Expression data from ADORA1 knockout and ADORA1 APOE double-knockout mice

Project description:The goal of this study is to determine whether A1 adenosine receptor (ADORA1) plays a role in atherosclerosis development and its possible mechanisms. This dataset compares gene expression (aortas) of ADORA1 knockout mice to ADORA1+APOE double-knockout mice. Mice deficient in both ADORA1 and APOE (DKO) demonstrated reduced atherosclerotic lesions in aortic arch (en face), aortic root, and innominate arteries when compared to (APOE-KO) of the same age. Treating APOE-KO with the ADORA1 antagonist DPCPX also achieved concentration-dependent reduction in lesions. The total plasma cholesterol and triglyceride levels were not different between DKO and APOE-KO, however, higher triglyceride was observed in DKO fed a high-fat diet. DKO also were heavier than APOE-KO. Plasma cytokine levels (IL-5, IL-6, and IL-13) were significantly lower in DKO. Proliferating cell nuclear antigen (PCNA) expression was also significantly reduced in the aorta from DKO. Despite smaller lesions in DKO, the composition of the innominate artery lesions and cholesterol loading and effusion [export] from bone-marrow-derived macrophages from DKO were not different from APOE-KO.

Single cell sequencing of murine CD4+ T cells from atherosclerotic aortas

Project description:We report the single cell transcriptome of CD4+ T cells from atherosclerotic aortas from ApoE-deficient mice that have been fed a western diet (WD) with aggravated atherosclerosis or with a standard chow diet (CD) with moderate atherosclerosis. We show that aortic T cells have a unique transcriptome with a mixed phenotype overlapping with T-regulatory and pathogenic T-helper type -1 and -17 cells.

Aortic macrophages and endothelial cells of apoE KO mice aortas after plaque regression induced apoE adenoviral transfer

Project description:Aortic macrophages and endothelial cells of apoE KO mice were sorted and analyzed by microarray 2 weeks after regression was induced by adenoviral transfer of apoE. Aortic macrophages (CD45+ F4/80+ CD11b+) and endothelial cells (CD45- CD31+) were sorted from apoE KO mice and the RNA extracted and hybridized to Affymetrix Mouse Gene 1.0 ST array. We pooled aortas from 5 mice for each sort.

miRNA expression profile in aortas from LysM-Cre/Dicerflox/flox/Apoe–/– mice compared with LysM-Cre/Dicerwt/wt/Apoe–/– mice fed with high-fat diet for 12 weeks

Project description:The global change of the miRNA expression profile during atherosclerosis is due to the infiltration of different types of leukocytes into the arterial vessel wall in addition to disease-specific regulation in vascular cells. Monocyte-derived macrophage accumulation in the subintimal region is critical in the formation of atherosclerotic plaques. However, the role of Dicer, the key enzyme for miRNA biogenesis, during the development of atherosclerosis is currently unknown. To detect the effect of Dicer on miRNA expression in lesional macrophages, the comparison of the miRNA expression profiles was performed in aortas from Apoe deficient mice with or without a myeloid cell-specific knockout of Dicer after 12-week high-fat diet (HFD) programme. This screening combined with functional study may help to identify the crucial miRNAs that play a role in the macrophage function during atherogenesis.
