And we further evaluated the role of miRNAs in inflammation induced by flagellin.

Project description:Macrophages are an important component of the innate immune system. Flagellin has been extensively studied for its adjuvant activity owing to its TLR5 and NLRC4 binding sites. However, few studies have comprehensively investigated the effects of flagellin in macrophages using transcriptome sequencing. In this study, RNA sequencing (RNA-seq) was used to analyze the expression patterns of RAW264.7 cells induced by flagellin (FliC) of Salmonella typhimurium compared with unstimulated cells, to identify novel transcriptomic signatures in macrophages.

Project description:miRNA expression profiling in tumor-associated BMDCs induced by tumor cells 1D8 or CT-26 in comparison with those cultured in medium. To prepare tumor-associated BMDCs(Bone Marrow-derived Dendritic Cells), murine BMDCs were co-cultured with 1D8 or CT-26 cells in a transwell plate in 1640 medium supplemented with 3% FCS and 1% penicillin and streptomycin for 24 hrs. Total RNAs were isolated using the TRIZOLM-BM-. Reagent(Invitrogen) according to manufacturerM-bM-^@M-^Ys instructions.The samples were labeled using the miRCURYM-bM-^DM-" Array Power labeling kit (Exiqon). miRNA array hybridization was performed, and unbound miRNA Labels were washed away. Microarrays were analyzed using GenePix 4000B scanner and GenePix Pro 6 software (Molecular Devices).
Project description:Purpose: The purpose of this study is to identify the genome-wide binding sites for IRF4 interaction partners PU.1, BATF, and JunB in dendritic cells. These ChIP-seq data were integrated with gene expression analysis in IRF4-sufficient and -deficient BMDCs in order to assemble an IRF4 gene regulatory network. Hematopoietic bone marrow progenitors from C57BL/6 mice were differentiated with GM-CSF and IL-4 for 5 days. On day 6, BMDCs were stimulated for 6 hours with 100ng/ml LPS. Fixed chromatin was immunoprecipitated with anti-PU.1, BATF, and JunB antibodies and subjected to high-throughput sequencing. The sequencing data for the input DNA was previously submitted as GSM999807.
