Project description:The mammary gland redeveloped to the pre-pregnancy state during involution, which shows that the mammary cells have the characteristics of remodeling. The rapidity and degree of mammary gland involution are different between mice and dairy livestock (dairy cows and dairy goats). However, the molecular genetic mechanism of involution and remodeling of goat mammary gland has not yet been clarified. Therefore, this study carried out the RNA-sequencing of nonlactating mammary gland tissue of dairy goats in order to reveal the transcriptome characteristics of nonlactating mammary tissues and clarify the molecular genetic mechanism of mammary cell involution and remodeling.

Project description:In this study, two small RNA libraries were constructed using dry period and peak lactation dairy goat mammary gland tissues and sequenced by the Illumina Solexa high-throughput sequencing system. A total of 346 conserved and 95 novel miRNAs were identified in the dairy goat. The expression of miRNAs was confirmed by qRT-PCR in nine tissues and the mammary gland during development cycles. In addition, several candidate miRNAs that may be involved in mammary gland development and lactation were found by the comparison of miRNA expression profiles among different tissue and developmental stages of the mammary gland. This study provides the identification and profile of miRNAs related to the biology of the mammary gland in the dairy goat. The identification of these miRNAs could contribute to understanding the molecular mechanisms of lactation physiology and the development of the mammary gland in the dairy goat.

Project description:Milk fatty acids secreted by the mammary glands are one of the most important determinants of the nutritional value of goat milk. However, there are few data available on the transcriptome-wide changes across lactation in the goat. In this study, goat mammary tissue from the periods encompassing lactation, cessation of milking and involution were used for digital gene expression sequencing (DGE) using the goat transcriptome as reference sequence. A total of 51299 unigenes were identified and annotated to 12763 genes, of which 9131 genes were differentially expressed (DEG) among the various stages of lactation. Functional classification through KOG, GO, and KEGG was used for selection of the top expressed genes and the differentially expressed genes (DEG). DEG series-cluster analysis revealed 16 possible expression patterns. Using co-expression analysis, within the lipid transport and metabolism KOG categories, PLA2, CPT1, PLD had the most number of correlated genes. GGA, SRPRB and AP4S1 were the top 3 with the highest number of correlated genes associated with intracellular trafficking, secretion, and vesicular transport. These data may provide candidate genes with a high probability of having functional roles in regulating of goat fatty acid metabolism during the various stages of lactation.

Project description:Mammary gland branching morphogenesis is thought to depend on the mobilization of proteolytic machinery from the matrix metalloproteinase (MMP) family, namely MT1-MMP/MMP14, to drive coordinated epithelial cell invasion through the interstitial extracellular matrix, but the dominant effector has remained undefined. Unexpectedly, we find MMP14 controls postnatal mammary gland branching from the periductal stroma. Transcriptome profiling of stromal cell-targeted mammary glands was used to characterize the impact of stromal Mmp14-targeting on the growth factor and signaling cascades implicated in mammary gland morphogenesis. Transcriptome profiling of ductal networks and associated stroma was used to investigate the functional roles of MMP14 in the postnatal mammary gland stroma in an unbiased fashion.

Project description:Mammary epithelial cells (MECs) have strong secretory function, which is an important target cell for studying mammary gland bioreactor. It is also a unique cell with lactation function in mammary gland. It can be used to study cell biological characteristics and molecular biological mechanism in the process of mammary gland growth, development and lactation. At present, mammary epithelial cells are obtained from mammary tissue, and the extraction of tissue block involves surgical anesthesia, which may lead to breast tissue injury and infection. This study aimed to describe the process of isolating and culturing goat mammary epithelial cells (GMECs) from goat milk, and to identify the biological characteristics of GMECs. Our study laid a foundation for the study of the regulation mechanism of mammary gland development and lactation, the improvement of milk quality, the increase of milk production and the establishment of mammary gland bioreactor.

Project description:Mycoplasma agalactiae (Ma) is one of the main aetiological agents of intramammary infections in small ruminants, causing contagious agalactia. To better understand the underlying disease patterns a primary goat mammary epithelial cell (pgMEC) culture was established from the mammary tissue and it was challenged with Ma. High-throughput mRNA sequencing was performed to reveal differentially expressed genes (DEG) at different time-points (3 h, 12 h, and 24 h) post infection (PI). The pathway enrichment analysis of the DEG showed that infection significantly affected pathways associated with immune response, steroid metabolism, fatty acid metabolism, apoptosis signalling, transcription regulation, and cell cycle regulation. Based on the results we suggest that mammary epithelial cells in vivo contribute to the immune system by the induced expression of cytokines and other chemotactic agents, activation of the complement system and apoptosis pathways, and expression of genes coding for antimicrobial molecules and peptides. In our study we attempted to interpret the detected transcriptomic changes in a biological context and infer mammary infection resistance candidate genes, interesting for further validation. Additionally, the results represent comprehensive goat mammary transcriptome information and demonstrate the applicability of the comparative genomics approach for annotation of goat data, using transcriptome information of a closely related species (Bos taurus) as a reference.
