Psychobiotic Lactobacillus plantarum JYLP-326 relieves anxiety, depression, and insomnia symptoms in test anxious college students via modulating the gut microbiota and its metabolism

Project description:Test anxiety frequently occurs in college students and harms their physical and psychological health, but suitable interventions or therapeutical strategies are still missing. The present study aims to evaluate the potential effects of Lactobacillus plantarum JYLP-326 on test anxious college students. Sixty anxious students were enrolled and randomly allocated to the placebo group and the probiotic group, which were instructed to take placebo and JYLP-326 products twice per day for three weeks, respectively. Thirty unanxious students with no treatments were assigned to a regular control group. The anxiety, depression, and insomnia questionnaires were used to measure students’ mental states at the baseline and the end of this study. 16S rRNA sequencing and untargeted metabolomics were performed to analyze the changes in the gut microbiota and fecal metabolism. The questionnaire results suggested that JYLP-326 administration could relieve the symptoms of anxiety, depression, and insomnia in test anxious students. The gut microbiomes of the placebo group showed a significantly greater α diversity index than the control group (p < 0.05). An increased abundance of Bacteroides and Roseburia was observed in the placebo group, and the relative abundance of Prevotella and Bifidobacterium decreased. Whereas JYLP-326 administration could partly restore the disturbed gut microbiota. Additionally, test anxiety was correlation with disordered fecal metabolomics such as a higher Ethyl sulfate and a lower Cyclohexylamine, which could be reversed after taking JYLP-326. Furthermore, the changed microbiota and fecal metabolites were significantly associated with anxiety-related symptoms. These results indicated that the intervention of L. plantarum JYLP-326 could be an effective strategy to alleviate anxiety, depression and insomnia in test anxious college students.

Whole transcriptome profiling reveals major cell types in the cellular immune response against acute and chronic active Epstein-Barr virus infection

Project description:Epstein–Barr virus (EBV) is a common human pathogen that infects over 95% of the population worldwide. In the present study, the whole transcriptome microarray data were generated from peripheral blood mononuclear cells from Chinese children with acute infectious mononucleosis (AIM) and chronic active EBV infection (CAEBV) that were also compared with a publicly available microarray dataset from a study of American college students with AIM. Our study characterized for the first time a broad spectrum of molecular signatures in AIM and CAEBV. The key findings from the transcriptome profiling were validated with qPCR and flow cytometry assays. The most important finding in our study is the discovery of predominant γδ TCR expression and γδ T cell expansion in AIM. This finding, in combination with the striking up-regulation of CD3, CD8 and CD94, suggests that CD8+ T cells and CD94+ NK cells may play a major role in AIM. Moreover, the unique up-regulation of CD64A/B and its significant correlation with the monocyte marker CD14 was observed in CAEBV and that implies an important role of monocytes in CAEBV. In conclusion, our study reveals major cell types (particularly γδ T cells) in the host cellular immune response against AIM and CAEBV.

Project description:Illumina high-throughput sequencing was used to analyse the intestinal bacteria of these two species during different wintering periods at Shengjin Lake. We tested whether contact time enhances the trans-species spread of gut bacteria. Our results indicate that although intestinal microflora of hooded crane and the bean goose were different, direct or indirect contact in the mixed-species flock caused the spread of gut bacteria trans-species, and a very high proportion of common pathogens among these two hosts.

Detection of antibiotic resistance genes in human gut microbiota

Project description:We used a DNA microarray chip covering 369 resistance types to investigate the relation of antibiotic resistance gene diversity with humans’ age. Metagenomic DNA from fecal samples of 123 healthy volunteers of four different age groups, i.e. pre-school Children (CH), School Children (SC), High School Students (HSS) and Adults (AD) were used for hybridization. The results showed that 80 different gene types were recovered from the 123 individuals gut microbiota, among which 25 were present in CH, 37 in SC, 58 in HSS and 72 in AD. Further analysis indicated that antibiotic resistance genes in groups of CH, SC and AD can be independently clustered, and those ones in group HSS are more divergent. Project description:Commensal microbiota contribute to gut homeostasis and influence mucosal gene expression. We harvested mucosal lining of middle and distal part of the small intestine and colon from germ-free (GF) and gnotobiotic mice monocolonized either with the E.coli strain O6K13 (O) or Nissle 1917 strain (N). The expression profiles of the mucosa samples were compared to the corresponding tissue isolated from conventionally reared mice in order to disclose genes differentially expressed in response to the change in the intestinal microflora composition.
