Project description:Bioassay-guided fractionation of a CH(2)Cl(2)-MeOH extract of the aerial parts of Albizia inundata resulted in the isolation of two new natural oleanane-type triterpene saponins {3-O-[?-L-arabinopyranosyl(1?6)]-2-acetamido-2-deoxy-?-D-glucopyranosyl oleanolic acid (1) and 3-O-[?-L-arabinopyranosyl(1?2)-?-L-arabinopyranosyl(1?6)]-2-acetamido-2-deoxy-?-D-glucopyranosyl acacic acid lactone (2)} along with seven known saponins {3-O-[?-L-arabinopyranosyl(1?6)]-2-acetamido-2-deoxy-?-D-glucopyranosyl echinocystic acid (3), 3-O-[?-D-xylopyranosyl (l?2)-?-L-arabinopyranosyl(l?6)]-2-acetamido-2-deoxy-?-D-glucopyranosyl acacic acid lactone (concinnoside D) (4), 3-O-[?-D-glucopyranosyl(l?2)]-?-D-glucopyranosyl oleanolic acid (5), 3-O-[?-L-arabinopyranosyl(1?2)-?-L-arabinopyranosyl(l?6)]-?-D-glucopyranosyl oleanolic acid (6), 3-O-[?-D-xylopyranosyl(1?2)-?-L-arabinopyranosyl(l?6)]-?-D-glucopyranosyl oleanolic acid (7), 3-O-[?-L-arabinopyranosyl(l?2)-?-L-arabinopyranosyl(1?6)-[?-D-glucopyranosyl(l?2)]-?-D-glucopyranoside echinocystic acid (8), and 3-O-[?-D-xylopyranosyl(l?2)-?-L-arabinopyranosyl(1?6)-[?-D-glucopyranosyl(l?2)]-?-D-glucopyranoside echinocystic acid (9)}. The structures of 1 and 2 were established on the basis of extensive 2D NMR ((1)H-(1)H COSY or DQF-COSY, HSQC, HMBC, TOCSY, and HSQC-TOCSY) spectroscopic, ESIMS, and chemical methods. Saponins 1, 3, 6, and 7 showed cytotoxicity against human head and neck squamous cells (JMAR, MDA1986) and melanoma cells (B16F10, SKMEL28) with IC(50) values in the range 1.8-12.4 ?M, using the MTS assay.

Project description:The bark, leaves, and roots of Albizia adianthifolia are highly sought after in tropical Africa as herbal medicines. Therefore, the aim of this study was to review the botany, medicinal uses, phytochemistry, and pharmacological properties of A. adianthifolia so as to provide baseline data required for evaluating the therapeutic potential of the species. Information on the botanical profile, medicinal uses, phytochemistry, and pharmacological properties of A. adianthifolia was undertaken using databases such as ScienceDirect, SciFinder, Pubmed, Google Scholar, Medline, SCOPUS, EThOS, ProQuest, OATD, and Open-thesis. Preelectronic literature search of conference papers, scientific articles, books, book chapters, dissertations, and theses was carried out at the University library. Literature search revealed that A. adianthifolia is used as purgative and herbal medicine for diabetes, eye problems, gastrointestinal problems, haemorrhoids, headache, neurodegenerative disorders, reproductive problems in women, respiratory problems, wounds and pain, skin diseases, sexually transmitted infections, and ethnoveterinary medicine. Phytochemical compounds identified from the species include apocarotenoids, chalcone, dipeptide, elliptosides, essential oils, fatty acids, flavonoids, histamine, imidazolyl carboxylic acid, prosapogenins, steroids, triterpene saponins, and triterpenoids. Pharmacological studies revealed that A. adianthifolia extracts and compounds have acetylcholinesterase enzyme inhibitory, anthelmintic, antiamoebic, antibacterial, antimycobacterial, anti-sexually transmitted infections, antifungal, anti-inflammatory, antioxidant, anxiolytic, and antidepressant, cognitive-enhancing, haemolytic, hypoglycemic and antihyperglycemic, immunomodulatory, and cytotoxicity activities. Detailed studies on the pharmacokinetics, in vivo, and clinical research involving compounds isolated from A. adianthifolia and extracts of the species are required.

Project description:Bioassay-guided fractionation of an EtOH extract obtained from the roots of the Madagascan plant Albizia gummifera led to the isolation of three new cytotoxic oleanane-type triterpenoid saponins, gummiferaosides A-C (1-3). The structures of these new compounds were elucidated using 1D and 2D NMR experiments and mass spectrometry. Compounds 1-3 showed cytotoxicity against the A2780 human ovarian cancer cell line with IC50 values of 0.8, 1.5, and 0.6 microg/mL, respectively.

Project description:Albizia julibrissin Durazz. is one of the most common herbs used for depression and anxiety treatment, but its molecular basis and mechanism of action as an antidepressant or anxiolytic drug are not understood. In this study, we separated and identified two lignan glycosides that inhibit serotonin transporter (SERT) noncompetitively by decreasing Vmax with little change in Km for its fluorescence substrate. In addition, treatment with lignan glycosides did not alter total and cell surface expression levels of the transporter protein. The two compounds decreased the accessibility of a cysteine residue placed in the extracellular substrate permeation pathway by inducing a conformational shift toward an outward-closed state of SERT. These results are consistent with molecular docking for the association of the lignan glycosides to the allosteric site in SERT. The present work supports the proposal that these compounds act on SERT by a novel underlying mechanism of action different from that of conventional antidepressant drugs.

Project description:The EFSA Panel on Plant Health was requested to prepare and deliver risk assessments for commodities listed in the relevant Implementing Acts as 'High risk plants, plant products and other objects' [Commission Implementing Regulation (EU) 2018/2019 establishing a provisional list of high-risk plants, plant products or other objects, within the meaning of Article 42 of Regulation (EU) 2016/2031]. The current Scientific Opinion covers all plant health risks posed by Albizia julibrissin imported from Israel, taking into account the available scientific information, including the technical information provided by Israel. The relevance of an EU-regulated pest for this opinion was based on evidence that: (i) the pest is present in Israel; (ii) A. julibrissin is a host of the pest and (iii) the pest can be associated with the commodity. The relevance of this opinion for other non EU-regulated pests was based on evidence that (i) the pest is present in Israel; (ii) the pest is absent in the EU; (iii) A. julibrissin is a host of the pest; (iv) the pest can be associated with the commodity and (v) the pest may have an impact and can pose a potential risk for the EU territory. Three pests (two insects, Aonidiella orientalis and Euwallacea fornicatus; one fungus, Fusarium euwallaceae) that fulfilled all criteria were selected for further evaluation. For the three selected pests, the risk mitigation measures proposed in the technical dossier were evaluated. Limiting factors on the effectiveness of the measures were documented. For the selected pests, an expert judgement on the likelihood of pest freedom is given taking into consideration the risk mitigation measures acting on the pest, including uncertainties associated with the assessment. The Panel is 95% sure that 9,950 or more units per 10,000 will be pest free from these three pests.
