Project description:The aim of this study was to undertake an in-depth and comparative study of the protein expression patterns of subcutaneous and visceral adipose tissues in goats, combined to an mRNA expression study of proteins involved in immune and inflammatory response. Samples were obtained from four healthy goat-kids, Alpine breed, naturally reared by their mothers. Animals were slaughtered at the age of 30 days, during routinely slaughtering procedures, and four different adipose tissues were collected from each animal. Subcutaneous fat was taken from sternum and base of the tail; visceral fat was taken from perirenal and omental depots. The four adipose tissues deposits were selected due to their frequent use in experimental studies on fat tissue. Liver samples were also collected, and used as reference samples. Tissue samples were snap frozen in liquid nitrogen and stored at –80 degrees C until analysis. The 20 runs included in this dataset are from the 4 animals x 5 tissues (sternum, base of the tail, perirenal area, omentum and liver). A 2-D label free LC-MS/MS approach followed by cluster analysis was used for comparing the subcutaneous and visceral fat tissue proteomes. Each sample was analysed in ~12 SCX fractions which correspond to the WIFF files included in each RAW_GOAT0xx.zip files.

Project description:This is the expression dataset for two studies: 1) Characterization of visceral and subcutaneous adipose tissue transcriptome and biological pathways in pregnant and non-pregnant women: Evidence for pregnancy-related regional-specific differences in adipose tissue and 2) Characterization of visceral and subcutaneous adipose tissue transcriptome in pregnant women with and without spontaneous labor at term: Implication of alternative splicing in the metabolic adaptations of adipose tissue to parturition. The studies compare expression profiles and exon usage between adipose tissue regions and groups of women (pregnant vs non-pregnant) and in labor vs not in labor.

Adipose Kiss1 controls aerobic exercise-related adaptive responses in adipose tissue energy homeostasis

Project description:Adipose Energy Homeostasis is the important guarantee to maintain the body's energy balance. Recently, it is reported that, in the adipose cell, Kisspeptins play important role in cell proliferation, differentiation, lipid metabolism and some adipocytokine secretion, so Kisspeptins maybe the novel targets of adipose energy homeostasis regulation. Adipose tissue is the core organs that regulates adipose energy homeostasis. Our early studies observed that there is organizational difference that exercise regulated adipose energy homeostasis, and the response of the Kisspeptins to exercise is closely related to the energy state of the body, so we speculated that Kisspeptins play some important role in the exercise regulated adipose energy homeostasis. Based on the cell experiments to clarify the role of Kisspeptins in regulating adipose energy homeostasis, the role of Kisspeptins in the regulation of adipose energy homeostasis were determined in the adipose tissue of conditioned Kiss1 gene knockout mice of CRISPR/Cas9, then we use the single cell transcriptome sequencing, untargeted proteome and targeted metabolome techniques to further explore and elucidate the possible pathways and mechanisms of Kisspeptins mediated adipose energy homeostasis regulated by exercise.
