonia were found to lower slaughter rate and broiler breast meat yield significantly (P < 0.05). To explore the candidate genes that ammonia regulates breast meat yield of broilers, high throughout RNA-Seq was used to compare the transcriptome of breast muscle with different ammonia exposure (50 ppm vs 3 ppm). In total, 129 differentially expressed genes (DEGs) were identified (P-value < 0.05; fold-change ≥ 2), among which 87 genes were significantly down-regulated and 42 were up-regulated. Bioinformatics analysis suggested that DEGs (such as PDK4, ACSL1, GLUL, FBXO32) were involved in fatty acid degradation/metabolism, nitrogen metabolism, PPAR signaling and adipocytokine signaling pathways. Functional annotation showed that DEGs were mainly enriched in reactive oxygen species metabolic process and muscle contraction. It can be concluded that decreased meat yield was due to the DEGs participating in above biological processes and pathways. This study provides novel insights into transcriptional differences in breast meat between high- and low-ammonia exposed broiler chickens.

Project description:<p><strong>BACKGROUND:</strong> Chicken meat has become a major source of protein for human consumption. However, the quality of the meat is not yet under control, especially since pH values that are too low or too high are often observed. In an attempt to get a better understanding of the genetic and biochemical determinants of the ultimate pH, two genetic lines of broilers were divergently selected for low (pHu−) or high (pHu+) breast meat pHu. In this study, the serum lipidome of 17-day-old broilers from both lines was screened for pHu markers using liquid-chromatography coupled with mass spectrometry (LC-HRMS).</p><p><strong>RESULTS:</strong> A total of 185 lipids belonging to 4 groups (glycerolipids, glycerophospholipids, sterols, sphingolipids) were identified in the sera of 268 broilers from the pHu lines by targeted lipidomics. The glycerolipids, which are involved in energy storage, were in higher concentration in the blood of pHu− birds. The glycerophospholipids (phosphatidylcholines, phosphatidylethanolamines) with long and polyunsaturated acyl chains were more abundant in pHu+ than in pHu− while the lysophosphatidylcholines and lysophosphatidylethanolamines, known to be associated with starch, were observed in higher quantity in the serum of the pHu− line. Finally, the concentration of the sterols and the ceramides, belonging to the sphingolipids class, were higher in the pHu+ and pHu−, respectively. Furthermore, orthogonal partial least-squares analyses highlighted a set of 68 lipids explaining 77% of the differences between the two broilers lines (R2Y = 0.77, Q2 = 0.67). Among these lipids, a subset of 40 predictors of the pHu value was identified with a Root Mean Squared Error of Estimation of 0.18 pH unit (R2Y = 0.69 and Q2 = 0.62). The predictive model of the pHu value was externally validated on 68 birds with a Root Mean Squared Error of Prediction of 0.25 pH unit.</p><p><strong>CONCLUSION:</strong> The sets of molecules identified will be useful for a better understanding of relationship between serum lipid profile and meat quality, and will contribute to define easily accessible pHu biomarkers on live birds that could be useful in genetic selection.</p>

Project description:Background: High concentrations of atmospheric ammonia are one of the key environmental stressors affecting broiler production performance, which causes remarkable economic losses as well as potential welfare problems of the broiler industry. Previous reports have demonstrated that high levels of ammonia can alter body fat distribution and meat quality of broilers. However, the molecular mechanisms and metabolic pathways in breast muscle altered by high concentrations of ambient ammonia exposure on broilers are still unknown. Results: This study utilized RNA-Seq to compare the transcriptomes of breast muscles to identify differentially enriched genes in broilers exposed to high or low concentrations of atmospheric ammonia. A total of 267 promising candidate genes were identified by differential expression analysis, among which 67 genes were up-regulated and 189 genes were down-regulated. Bioinformatics analysis suggested that the up and down-regulated genes were involved in the following two categories of cellular pathways and metabolisms: Steroid biosynthesis (gga00100) and peroxisome proliferator-activated receptor (PPAR) signaling pathway (gga03320), which both participate in the lipid metabolism processes. Conclusions: This study suggests that longtime exposure to high concentrations of aerial ammonia can change fat content in breast muscle, meat quality and palatability via altering expression levels of genes participating in important lipid metabolism pathways. This study maybe provides new information that could be used for genetic breeding and nutritional intervention in production practice of broilers industry in the future.

Project description:Domestic chicken has been intensively studied because of its role as an efficient source of lean meat. However, commercial broilers resulting from genetic selection for rapid growth demonstrate detrimental traits, such as excess deposition of abdominal adipose tissue, metabolic disorders, and reduced reproduction. Therefore fast-growing broilers represent “obese” chickens compared to slow-growing egg layers (e.g, Leghorn) or wild strain of meat-type chickens (e.g., Fayoumi). Fayoumi chickens, originating from Egypt, represent a harder stain of chickens, which are more resistant to diseases. Leghorn chickens are the original breed of commercial U.S layers. Both lines were maintained highly inbred by Iowa State University poultry geneticists with an inbreeding coefficient higher than 0.95. Both Fayoumi and Leghorn demonstrated lean phenotype compared to broilers, and these three lines of chickens are genetically distant from each other. In this study, we used affymetrix microarray to profile global gene expression of three distinct genetic lines of chickens to identify functional pathways associated with leanness of domestic chickens.
