Project description:Bmi1tm1(cre/ERT2)Mrc/J (a.k.a. Bmi1creERT2 mice), B6.Cg-Gt(ROSA)26Sortm14(CAT-tdTomato)Hze/J (a.k.a. Rosa26tom mice) were crossed. The mice were fed with New Western diet 1 (NWD1) or Americal Instititue of Nutrition 76A (AIN76A or AIN) diet from weaning for 3 months. Tamoxinfen was injected to induce Bmi1 cell dedifferentiation, featuring the expression of tomato red fluoresence in the Bmi1 locus. Single intestinal epithelial cells were FACS isolated by the Epcam+, CD45- and Tomato (red fluorescence)+, and subject to scRNAseq assay.

Bmi1 defines long-term adult cardiac stem cells in heart homeostasis and repair

Project description:We have found the existence of a Bmi1+ population in the adult heart contributing to the organ low-rate turnover and repair with the generation of new cardiomyocytes. We show that the Bmi1+ population is a sub-population of the cardiac Sca-1+ progenitor cells. We have analyzed the gene profile by deep-sequencing (RNA-Seq) of Bmi1+ and Sca-1+Bmi1- cells in homeostatic heart condition. On the other hand, we have compared gene profile by deep-sequencing (RNA-Seq) of Bmi1+ cells in homeostatic condition versus Bmi1+ cells 5 days after myocardial infarction (MI). Analysis of RNA-Seq data revealed a differential expression signature between both subsets of cardiac stem/progenitors cells in homeostatic condition and also differences between Bmi1+ cells after AMI versus homeostatic condition.

Reprogramming of fibroblasts into induced pluripotent stem cells with Bmi1

Project description:Somatic cells can be reprogrammed into induced pluripotent stem (iPS) cells by Oct4, Sox2, Klf4, plus c-Myc. Recently, Sox2 plus Oct4 were shown to reprogram fibroblasts and Oct4 alone to reprogram mouse and human neural stem cells (NSCs) into iPS cells. Here we report that Bmi1 leads to dedifferentiation of mouse fibroblasts into NSC-like cells and, in combination with Oct4, replaces Sox2, Klf4 and c-Myc during reprogramming fibroblasts to iPS cells. Furthermore, activation of sonic hedgehog signalling (by Shh, purmorphamine, or oxysterol) replaces the effects of Bmi1, and, in combination with Oct4, reprograms mouse embryonic and adult fibroblasts into iPS cells. One-and two-factor iPS cells are similar to mouse embryonic and adult fibroblasts into iPS cells in global gene expression profile, epigenetic status, in vitro and in bibo differentiation into all three ferm layers, as well as teratoma formation and germline transmission in vivo. These data support that fibroblasts can be reprogrammed into iPS cells by Oct4 alone. Total RNAs were isolated from indicated cells and labeled with Cy3. Hybridization was performed once for each sample.

Project description:We report the application of single cell RNAseq assay in examining the effect of Ppargc1a gene expression in intestinal stem cells on mucosal homeostasis. Lgr5EGFPcreERT2 mice were crossed with Ppargc1a F/+ or F/F to selectively target gene expression in Lgr5 expressing intestinal stem cells. Mice were fed with control diet AIN76A from weaning for 3 months, and deletion of Ppargc1a gene was conducted via Tamoxifen injection. Mice were sacrificed 3 days post tamoxifen injection.

Gene expression in the liver of Bmi1 knockout mice

Project description:To explore the potential targets of Bmi1 in the liver development of hepatic carcinogenesis, we assayed the gene expression level in the liver of Bmi1 knockout mice. We isolated the liver tissue of Bmi1 WT and KO mice around 6-8 weeks. Then we extracted total RNA and run the microarray detection. Gene expression in Bmi1 KO mouse livers was compared with that in Bmi1 WT mouse livers to screen potential targets of Bmi1.
