Project description:Candida albicans can form biofilm on the surface of indwelling medical devices. Biofilm formation is an importanat clinical problem because biofilm-grown cells have decreased susceptibility to antifungal agents. Microarray technology was used to identify changes in gene expression during biofilm development. Two biofilm substrates (denture and catheter) were used, with two C. albicans strains tested on each substrate. Three phases of biofilm development were studies: early (6 h), intermediate (12 h), and mature (48 h). Planktonic specimens were collected at the same time points. Comparison between biofilm and planktonic cell transcriptional profiles at each time point showed differential expression of approximately 3% of the genome in biofilm. Fewer than half of these genes were up-regulated in biofilm, compared to planktonic cells. Transcriptional profiles were also analyzed over the time course of biofilm development. Genes up-regulated during the early phase (6 h) primarily were involved in glycolytic and non-glycolytic carbohydrate assimilation, and amino acid metabolism. The largest number of differentially expressed genes were identified at the intermediate phase (12 h) of biofilm development where the largest increase in biofilm biomass occurs. Genes up-regulated at 12 h were involved in transcription, protein synthesis/translation, energy generation, cellular transport, and nucleotide metabolism. At mature phase (48 h), few genes were up-regulated compared to the 12 h time point. These data define phase-dependent changes in gene expression that occur during biofilm development and show how genes belong to different, but interconnected, functional categories regulate the morphology and phenotype of C. albicans biofilm Keywords: phase-dependent gene expression; comparative genomic hybridization; cell type comparison

Project description:Campylobacter jejuni, an important pathogen of bacterial gastrointestinal infections, forms biofilms that are crucial for its survival in different environments. We used RNA sequencing to investigate the changes in gene expression at different stages of biofilm formation (16 h, 24 h, 48 h and 72 h). The results showed that early biofilms (16 h and 24 h biofilms) show increased expression of genes involved in cysteine and methionine amino acid metabolism, while mature biofilms (48 h and 72 h biofilms) show decreased expression of capsular polysaccharides and lipooligosaccharides. Both early and mature biofilms are characterized by increased expression of genes involved in flagella formation, leucine amino acid metabolism and oxidative stress response, and decreased expression of genes involved in energy metabolism, iron acquisition and transmembrane transport. Thus, we provide a deeper understanding of the molecular mechanisms of C. jejuni biofilm maturation, environmental resistance and the dynamic nature of gene expression that facilitates adaptation to biofilm-associated lifestyles

Project description:Salmonella spp. biofilms have been implicated in persistence in the environment and plant surfaces. In addition, Salmonella is able to form biofilms on the surface on cholesterol gallstones. The ability of Salmonella spp. on these surfaces is superior to biofilm formation on surfaces on glass or plastic. Thus, we hypothesized that Salmonella gene expression is specific during biofilm development on cholesterol surfaces.

Project description:The aim of this study was to investigate the plastic colonisation process, to identify the active taxa involved in biofilm formation and the mechanisms used to initiate colonisation. To achieve this, a marine plastisphere characterised by active hydrocarbonoclastic genera was used as the inoculum for a short-term microcosm experiment using virgin low-density polyethylene as the sole carbon source. Following incubation for 1 and 2 weeks (representing early and late colonisation, respectively), a taxonomic and comparative metaproteomic approach was used to explore shifts in diversity and function.

Project description:In this work we have demonstrated increased mutability of Staphylococcus aureus and S. epidermidis in biofilms and have explored the mechanisms underlying the enhanced mutability. A novel static biofilm model, utilising cellulose filter disks, was developed to support the formation of mature biofilms with sufficiently high cell densities to permit determination of mutation frequencies. The mutability of biofilm cultures increased up to 60 fold and 4 fold for S. aureus and S. epidermidis, respectively, compared with planktonic cultures. Incorporation of antioxidants into S. aureus biofilms reduced mutation frequencies, indicating that increased oxidative stress underlies increased mutability in the biofilm. Transcriptional profiling revealed upregulation of the superoxide dismutase gene, sodA, in early biofilm cultures, also suggesting enhanced oxidative stress in these cultures. However, loss of the genes encoding superoxide dismutases or peroxidases did not specifically exacerabate biofilm mutability. In S. aureus SH1000, hydrogen peroxide was found to contribute to biofilm mutability.
