Project description:To analyze both early and late events associated with Smad3-inactivation, and to identify how specific smooth muscle cell (SMC) phenotypes may confer increased susceptibility to disease in the aortic root, we performed single-cell transcriptomic analysis on aortic cells from control (Smad3+/+; Myh11-CreER; + or – EGFP-L10a mice are refered to as Smad3+/+) and Smad3-deficient (Smad3lox/lox; Myh11-CreER; + or - EGFP-L10a are referred to as Smad3SmKO) mice at 10- and 18-weeks post-deletion, when aortic dilation is undetectable and moderate, respectively. Analysis of SMC-specific transcriptional changes show that, in addition to the expected decrease in TGF-β -induced expression of extracellular matrix components, Smad3-inactivation resulted in a broad reduction in the expression of components and modulators of focal adhesions, including that of several integrins, anchoring proteins, adaptors and modulators.

Project description:The role of neutrophils in tumor is still controversial and largely unknown. We previously identified an anticancer tumor microenvironment from mice lacking Smad3, but the underlying mechanism is still largely unclear. Interestingly, depletion of neutrophils largely suppressed the anticancer phenotype of Smad3-KO mice against syngeneic murine Lewis lung carcinoma (LLC). Here, we discovered that Smad3 is essential for promoting N1/N2 polarization of the tumor_x0002_associated neutrophils (TANs) in lung carcinoma. We found that Smad3 deficiency dramatically increased N1 TANs associated with an anticancer phenotype in mice and detected a negative correlation between Smad3 activation and anticancer N1 associated with a higher mortality of NSCLC. By conducting a neutrophil-specific single-cell RNA-sequencing, we uncovered that Smad3 is essential for completing the N1/N2 polarization of TANs, whereas N1 will be the dominated phenotype in the Smad3-KO TME by analyzing the developmental pathways of TANs with pseudo-time at transcriptome level. Mechanistically, direct binding of Smad3 on the developmental genes in neutrophils under cancer condition was evidenced by ChIP-sequencing at genomic level. Importantly, both neutrophil-specific and pharmaceutical inhibition of Smad3 effectively enhanced the anticancer activity of human and murine TANs, thereby suppressing the progression of lung carcinoma in vivo. Thus, Smad3 may represent a precision therapeutic target for enhancing the anticancer activity of TANs via blocking N1/N2 polarization in NSCLC. in C57BL6 (wild-type) and Smad3-deficient (Smad3-/- ) mice and submitted for cell encapsulation and library construction by Chromium controller with 5’ expression kit (10x genomics)

Project description:Granulosa cell tumors (GCTs) are a rare form of stromal cell malignant cancer accounting for one in twenty cases of ovarian cancer. More than 400 tumors from aGCT patients of diverse ethnicities showed one single mutation in the Forkhead box L2 transcription factor gene FOXL2: Cys134Trp (c.402C<G). In this work, seeking more comprehensive profiling of the mutant FOXL2C134W transcriptomic activity in a model of adult Granulosa Cell Tumor (aGTC), we performed an RNA-seq analysis comparing the effect of FOXL2WT/SMAD3 and FOXL2C134W/SMAD3 overexpression in presence/absence of FOXO1 in an established human GC line (HGrC1).

Project description:Understanding the molecular mechanisms of pathological vascular remodeling is important for treating cardiovascular diseases and complications. Recent studies have highlighted a role of deubiquitinases in vascular pathophysiology. In this study, we investigated the role of a deubiquitinase, OTUD1, in angiotensin II (Ang II)-induced vascular remodeling. We identified upregulated OTUD1 in the vascular endothelium of Ang II-challenged mice and showed OTUD1 deletion attenuated vascular remodeling, collagen deposition, and EndMT. Conversely, OTUD1 overexpression aggravated these pathological changes both in vivo and in vitro. Mechanistically, SMAD3 was identified as a substrate of OTUD1 using co-immunoprecipitation followed with LC-MS/MS. We found OTUD1 stabilizes SMAD3 and facilitates SMAD3/SMAD4 complex formation and subsequent nuclear translocation through both K48- and K63-linked deubiquitination. OTUD1-mediated SMAD3 activation regulate the transcription of genes involved in vascular EndMT and remodeling in HUVECs. Finally, SMAD3 inhibition reversed OTUD1-promoted vascular remodeling. Our findings demonstrate endothelial OTUD1 promoted Ang II-induced vascular remodeling by deubiquitinating SMAD3. This study identified SMAD3 as a target of OTUD1 and indicates OTUD1 as a potential therapeutic target for the diseases related to vascular remodeling.

Project description:Atherosclerosis is a persistent inflammatory state accompanied by lipid overload. Vascular fibrosis is one of the primary causes of atherosclerosis development. Although ligustilide (Lig) was shown to exert obvious antiatherogenic effects in previous studies, its precise mechanism has not been comprehensively discussed. In this paper, pharmacologic studies were performed to explore the pharmacodynamic effects of Lig on protecting aorta vascular wall structures and modulating serum inflammatory factors in ApoE-/- mice. Chemical proteomics based on a Lig-derived photoaffinity labelling (Lig-PAL) probe were applied to identify potential therapeutic targets. Mothers against decapentaplegic homologue 3 (SMAD3), which is closely related to the development of vascular fibrosis and atherosclerosis, was identified as a potential target of Lig. Lig suppressed the phosphorylation and nuclear translocation of SMAD3 by blocking the interaction between SMAD3 and transforming growth factor-β (TGF-β) receptor 1, thereby inhibiting the collagen synthesis process, preventing vascular fibrosis and improving atherosclerosis. The quantitative proteomics results from Lig-treated atherosclerotic ApoE-/- mice also indicated that Lig inhibits the expression of collagens I and III, interferes with collagen fibril organization processes and protects the aorta from vascular fibrosis. Hence, developing a novel SMAD3 inhibitor may present another therapeutic option for preventing atherosclerosis.

Project description:Gene expression profiling was carried out in six (wild type, β2SP+/-, β2SP-/-, SMAD3+/-, SMAD3-/- and β2SP+/-/ SMAD3+/-) different mouse knockout embryonic fibroblast (MEF) cells. Beta-2-spectrin (β2SP) is a dynamic intracellular non-pleckstrin homology (PH)-domain protein that belongs to a family of polypeptides that have been implicated in conferring cell polarity. Spectrins have been linked to multiple signaling pathways, including cell cycle regulation, DNA repair and TGFβ signaling. In this study, we report a major role of the TGFβ/Smad3 adaptor β2-Spectrin in conserving genomic integrity from alcohol-induced DNA damage and describe a novel pathway that protects genomes from genotoxic stresses. To determine the mechanism for the oncogenic switch, and whether it is related to the role of β2SP in TGF-β signaling transduction or secondary to its cytoskeletal functions, we analyzed disruption of two elements of the TGF-β pathway by generating double heterozygous Sptbn1+/−/Smad3+/− mice.
